Apple scar skin viroid (ASSVd) is a serious pathogen of apple fruits that results in severe loss in apple production. Up to nowadays, many ASSVd management options are applied to resist the disease, but the diserable resulits are not achieved. Therefore, this study was conducted in 2010–2012 at experimental field of Penglai City, Shandong Province, China (E 120°57’22’’, N 370°38’33’’) to investigate whether arbuscular mycorrhizal (AM) Glomus versiforme protects Red Fuji apple trees (Malus × domestica Borkh) against apple scar skin viroid. Red Fuji apple trees were inoculated with Glomus versiforme and then potential protection mechanism was explored and compared to noninoculated plants. The results showed that inoculation with Glomus versiforme significantly increased root length colonization rate and clearly decreased the percentage of disease severity of apple scar skin disease. Compared to non-inoculated plants, Glomus versiforme obviously enhanced total nitrogen and phosphorus concentrations in leaves. Root colonization by Glomus versiforme induced an increase in defense-related enzyme activities in fruits, such as the enhanced activities of catalase, ascorbate peroxidase, chitinase and glucanase. Significant differences in acid content of fruit and fruit yield were observed as apple roots were colonized by Glomus versiforme. It is therefore concluded that Glomus versiforme can be regarded as a biocontrol agent to protect apple trees against the infection with ASSVd.
The study aimed at demonstration of involvement of CD8+ cells in protective mechanisms in canine mammary gland carcinomas. Material for the studies involved metastases to lungs sampled at autopsy from crossbreed bitches aged 11 to 13 years. The tumours were verified histopathologically as adenocarcinomas. The presence of CD8+ cells infiltrating the examined tumour tissues was estimated in cryo-sections by immunofluorescence methods. Parental tumours free of metastases were accompanied by slight infiltration of CD8+ cells. In the case of metastasing tumours, and especially in their pulmonary metastases, the increased number of CD8+ cells was observed. Taken together, metastasing potential of canine mammary adenocarcinomas was positively correlated with the number of CD8+ cells in the tumour stroma.
In the contemporary systems of cattle production, transport stress is the most essential polyetiological factor responsible for inducing unfavourable reactions in the animals. The main reason for this phenomenon is the immunosuppressive effect of steroid hormones on cellular and humoral protective mechanisms. The purpose of the present study was to establish the relationship between the Cortisol concentration as an indicator of the stress reaction occuring directly after the transportation of calves and the specific humoral immune response to the leukotoxin (Lkt) antigen produced by the M. haemolytica strain. The experiment was carried out on 19 clinically healthy calves, weighing about 100 kg and transported by track for about 2 hours. After the delivery of the animals for feeding to the traditional cattle-house, the calves were immunized s.c.: group I with 1 ml Lkt (in conc. - 10 μg/ml) with 1 ml of adjuvant on the 1st and 14th day after the transportation, group II with the same Lkt doses on the 3rd and 16th day after the transportation. The animals of the control group were vaccinated on the 1st and 14th day after the transportation with the twice diluted adjuvant. In examined sera the Cortisol concentrations and the level of Lkt antibodies were measured by ELISA test. The cytotoxin neutralizing (CN) antibody level (cytotoxity assay) was determined with a simple visual assay. The study revealed, significant differences (P ≤ 0.01) in serum Cortisol leves between the control and experimental animals. The analysis of the absorbance of the sera in both groups immunized with Lkt showed substantial differences (P ≤ 0.05) from the 6th through to 22nd day of the experiment compared with the control group. The analysis of the results of CN antibody titers showed no differences between the sera from group I and II. Based on the results obtained in this experiment it can be assumed, that a short transportation stress has no important influence on the level of specific humoral anti-Lkt response.
