In this study, a hundred Klebsiella pneumoniae strains isolated from urinary tract infections were evaluated in terms of genotyping, susceptibility to certain antibiotics and detection of extended spectrum of beta lactamase (ESBL) production. The random amplified polymorphic DNA (RAPD-PCR) method was used to identify the genetic differentiation of K pneumoniae isolates. A total of 26 different DNA bands ranging between 334 bp and 28033 bp were detected among the strains. It was found that 100 K. pneumoniae strains revealed 11 different RAPD profiles. Antibiotic susceptibility tests were conducted using a disc diffusion method against 16 antibiotics. Fifty-five different resistance profiles were determined among the strains. ESBL-productions of the strains were determined by the double disc synergy test (DDST) and ESBL E-test methods. ESBL production rates among the strains were found to be 55% by E-test method and 45% by DDST method. While ESBL-producing K. pneumoniae strains showed the greatest resistance to penicillin G (100%), followed by piperacillin (92.7%) and erythromycin (85.4%), the resistance rates of non ESBLproducing strains to those antibiotics were determined as 97.8%, 88.8% and 88.8%, respectively. Both groups of strains showed the highest sensitivity to meropenem. Based on the results obtained from the study, it was concluded that the detection of ESBL-producing strains by the E-test method was more sensitive than by the DDST method. Phenotypic and genotypic identification methods should be used together to detect ESBL presence. The RAPD-PCR method alone will not be adequate in the genotyping of the strains and alternative DNA-based methods should be used.
The purpose of this study was to evaluate the applicability of endoscopic examination as a diagnostic method in urethra and bladder ailments in dogs. The experiment involved 60 dogs of different breeds and gender, weighing from 5 to 65 kg, aged between 7 months and 12 years. The dogs were divided into two groups. Group I consisted of clinically healthy animals on which all the lower urinary tract endoscopic examinations described in the literature as well as biopsies of mucosa were performed. Group II consisted of clinical patients, on which endoscopic diagnostic examination was conducted and, on some of them, biopsies for histopathologic examination were made. The results of the experiment suggest that the lower urinary track endoscopy is a suitable diagnostic technique for recognition and differentiation of mucosa inflammation, establishment of causes of haematuria, diagnostics of neoplasms and detection of the existence of an ectopic ureter. Biopsy and histopathologic examination improved the diagnostic value of endoscopy. The techniques tested in our study, inclusive of more invasive method like PPC and cystoscopy on males preceded by the perineal urethrotomy, enable doctors to perform the lower urinary tract endoscopic examination on dogs, especially up to 5 kg. regardless of their gender.
Although integrons by themselves are not mobile, due to their presence in plasmids and transposons, they can be transferred horizontally. For these reasons integrons are a major mechanism for the spread and maintenance of multidrug resistance (MDR). This study describes the distribution of integron gene cassette classes in a collection of uropathogenic Escherichia coli (UPEC) isolated from children with community acquired urinary tract infection in Jahrom, Iran. E. coli strains isolated from urine samples were tested for susceptibility to 14 different antibiotics using the disk diffusion method and for integron classes by RFLP-PCR. Totally 96 strains of E. coli were isolated from urine samples. High prevalence of resistance to ampicillin (80.2%), co-trimoxazole ((76%) and tetracycline (70.8%) was seen among the UPEC isolates. All isolates were 100% sensitive to imipenem. Sixteen strains (16.6%) had the evidence of integron sequences with the prevalence of 6.25% (n = 6) and 10.41% (n = 10) for intI1 and intI2, respectively. No intI3 was detected in the isolates. The presence of integrons was significantly associated with resistance to certain antibiotics including gentamicin and ampicillin. Considering the MDR patterns and the low prevalence of integrons among the E. coli strains under the study, we suggest that the antibiotic resistance cassettes in these strains presumably are mostly carried on the other transposable elements rather than integrons.
Urinary tract infections are frequently caused by Proteus mirabilis strains. In the previous studies there were defined the complete structures of O-polysaccharide parts of lipopolysaccharides from strains: P. mirabilis O3 (S1959), P. mirabilis O9 and P. mirabilis O18. In the present study it was investigated bactericidal effect of normal human serum (NHS) to P. mirabilis strains. We also focused on the diversity of outer membrane proteins (OMPs) being separated on a gel isolated from tested strains. Serial passage of P. mirabilis O18 in 90% normal bovine serum (NBS) contributed to over-expressing some classes of OMPs.
