Studies on the possibility of application of the attenuated strain of DHV (designated as P-50) for duckling immunization against duck virus hepatitis (DVH) have been undertaken. It has been shown that minimal dose of the virus immunizing 100 per cent of ducklings was 10 3.4 EID₅₀. Strain P-50 was reisolated from the liver of vaccinated ducklings and was excreted with faeces up to 9 days postvaccination. During 10 passages in susceptible ducklings, the reversion to virulent form of the attenuated strain have not been noted. The attenuated strain P-50 of DVH induced a quicker immunization effect and higher level of neutralizing antibodies in one-day-old ducklings than vaccinal strain E-52.

A strain of duck hepatitis virus (DHV) isolated from a field case of duck virus hepatitis (DVH) was passaged 50 times in chicken embryos. Influence of serial passages on EID₅₀ and ELD₅₀ titers, virus concentration in the amnioallantoic fluid and in embryo with chorioallantoic membrane, pathogenic properties of the selected virus passages for ducklings have been determined. It has been shown that with the increased number of passages both titers were becoming higher. EID₅₀ was higher than ELD₅₀ up to the 36th passage, and thereafter they remained at the same level. In each examined passage, the concentration of DHV in embryo and chorioallantoic membrane was higher when compared with amnioallantoic fluid. Between the 30th and the 40th virus passages, DHV appeared to be apathogenic for one-day-old ducklings. Histopathological examinations of the liver sections of ducklings inoculated with the attenuated strain did not show lesions characteristic of DVH.
