The mucosa of the uterine cervix is normally devoid of melanocytes; therefore, melanin-containing lesions are very rare in this site. A new case of a common blue nevus in the cervix of a 57-year-old woman is reported. The lesion was an incidental finding in a total hysterectomy specimen performed for atypical endometrial hyperplasia. Gross and histological examination revealed minute dark macula on the mucosa of the posterior aspect of the endocervical canal, composed of loose conglomerates of spindle-shaped and dendritic cells located superficially within the stroma, containing multiple brownish granules, which exhibited positive immunostaining for HMB45 and melan A. Although the blue nevi seem to be lesions of low clinical significance, they require careful differential diagnosis with malignant melanoma, especially in scanty endocervical curettage or cervical biopsy specimens. Sebaceous glands have been extremely rare findings in the female genital system. Excluding the vulva and recent findings, very few cases have been described. The origin of the lesions remains a topic of speculation. However, it appears that prolonged irritation induces a metaplastic response in the ectocervical epithelium. A new case of sebaceous glands in the ectocervix of a 46-year-old woman is reported. The lesions were unexpectedly found in a hysterectomy specimen. The procedure was carried out for multiple leiomyomas of the uterine corpus. Histological examination revealed three mature sebaceous glands located distally to the transformation zone, which opened directly onto the surface epithelium. It could be concluded that sebaceous glands in the ectocervix are rare lesions of unclear origin and low clinical significance. However, the glands could potentially be associated with sebaceous carcinoma of that anatomical site. (Folia Morphol 2009; 68, 4: 287–289)
The effect of endophytic and rhizospheric bacteria was studied on salt stress in a local paddy rice (Oryza sativa L.) variety GJ-17. Plants inoculated with endophytic bacterium Pseudomonas pseudoalcaligenes showed significantly higher concentration of glycine betaine-like quaternary compounds and higher shoot biomass at lower salinity levels. While at higher salinity levels, mixture of both P. pseudoalcaligenes and Bacillus pumilus showed better response against the adverse effects of salinity. However, accumulation of proline showed an opposite trend against plant growth promoting rhizobacteria (PGPR) treatment in salinity stress. Proline concentration increased with salinity but decreased in plants inoculated with either of the PGPRs or mixture of both P. pseudoalcaligenes and B. pumilus. The present study shows that inoculation of paddy rice (Oryza sativa L.) with a mixture of endophytic and rhizospheric bacteria could serve as a useful tool for alleviating salinity stress.
PML is a tumor-suppresor protein involved in the pathogenesis of promyelocytic leukemia. In proliferating mammalian cells PML is a principal component of characteristic nuclear bodies, which contain other proteins but do not contain nucleic acids. There are several PML bodies per nucleus. The molecular function of PML protein is unclear, yet the majority of data points to its involvement in regulation of gene-expression and/or intranuclear protein storage and degradation. In the brain PML has been implicated in the pathogenesis of neurodegenerative disorders, glioma and in the control of embryonic neurogenesis. It is not clear whether the protein is expressed, and has a function, in the normal adult brain. Therefore we have investigated the expression and localization of PML at the cellular and subcellular levels, in the adult mouse brain. By immunofluorescence, typical PML bodies were found in a subset of neurons in the cerebral cortex, hippocampus and amygdala. In the cortex, the protein was present predominantly in layer II/III. Stimulation of neuronal activity by a) pentylenetetrazole seizures or b) immobilization stress, greatly increased the size and signal-intensity of the neuronal PML bodies. Our results suggest that function of PML protein in the brain can be associated with neuronal activity and plasticity.
The activities of vertebrate lysosomes are critical to many essential cellular processes. The yeast vacuole is analogous to the mammalian lysosome and is used as a tool to gain insights into vesicle mediated vacuolar/lysosome transport. The protein SAND, which does not contain a SAND domain (PFAM accession number PF01342), has recently been shown to function at the tethering/docking stage of vacuole fusion as a critical component of the vacuole SNARE complex. In this publication we have identified SAND in diverse eukaryotes, from single celled organisms such as the yeasts to complex multicellular chordates such as mammals. We have demonstrated subfamily divisions in the SAND proteins and show that in vertebrates, a duplication event gave rise to two SAND sequences. This duplication appears to have occurred during early vertebrate evolution and conceivably with the evolution of lysosomes. Using bioinformatics we predict a secondary structure, solvent accessibility profile and protein fold for the SAND proteins and determine conserved sequence motifs, present in all SAND proteins and those that are specific to subsets. A comprehensive evaluation of yeast and human functional studies in conjunction with our in silico analysis has identified potential roles for some of these motifs.
