Application of immunoblotting method to detect native lipases of rapeseed

The purpose of the study was to determine whether the immunoblotting method can be used to detect native rapeseed lipase in different fractions of rapeseed enzymie extracts of low-erucic Bolko variety and high-erucic Skrzeszowicki variety. Antigen-lipase for production of polyclonal antibodies against the native lipase of rapeseed was obtained after Sephadex G-100 gel separation of supernatant proteins obtained by centrifuging (23 000g x 30 min) the enzymie extract from rapeseed of Bolko variety. In the enzymie extracts (crude enzymie extract, fraction of lipid bodies, microsomal fraction) from the germinated seeds of both rapeseed varieties the affinity to the obtained antibodies was found in the protein fractions with electrophoretic mobilitis corresponding with to those of proteins with the molecular weight of 97 kDa, 84 kDa and 65 kDa-64 kDa-62 kDa.
