The prolactin receptor (PRLR) gene was studied as a candidate gene for the high prolificacy of Jining Grey goats. Polymorphisms in intron 1 and intron 2 of PRLR gene were detected in high prolificacy (Jining Grey) and low prolificacy (Boer, Wendeng dairy, Liaoning Cashmere and Beijing) native goats using PCR-SSCP. For intron 1, five genotypes (AA, AH, AK, HH and HK) were identified in Jining Grey goats, and two (AA and AK) in the other four breeds. The Jining Grey does of genotype HH, HK, AH and AK delivered by 0.65, 0.62, 0.59 and 0.57 more kids (P<0.01) than those of genotype AA, respectively. For intron 2, three genotypes (CC, CD and DD) were detected in Boer goats, and two (CC and CD) in the other four breeds. The Jining Grey does of genotype CD delivered by 0.55 (P<0.01) more kids than those of genotype CC.
In marker-assisted selection (MAS) of dairy cattle certain genes are proposed as potential candidates associated with dairy performance traits. Among different candidates, prolactin receptor gene (PRLR) seems to be promising because of its crucial role in transmitting signal from lactogenic hormones to milk protein gene promoters. In this study nine PCR fragments representing most important functional domains of PRLR were screened for polymorphism. Using SSCP method one SNP (A→C) was found in intron 9. The SNP was deposited in GenBank AY484400 and AY339393 at position of 205 nt, for Jersey and Polish Black-and-White cattle, respectively. Allele frequency was estimated in 186 Polish Black-and-White (0.981 and 0.019 for A and C, respectively) and 138 Jersey (0.812 and 0.188 for A and C, respectively) cows. Preliminary analysis showed no significant associations between PRLR genotypes and milk performance traits. However, Jersey cows of CC genotype produced more milk with higher protein content than those of AA and AC genotypes. Because of the low number of cows of CC genotype, it is necessary to investigate more numerous population of cattle in which all genotypes will be efficiently represented.
The prolactin receptor gene (PRLR), located on chromosome 16 in pigs, is a candidate gene for reproductive traits. The experiment was aimed to detect the DNA mutations in this gene and to find probable relations between the genotype and some reproductive traits in boars. The polymorphism in the PRLR gene was identified by PCR-RFLP method using specific primers and the restriction enzyme AluI. In total 229 boars of various breeds were genotyped. The frequency of allele A was estimated at 0.62 and allele В at 0.38. Genotype AA was found at a frequency of 0.45, AB at 0.35 and BB at 0.20. We found associations between PRLR genotype and ejaculate volume, sperm concentration, percentage of live sperm, and number of live sperm in the ejaculate (P < 0.01).
