Liposomal formulations of cyclosporin A have been investigated in a series of studies. One of the most used arguments for the development of this pharmaceutical preparation was the high lipophilicity of cyclosporin A, which ensures an encapsulation of cyclosporin A in the liposomal bilayer. From this study on the interaction of cyclosporin with lipids spread as a monolayer on the air/water interface we demonstrate, that one molecule of cyclosporin A occupies an area of 260 Å2 in the lipid monolayer. These data correlate well with molecular modelling data, calculating a cross-sectional area of 230 Å2 for a cyclosporin A molecule. Calculation of a partition coefficient using these data at different surface pressures results in a value of about 4000 at a surface pressure of about 31 mN/m. This partition coefficient is also found in bilayer membranes supporting the notion, that phospholipid bilayer systems have a surface pressure of about 32 mN/m. If the lipid:cyclosporin A ratio exceeds a value of 20, the lipid monolayer becomes nonlinear. Cyclosporin A starts to detoriate liposomal membranes at similar lipid:cyclosporin ratios. It could also be concluded from our measurements at the air/water interface, that cyclosporin A is able to exchange binding places between membranes rapidly with a time constant of about 2.5 min. Lipid monolayer experiments are not only an easy and fast method to obtain information about molecule interactions with lipid membranes, but is also a suitable method to develop liposomal formulations of drugs.
Prepenetration and penetration phenomena after inoculation of roots of Pinus sylvestris seedlings grown in vitro with the P-, S- and F-group isolates of Heterobasidion annosum were observed using scanning electron microscop. There were no differences in the behaviour of hyphae and in the appearence of mycelia formed by the three IS-group isolates. Four types of the root penetration by hyphae were observed. In the first, the entrace of hypha into root was achieved throught tiny pore formed in walls of the cortical cells. In the second, swellings resembling appressoria were formed by the hyphal tips at the points of contact with the root surface. In the third, the hyphae penetrated cells through holes in eroded areas of roots, and in the fourth by the natural crevices at points of the cortical cell junctions. It is concluded that the different frequencies of the four types of penetration observed after inoculation with isolates of the three IS group should be carefully explained in term of their different pathogenic and saprobic capabilities on pine.
The effectiveness of penetration of erythrocyte membrane by sodium salt of 2,4-dichlorophenoxyacetic acid was analyzed. The experiment was executed in a dependence on different doses of the herbicide and at different times of incubation of red blood cells with 2,4-D-Na. It is known that the main mechanism of detoxification of the cell from xenobiotics including 2,4-D is to bind them with proteins contained in blood plasma. In the case of exposure of blood to high doses of 2,4-D-Na, the unbound part of xenobiotics may penetrate into erythrocytes and change the activity of numerous parameters of the cells. The results obtained by the use of high pressure liquid chromatography (HPLC) revealed that 2,4-D-Na is adsorbed by erythrocytes of 5% haematocrite in the amount of ~1% of the initial concentration. Moreover, it was observed that 2,4-D-Na is capable of accumulating in erythrocyte’s membrane and haemolysate in the amounts of 0.15% and 1.23% of the initial concentration, respectively. It was also stated that penetration of 2,4-dichlorophenoxyacetic acid into erythrocytes is not associated with incubation time (the similar concentrations of 2,4-D-Na were detected after different incubation times of 0.5 to 3 hours); however, it was related with concentrations of the herbicide. We suggest that 2,4-D-Na was transported with concentration gradient in human erythrocytes.
The composition of mycoflora in storage rooms, and other rooms in a poultry-processing plant, as well as on the surfaces of egg shells was observed. The concentrations of both aflatoxin B₁ and ochratoxin A were determined in the shell eggs at room temperature and humidity, at a higher temperature and humidity, and in the eggs previously contaminated by Aspergillus flavus. We found that there was a reciprocal correlation between the presence of microscopic filamentous fungi in the air and on the working tables (Cladosporium spp. 45.5%, Penicillium spp. 36.4%, Mucor spp. 9.0%). The penetration of mycotoxins through the egg shell was relatively low and the residue limit of aflatoxin B₁ allowed (5 μg.kg⁻¹) was not exceeded in any sample of egg tested. However, the residue limit of ochratoxin A (20 μg.kg⁻¹) was exceeded in one case.
The characteristics of air jet together with travel velocity of the sprayer are crucial factors influencing the penetration of air in the tree canopy. The objective of the study was to determine the volume of air penetrating into the canopy of apple trees, produced by three orchard sprayers with different fan types and operated at different travel velocities. Due to different fans, the following discharge systems were available: radial air flow, cross-flow and directed air flow. The sprayers were driven at 4, 6 and 8 km/h along the trees inside which the air velocity was measured in 9 points, placed in 3 vertical layers. The obtained data was processed to get the air volume penetrating the tree canopy and being delivered to the points of measurements. The influence of travel velocity on air penetration in the tree depended on the character of the air jet produced by the sprayer. For high volume/low speed air jets (radial air flow and cross flow) the decrease of travel velocity caused consid.
Pathogenicity and ultrastructural investigation of the inoculation of peppermint stems and rhizomes with Phoma strasseri conidia was undertaken using scanning and transmission electron microscopy to examine the host-parasite relationship. Pathogenicity experiments demonstrated that all tested P. strasseri isolates had infected the stems and rhizomes of peppermint. Of all inoculation methods, direct placement of colonized agar plugs on damaged epidermis and soaking stems and rhizomes in conidial suspension were the most e#ective. !e behavior of the conidia deposited on the stems and rhizomes was investigated at different time intervals after inoculation: 6, 16, 24, 36 and 48 h. Conidia produced an appressorium directly at the end of a short germ tube. Appressoria were formed over the cuticle, but never over stomata. Direct penetration to host tissue through the cuticle was observed. The spore and hyphae were covered with a mucilaginous sheath.
