The fraction of sterol acetates from the inflorescences of Hieracium pilosella has been isolated in the typical way from petroleum ether extract. By means of the weight method the total amount of sterols was determined (0.2659%). The mixtures of sterol acetates and free sterols were investigated using GC-MS techniques. The occurrence of about 18 sterols has been observed. Cholesterol, cholest-8(14)-en-3b-ol, cholesta-5.7-dien-3b-ol, cholest-7-en-3b-ol, ergosta-5.24-dien-3b-ol, campesterol, stigmasterol, b-sitosterol, fucosterol, 5a-stigmast-7-en-3a-ol were identified. The probable structures of lophenol, isofucosterol, 5a-stigmasta-7.24-dien-3b-ol, lanosta-9(11).24-dien-3b-ol and 24-ethylidene lophenol were stated on the basis of literature data. The last 4 sterols occur in a vestigial quantity, which made its identification impossible. Sitos erol and cholesterol are remarkably dominating sterols in the fraction.
Meadow fescue (Festuca pratensis Huds.) and spring barley (Hordeum vulgare L.) are different in degree of sensitivity to leaf spots, disease caused by fungus Bipolaris sorokiniana. The aim of presented work was the investigation, if metabolites of this fungus change the composition of fatty acids and sterols in callus tissue of both mentioned species. The changes of fatty acid and sterol amount in tissues under influence of fungal elicitor were evaluated dynamically: 1, 3, 10, 24, 48, 72 and 168 hours after elicitation. The content of palmitic, palmitooleic, stearic, oleic, linoleic and linolenic acid was estimated in three lipid classes: monogalactolipids, digalactolipids and phospholipids. In studied tissues the stigmasterol, β-sitosterol and campesterol were found. The obtained results showed that fungal metabolites changed considerably fatty acid and free sterol composition in cells of both studied species. Fescue and barley were unlike in the case of ability to rebuilding of lipid composition under influence of fungal metabolites. In the case of fescue significantly higher content of palmitic, palmitooleic, oleic and linolenic acids in elicited calli was recorded, while in barley ones lower content of oleic, linoleic and linolenic acids was observed. Fescue as less sensitive was characterized by higher ratio of unsaturated/saturated fatty acids than barley. Elicited tissue of barley demonstrated less content of free sterols in comparison with fescue. However, observed changes proceeded dramatically within whole experiment, what make an interpretation of obtained results and a connection them with sensitivity degree to studied pathogen impossibly.
