Malinowska, В., Pietraszek, M., Chabielska, E. and Buczko W.: The influence of propranolol on the hypotensive action of ketanserin in normotensive rats. Acta physiol, pol., 1989 40 (4): 356-362. In normotensive rats the effect of different doses of propranolol (1.0, 5.0 and 10.0 mg/kg i.p.) and ketanserin (10.0 mg/kg p.o.) on mean blood pressure and heart rate and on cardiovascular response to noradrenaline (0.1, 0.3, 0.5, 0,7 and 1.0 µg/kg i.v.) was examined. The drugs were given separately or together. Propranolol slightly reduced the hypotensive effect of ketanserin. On the other hand a decrease in heart rate caused by propranolol was not affected by ketanserin. Our results show that propranolol given with ketanserin did not change the effect of the latter on the cardiovascular system.
Nociceptin is the endogenous ligand of the opioid OP4 or ORL1 (opioid receptor-like1) receptor. It decreases blood pressure and heart rate in anesthetized and conscious rats and mice after its intravenous and intracerebroventricular injection in a manner sensitive to OP4 but not to OP1-3 (or , and µ opioid) receptor antagonists. OP4 receptors involved in the cardiovascular effects of nociceptin were identified on sensory afferent fibres, in brain areas including the nucleus tractus solitarii and the rostral ventrolateral medulla, on preganglionic and/or postganglionic sympathetic and parasympathetic nerve fibres innervating blood vessels and heart or directly on these target organs. These receptors do not seem to be tonically activated but may play a role in the pathophysiology of inflammation, arterial hypertension and cardiac or brain circulatory ischemia.

Over the years 1996-1997 mycological examination were performed in 367 persons with cutaneous lesions, returning from the tropics. Fungal infection was diagnosed in 141 patients. Twenty nine (42.65%) of the cases were of single-focal, 32 (47.06%) of bifocal and 7 (10.29%) of multifocal nature. One hundred forty one isolates included 50 (35.6%) moulds 23 (16.3%) yeast - like fungi and 68 (48.1%) dermatophytes. Genus classification of the dermatophytes was as follows: Microsporum - 16 (23.5%), Epidermophyton - 15 (22.1%), Trichophyton - 37 (54.4%). Over the years 1996-1997 the following dermatophyte species were isolated: M audouini, M. ferrugineum, M canis, T. rubrum, T. mentagrophytes var. granulosum, T. tonsurans, T. violaceum, E. floccosum. Microsporum genus was isolated from persons returning from East, West and Central Africa, and from South Europe, Trichophyton genus was isolated from persons returning from Asia and South America.

Due to dramatic development of modern means of transport, exotic countries located on distant continents are today within a few-hour reach by a jet. Every year several million people travelled by air in business or as a tourists. This results in easy transmission of parasitic and infectious factors from continent to continent or from country to country. Plasmodium vivax infection: diagnosis concerned 6 men who had returned from Thailand, India, Senegal and Zaire. The parasitosis was detected as a result of thin smear of periferal blond tests - there were found P. vivax trophozoites an shizontes in erythrocytes. In the patients we found haematologic and termoregulation disturbances as well as hapepatosplenomegaly. Halfan was applied in therapy. Plasmodium falciparum infection: the infection was found in a man and a women after return from Kenya and Sudan. The parasitosis was detected as a result of thin smear of peripheral blond test - there found trophozoites of the parasite in erthorocytes. The course of malaria in the woman was very severe with cerebral malaria, DlC syndrome and blond circulation disturbances, hepatocellular and nephrocellular damage. Entamoeba histolytica, HAV and Candida albicans mixed infections: found in men who had returned from India. Severe E. histolytica infection: diagnosed in a women after return from Greece. There were found massive lesion of large intestine mucosa and high-degree disturbances. Leptospirosis icterohaemorrhagiae infection: found in a man after return from Belarus. Haematologic disturbance and hepatocellular and nephrocellular damage were observed.
The aim of our study was to examine whether non ß1-/ß2-adrenoceptors participate in the relaxation of the human pulmonary artery. For this purpose the vasodilatory effect of the non-conventional partial ß-adrenoceptor agonist cyanopindolol was examined. Cyanopindolol (1 - 300 µM), studied in the presence of the ß1-/ß2-adrenoceptor antagonist propranolol, relaxed the human pulmonary artery preconstricted with serotonin 1 µM in a concentration-dependent manner (maximally by about 80%). This effect was diminished by bupranolol 10 µM (an antagonist of ß1-ß3-adrenoceptors and the low affinity state of the ß1-adrenoceptor) and CGP 20712 10 µM (known to antagonize the low-affinity state of the ß1-adrenoceptor at high concentrations). In further experiments, the effect of ß-adrenoceptor ligands on the serotonin-induced vasoconstriction was examined. The concentration-response curve for serotonin was not affected by cyanopindolol 30 µM, bupranolol 10 µM and CGP 20712 10 µM but shifted to the right by cyanopindolol 100 and 300 µM; the serotonin 5-HT2A receptor antagonist ketanserin 0.3 µM abolished the maximum contraction elicited by serotonin. In conclusion, the present study reveals that the vasodilatory effect of cyanopindolol in the human pulmonary artery consists of two components, i.e. activation of a propranolol-insensitive atypical ß-adrenoceptor and antagonism against 5-HT2A receptors.

