The activity of amylases obtained under chemostat conditions was found between 92 to 118 U/mL, but this highly effective biosynthesis was a short-lived process. Continuous amylase synthesis in a packed-bed reactor (carried out for 144 h) by the bacteria immobilized in calcium alginate resulted in slow reduction of cell productivity of amylases ranging from 69.5 to 44.7 U/g of biocatalyst. Amylase biosynthesis in this process was to a large extent dependent on the dilution rate (D). The highest effectiveness of bacteria production (Yp/x= 36.1 U/g beads) was obtained at D=0.8 h-1, while the highest volumetric productivity Qp= 98 kU/Lh was observed at D=6.4 h-1. The bacteria cells immobilized in chitosane gel beads exhibited high yield (70-73 U/g beads) for 96 h of incubation, which was followed by a rapid decrease.
The aim of the study was to determine optimal conditions for amylases biosynthesis by immobilized Bacillus polymyxa В-20 cells. Composition of the medium and immobilization conditions to a large extent affected the efficiency of enzyme production. Optimal production medium consisted of yeast extract (5-10 g/L), maltose, peptone and mineral salts. Stability of alginate biocatalyst beads was increased by 30 g/L gel concentration, increased bead size and chemical gel modification by glutaraldehyde. Increased quantities of synthesized enzymes were proportional to the increment of initial biomass present in beads, and to a lesser degree, depended on quantities of immobilized inoculum. In repeated batch culture, the bacteria immobilized in alginate were able to synthetize 32% more enzymes than those immobilized in chitosane. Operational stability of alginate and chitosane beads was maintained for 17 days.
The effect of two different inocula on growth and production parameters in citric acid fermentation (on glucose) by Yarrowia lipolytica A-101 was studied. For inoculum prepared in full growth medium the total acids yield was 5-12% higher and biomass yield about 10% higher than for inoculum prepared in a nitrogen-deficient medium. The latter inoculum, however, led to about 10-30% higher acid production and glucosce consumption rates.
A new criterion was proposed to evaluate the process of citric acid biosynthesis by the acetate negative mutant strain Yarrowia lipolytica AWG-7 cultivated on glucose syrup by the repeated-batch method. This criterion imparts the same weight to the overall amount of the citric acid production and to the trend in citric acid concentration. To determine the optimum levels of bacto-peptone, ammonium chloride and potassium dihydrogen phosphate which maximize the proposed objective function, a central composite design was developed using 50 ml repeated-batch fermentation. The design involved 20 processes conducted with various combinations of the five values of these three parameters. The experiments produced empirical values of the proposed criterion which were then approximated with the fourth-order polynomial. It was found that the optimal concentrations of bacto-peptone, ammonium chloride, and potassium dihydrogen phosphate equaled 8.5; 159.8 and 65 mgl-1, respectively.
Erytrytol, czterowęglowy alkohol wielowodorotlenowy, jest substancją słodzącą coraz częściej stosowanąjako zamiennik sacharozy. Jego produkcja na skalę przemysłową odbywa się wyłącznie w procesach biotechnologicznych. Celem pracy było porównanie wydajności i dynamiki produkcji erytrytolu z glicerolu odpadowego, pochodzącego z produkcji biodiesla, przez szczep Yarrowia lipolytica Wratislavia K1-UV21 w hodowli okresowej, okresowej zasilanej i powtórzeniowej hodowli okresowej. Hodowle prowadzono w bioreaktorze Biostat B Plus. Całkowite stężenie glicerolu wynosiło 150 i 250 g-dm-3, odpowiednio w hodowli okresowej i okresowej zasilanej. Powtórzeniową hodowlę okresową rozpoczęto jako hodowlę okresową zasilaną, a po wyczerpaniu glicerolu, z bioreaktora usunięto 40% jego zawartości i uzupełniono taką samą objętością świeżego podłoża. Po każdej wymianie stężenie glicerolu wynosiło 100 g-dm"3. Przeprowadzono 4 takie cykle. W hodowli okresowej i okresowej zasilanej drożdże produkowały odpowiednio 84,1 i 136 g-dm-3 erytrytolu. Wydajność produkcji tego związku była zbliżona (0,53 g^g-1) w obu systemach hodowlanych, natomiast szybkość objętościowa, jak i właściwa syntezy erytrytolu była wyższa w hodowli okresowej. Podczas powtórzeniowej hodowli okresowej wysoką wydajność (0,51-0,55 g^g-1), jak i dynamikę produkcji erytrytolu (0,80-1,05 g-dm^h-1) uzyskano podczas trzech pierwszych cykli.
