The paper presents a new way of obtaining viable and very homogeneous cucumber protoplasts. Protoplasts from cells formed in the shoot tip meristem culture were isolated from suspension. Plasmid pBI121 was introduced using impulse electric field. Effectiveness of transformation process was determined by the transient expression of ß-glucuronidase (GUS) gene, controlled by promotor 35S. The activity of ß-glucuronidase enzyme as a product of GUS reporter gene was estimated by fluorimetric method (JEFFERSON 1987). Parameters of electroporation process were optimized. The transient expression of GUS gene was measured 24 h after electroporation. The highest effectiveness of transformation process was achieved using three electric impulses at the initial voltage of 250-350 V at 10-sec. intervals as a result of discharging a 140 µF capacitor and 50-70 µg × cm⁻³ plasmid DNA in the presence of 50 µg × cm⁻³ carrier DNA. The system presented is an effective method of exogenic DNA transfer, which is indicated by a high transient expression of the reporter gene. In comparison to Agrobacterium tumefaciens and A. rhizogenes, this alternative method of gene transfer can be used for obtaining transgenic cucumber plants.
Freshly isolated mesophyll and suspensions-cell protoplasts of S. tuberosum cvs. Desiree and Maris Piper were cultured in different media i.e. modified MS, V-KM and MS-KM. Protoplast plating efficiencies were higher in MS-KM medium. Resulting protoplast-derived calluses were transferred either onto the medium of Bokelmann and Roest (1983) or that of Lam (1977) for shoot regeneration. Calluses derived from mesophyll cell protoplasts differentiated about 2 weeks earlier than calluses derived from suspension-cell protoplasts. Shoot initiation was also about 2 weeks earlier from calluses subcultured onto the former medium as compared to the latter.
Ca2+ acts as a second messenger which controls a wide variety of cellular functions. Ca2+ homeostasis is accomplished by a complex of mechanisms including pumps, ion channels and calcium buffers within storage compartments. Calreticulin is the low-affinity, high-capacity Ca2+-binding protein of most eucaryotic cells. The distribution of calreticulin was studied in isolated protoplasts and during different stages of somatic embryogenesis in carrot (Daucus carota L. cv. St Valery Lunga Rossa) suspension culture. Immunofluorescence staining of protoplasts showed the presence of calreticulin in the cytoplasm. No calreticulin was observed within vacuoles. During somatic embryogenesis calreticulin was distributed mainly in the protoderm of developing embryos in different stages.
The DNA relative content in nuclei from several Solanum species, which were used as partners for somatic hybridization, were determined using a flow cytometry method. The nuclei were isolated mechanically or via protoplasts from leaves of in vitro grown plants. In the case of S. nigrum as well as S. tuberosum cv. Bzura and dihaploid clone H8105, the nuclei were also obtained from suspension cultured cells by lysis of protoplasts. The source and the method of nuclei isolation affected the pattern of nuclear DNA in the genotypes studied. The mesophyll nuclei showed two distinct peaks on the DNA histograms, whereas the DNA peaks produced by cell suspension nuclei were broad and less distinct. The DNA content in the nuclei, calculated from the DNA histograms of the samples and a DNA standard historgam (Trout Red Blood Cells, having DNA content of 5.05 pg per nucleus), were much lower in mesophyll nuclei than in those obtained from the cell suspension for the same genotypes. The results are discussed in respect of the genetic instability of Solanum genotypes. The usefulness of a flow cytometry approach in somatic hybridization research is also discussed.
The possibility of the reversible tonoplast transport of oleanolic acid mono­glycosides was investigated in vacuoles isolated from Calendula officinalis leaf protoplasts. The obtained results point to the reversibility of the transport of monoglucoside I, whereas monoglucuronide F seems to be definitely stored in the vacuolar space.
