In vitro antifungal activity assay of different concentrations of 2-hydroxy-4-methoxybenzaldehyde isolated from Decalepis hamiltonii against six important seed-borne fungal pathogens viz., Alternaria alternata, Drechslera tetramera, Fusarium oxysporum, F. proliferatum, Pyricularia oryzae and Trichoconis padwickii isolated from paddy seeds revealed that, the compound 2-hydroxy-4-methoxybenzaldehyde showed significant antifungal activity. Among the fungi tested, F. proliferatum showed highest inhibitory activity, whereas P. oryzae showed least inhibitory activity. The minimal inhibitory concentration (MIC) varied between 350 μg/ml and 650 μg/ml depending on the fungal species. Comparative evaluation of the active compound with the synthetic fungicide thiram at recommended dosage revealed that, the antifungal activity of the active compound obtained from the plant was almost equivalent. Evaluation for nutritional parameters and dry matter losses (DML) revealed that, total carbohydrates, water soluble proteins, lipids and dry matter losses were significantly confined in 2-hydroxy-4-methoxybenzaldehyde treated paddy seeds compared with control seeds. This plant being an edible one can be exploited in the management of seed-borne pathogenic fungi and in the prevention of biodeterioration of grains and mycotoxin production during storage in an eco-friendly way.
The die-back of neem caused by Phomopsis azadirachtae is a devastating disease in India reducing the life span and seed production of neem. Six isolates of antagonistic bacteria and fungi, Bacillus cereus (MTCC 430), B. subtilis (MTCC 619), Pseudomonas aeruginosa (MTCC 2581), P. oleovorans (MTCC 617), Trichoderma harzianum (MTCC 792) and T. viride (MTCC 800) were evaluated against P. azadirachtae under in vitro conditions. Culture filtrates of all these microorganisms were extracted using ethyl acetate, and the obtained fractions were tested for their antifungal activity against P. azadirachtae at different concentrations. Ethyl acetate extracts of B. subtilis and P. aeruginosa were highly effective and completely inhibited the growth of P. azadirachtae at 25 ppm concentration. Both these isolates may be considered as factors for the biological control of die-back of neem.
A total of 40 seed samples of different cultivars of paddy, collected from southern part of India, were mycologicaly analyzed by agar plating method on Czapek's-Dox-Agar (CzA) medium and Standard Blotter Method (SBM), revealed the occurrence of 33 diverse fungal species belonging to 16 genera. The species of Drechslera oryzae (RP 82.5%) was recorded with the incidence and relative preponderance (RP), followed by Curvularia lunata (RP 67.5%) and Aspergillus niger (RP 65.0%) respectively. The data on the diversity and incidence of fungal species would be a great importance in the region for predicting the extent of pre-and post-infections. In vitro antifungal activity assay of methanol extract of 12 plants belonging to different families were tested against eight pathogenic fungal species viz., Alternaria alternata, Aspergillus flavus, Curvularia lunata, Drechslera oryzae, D. halodes, Fusarium moniliforme, Pyricularia oryzae and Trichoconis padwickii by poisoned food technique. The results revealed that, methanol extract of Acacia nilotica, Caesalpinia coriaria, Decalepis hamiltonii, Emblica officinalis, Lawsonia inermis and Mimosops elengi showed significant antifungal activity at 3500 μg/ml concentration. Comparative evaluation of the extracts with the synthetic fungicides viz., bavistin, blitox, captan, dithane M-45 and thiram at the recommended dosage revealed that, antifungal activity of methanol extract of D. hamiltonii, L. inermis and M. elengi was almost equivalent. These plants can possibly be exploited in the management of seed-borne pathogenic fungi and prevention of biodeterioration of paddy in an eco-friendly way.
