The tumblehome hull adopts some novelty designs such as low-tumblehome freeboard and wave-piercing bow. The new form design makes the ship have many special hydrodynamic performances. Especially the green water of tumblehome hull is different from that of hulls with flare free board. Green water is a strong nonlinear phenomenon of ship-wave interaction, the variation of free surface of liquid is complicated, and there are still some difficulties to solve green water problems well with numerical simulation method. In this paper firstly the motion responses of the tumblehome hull was calculated based on 3D potential theory, and then the dam-break flow model was used to calculate green water height and pressure distribution. According to the result of numerical simulation, some typical working conditions are chosen for 3D CFD simulation using RANS method. The results of numerical simulation methods are compared with the experimental results measured in towing tank. The influence of different ship form parameters and wave parameters to the green water of tumblehome hull is analyzed, and some regularities of green water on tumblehome hull in regular waves are summarized
Treating plants with abiotic or biotic factors can lead to the establishment of a unique primed state of defense. Primed plants display enhanced defense reactions upon further challenge with environmental stressors. Here, we report that trivalent chromium (Cr(III)) pretreatment can alleviate hexavalent chromium (Cr(VI)) toxicity in 2-week-old wheat plants. The data indicate that Cr(III)- pretreated wheat displayed longer survival times and less heavy metal toxicity symptoms under Cr(VI) exposure than the control. To investigate the possible mechanism from an antioxidant defense perspective, we determined the H₂O₂ and lipid peroxide content (TBARS), the activities of antioxidant enzymes (SOD, CAT, APX and GR) and the antioxidant metabolite content (ascorbate and glutathione content, AsA/DHA and GSH/GSSG ratios) in pretreated wheat roots. The results showed that 0.5 μM Cr(III) pretreatment can alleviate oxidative damage, such as H₂O₂ and TBARS accumulation, in root tissues compared to the control during the first 3 days of Cr(VI) exposure. Furthermore, we determined that this pretreatment can significantly increase the antioxidant enzyme activities and total ascorbate and glutathione contents compared to the control treatment. In addition, redox homeostasis declined slightly in pretreated wheat compared to the control in the presence of Cr(VI). We discuss a possible mechanism for Cr(III)-mediated protection of wheat.
Background: Osteosarcoma (OS) is a common malignant tumor that predominantly occurs in adolescents. Its most common metastasis is to the lungs. As shown in our earlier study, lysosome-associated membrane glycoprotein 3 (LAMP3) is highly upregulated in metastatic OS. However, its role in the regulation of OS cell viability and apoptosis remains unknown. Methods: We knocked down and overexpressed LAMP3 in OS cells and assessed the cell viability and apoptosis. Then, we investigated the expression of apoptosisassociated genes to identify the downstream gene(s) of LAMP3. Results: Knockdown of LAMP3 significantly inhibited OS cell viability and promoted apoptosis. TP53, which is involved in the apoptosis pathway, was found to be highly upregulated after knockdown of LAMP3. Overexpression of LAMP3 significantly increased cell viability and abrogated apoptosis. Importantly, subsequent knockdown of TP53 partially suppressed the increased OS cell apoptosis induced by the inhibition of LAMP3, suggesting that TP53 is a key functional downstream gene of LAMP3. Conclusions: Our findings suggest that LAMP3 promotes OS cell viability and survival by regulating TP53 expression.
Anthocyanins are natural bioactive pigments in plants that play important roles in many physiological functions. They are found in various tissues and can protect plants against different stress conditions. Anthocyanins are synthesized and accumulate in nutritional organs, which is crucial for plants to adapt to and resist adverse environmental conditions, including high exposure to light, ultraviolet light, low temperatures, drought, pests and disease. Some progress has been made in understanding the adaptability of anthocyanin to the external environment. Begonia semperflorens is an excellent model for studying the function and regulation of anthocyanin synthesis. To investigate the biosynthesis and regulation of anthocyanins, RNA sequencing techniques were employed to investigate anthocyanin biosynthesis induced by low temperature in B. semperflorens leaves. A total of 74,779 unigenes with a mean length of 1249 bp were assembled. Functional annotations were implemented using five protein databases. Differentially expressed genes involved in the process of anthocyanin biosynthesis were identified. This study represents the first report of a broad-scale gene expression study on B. semperflorens.
