The goal of the study was to determine the frequency of prion protein (PrP) alleles and genotypes in a flock of the Blackheaded sheep, carrying out the simulation of classical scrapie resistance breeding, as well as to investigate the association between PrP genotypes and lifetime prolificacy in the first, second, and third year of reproductive performance in this sheep. The study covered 95 sheep (79 ewes, 16 rams). Polymorphisms in codons 136, 154, and 171 of the prion protein gene were detected by means of PCR-RFLP method, using the following enzymes: BspHI (PagI), BspDI and BseLI. Based on obtained frequencies of PrP alleles, the classical scrapie resistance mating simulation was carried out. The relationship between PrP genotype and lifetime prolificacy was analysed by the means of χ-square test of independence. Two alleles - ARR and ARQ, and two genotypes - ARR/ARR and ARR/ARQ were identified in the investigated sheep. The most frequent was ARR allele (82.6%) in both ewes (81%) and rams (90.6%). The study on the simulation of classical scrapie resistance breeding revealed a continuous growth of frequency of ARR/ARR individuals, starting from generation FI. The result of χ-square test did not confirm the hypothesis of association between PrP genotype and lifetime prolificacy, which suggests that classical scrapie resistance breeding should not have an influence on reproduction traits of Black-headed sheep, at least on the analysed ones.
The objective of this research was to determine allele and genotype frequencies of the prion protein gene (PRNP) in a flock of sheep and categorise thereof into groups by susceptibility to scrapie. Additionally, a mating simulation was conducted, which aimed at decreasing sheep susceptibility to scrapie. Sheep of Kamieniecka breed from a single flock were investigated. Polymorphism in the PrP gene was identified with the use of the PCR-RFLP method. Four of the five alleles were found - ARR, ARQ, ARU. and VRQ, in nine different combinations - ARR/ARR, ARR/ARQ, ARQ/ARQ, ARR/ARH, ARH/VRQ, ARR/VRQ, ARQ/ARH, ARH/ARH, and ARQ/VRQ. The most frequent allele was the one being the most desired, namely ARR, (63.2%), and the least frequent was VRQ (5.4%). Mating simulation aiming at increasing the frequency of the ARR allele in the examined sheep group demonstrated that in the F₆ generation it is possible to obtain as many as 97.7% of homozygotic animals having the lowest susceptibility to scrapie.
The ovine PRNP gene is responsible for the production of cell prion protein and its polymorphism significantly affects the sheep susceptibility/resistance to scrapie. The aim of the study presented herein was to detect variation (CAG/AAG) occurring at codon 171 of the ovine prion protein gene. The test covered 241 animals from the following sheep breeds: Polish Merino, Blackheaded Mutton Sheep, Polish Mountain Sheep, Ile de France, Berrichon du Cher and Suffolk. The test itself was conducted with the application of the PCR-RFLP method and with the use of the MboI enzyme. A lysine coding triplet was found in the 171 position in case of the Blackheaded Mutton Sheep group. The XXK allele occurred in a heterozygous combination with a frequency of 0.69%, constituting 1.33% of genotypes of the analysed Blackheaded Mutton Sheep group and 0.4% of genotypes of the whole population subject to study.