Infekcje Toxocara w swietle materialow wlasnych

Toxocarosis clinical forms wers diagnosed according to KRAUS et al. (1995). There were three forms: l) asymptomatic - marked by hypereosynophilia and ELISA positive serologic reaction, 2) minor - with cutaneous, pulmomary and pseudorheumatic lesions, 3) major – with hepatosplenomegaly and multifocal inflammatory lesions of organ or sight. Over the years of 1994-1997 in Infectious, Parasitic and Tropical Diseases clinic of the Voivodship Specialist Hospital of Łódź we found and treated 137 Toxocara canis cases. Among them 63 (46.0%) asymptomatic, 57 (42.6%) minor and 17 (12.4%) major forms were diagnosed. The disease was detected in 80 (58.4%) adults and 57 (41.6%) children. Seventy six patients lived i towns and 61 in country. Minor form patients had the symptoms as follows: skin allergy, large joints lesions, augmentation in lymph nodes. Radiology examinations revealed pulmonary lesions. Using ultrasonography there was found hepatosplenomegaly and changes echogenity in patiems with major form. Biochemical tests showed elevation in hepatic enzymes activity. Lesions of organ of sight were multifocal and usually included uveitis, retinitis, inflammation of anterior chamber and inflammation of vitreous body.
Bupranolol is an antagonist at the cardiostimulatory low-affinity state of ß1-adrenoceptors and we were interested whether this effect is shared by its fluorine (GD-6), methyl (DZ-51) and isopropyl analogue (DZ-13) and by the analogue hydroxylated at the tertiary butyl moiety (DZ-52). (-)-Bupranolol and compounds (-)-GD-6, (+)-GD-6, (-)-DZ-13, (+)-DZ-13, DZ-51 and DZ-52 antagonized the CGP 12177-induced tachycardia in pithed rats with “apparent pA2 values” of 6.1, 6.1, 4.6, 5.5, 4.6, 5.1 and 5.3, respectively. For comparison, their potencies and affinities at the high-affinity state of ß1-adrenoceptors were determined, using the xamoterol-induced tachycardia in pithed rats and [3H]CGP 12177 binding to rat cerebrocortical membranes. The respective “apparent pA2 values” in the functional experiments were 7.9, 8.1, 5.4, 8.4, 5.7, 7.3 and 6.8 and the pKi values in the binding experiments were 8.8, 8.4, 6.9, 8.5, 6.7, 8.4 and 8.2. In conclusion, (-)-bupranolol and its fluorine analogue (-)-GD-6 are equipotent at the low-affinity state of ß1-adrenoceptors. The stereoselectivity of GD-6 and DZ-13 suggests that the low-affinity state is indeed a receptor.
