Pentatrichomonas hominis, a parasitic protozoan often detected in human diarrheic stools, is the cause of severe morbidity in newborns and children, particularly in tropical zones. The flagellate is resistant to many disinfectants and anti-protozoan drugs. Therefore in this study we have synthesized three novel 4,5,6,7-tetrabromobenzimidazole (TBBI) derivatives carrying a polyfluoroalkyl substituent in position 2 of the benzimidazole scaffold, namely 2-trifluoromethyl-TBBI (CF3-TBBI), 2-nonafluorobutyl-TBBI (C4F9-TBBI), and 2-nonadecafluorononyl-TBBI (C9F19-TBBI), that next we tested for their in vitro activity against P. hominis.Widely applied anti-protozoal drug, metronidazole as a reference was used. All the investigated agents were added to 24 h P. hominis cultures; each of them was administered at three different concentrations. Number of the moving trichomonads was determined and compared with the control cultures. Different anti-trichomonal activity occurred depending on a kind of compound and its concentration. C4F9-TBBI was the most effective TBBI derivative tested: the agent, at the highest concentration 24.2 μg/ml, after 72 h reduced the number of viable trichomonads to 44.3%; C9F19-TBBI, at the concentration 24 μg/ml reduced the number of the flagellates to 58.5%. Paradoxically, metronidazole after the same time given at the highest concentration increased trophozoite counts by 464.6% in comparison with the control cultures (100%).
This study was conducted to investigate the prevalence of parasitic protozoan agents in lambs with diarrhoea in the Van province (Eastern Turkey). Faecal samples from 132 diarrhoeic lambs aged from one day to two months old were collected at 12 different herds from January 2005 to May 2005. The faecal samples were examined by using, native-Lugol, Fulleborn's salty water technique, and a Modified Ziehl-Neelsen staining technique for Cryptosporidium. The parasitological examination revealed that 98 (74.24%) of the lambs were infected with one or more of parasitic protozoan agents. Eimeria, Giardia, and Cryptosporidium were detected in 80 (60.60%), 64 (48.48%), and 18 (13.63%) of the lambs, respectively. According to statistical analysis, Eimeria sp. oocysts were significantly higher (P<0.01) in lambs aged between 31 and 60 d (76.81%, 53 of 69) than in those of 16-30 d of age (50%, 21 of 42) and 1-15 d of age (28.57%, 6 of 21). There were significant differences in Giardia prevalance when lambs of 1-15 d of age were compared with those 16-30 d of age (P<0.05). Statistical analysis revealed that Cryptosporidium sp. oocysts were significantly higher (P<0.01) in lambs aged between 1 and 15 d (66.66%, 14 of 21) than in those of 16-30 d of age (4.76%, 2 of 42) and 31-60 d of age (2.89%, 2 of 69). Moreover, Giardia sp. and Eimeria sp. showed significant co-occurrence (34.84%). The results obtained confirmed the fact that protozoan infections are widespread in diarrhoeic lambs in this geographical region.
During the current century, 88 species of parasites have been recorded in Bison bonasus. These are 22 species of protozoa (Trypanosoma wrublewskii, T. theileri, Giardia sp., Sarcocystis cruzi, S. hirsuta, S. hominis, S. fusiformis, Neospora caninum, Toxoplasma gondii, Cryptosporidium sp., Eimeria cylindrica, E. subspherica, E. bovis, E. zuernii, E. canadensis, E. ellipsoidalis, E. alabamensis, E. bukidnonensis, E. auburnensis, E. pellita, E. brasiliensis, Babesia divergens), 4 trematodes species (Dicrocoelium dendriticum, Fasciola hepatica, Parafasciolopsis fasciolaemorpha, Paramphistomum cervi), 4 cestodes species (Taenia hydatigena larvae, Moniezia benedeni, M. expansa, Moniezia sp.), 43 nematodes species (Bunostomum trigonocephalum, B. phlebotomum, Chabertia ovina, Oesophagostomum radiatum, O. venulosum, Dictyocaulus filaria, D.viviparus, Nematodirella alcidis, Nematodirus europaeus, N. helvetianus, N. roscidus, N. filicollis, N. spathiger, Cooperia oncophora, C. pectinata, C. punctata, C. surnabada, Haemonchus contortus, Mazamastrongylus dagestanicus, Ostertagia lyrata, O. ostertagi, O. antipini, O. leptospicularis, O. kolchida, O. circumcincta, O. trifurcata, Spiculopteragia boehmi, S. mathevossiani, S. asymmetrica, Trichostrongylus axei, T. askivali, T. capricola, T. vitrinus, Ashworthius sidemi, Onchocerca lienalis, O. gutturosa, Setaria labiatopapillosa, Gongylonema pulchrum, Thelazia gulosa, T. skrjabini, T. rhodesi, Aonchotheca bilobata, Trichuris ovis), 7 mites (Demodex bisonianus, D. bovis, Demodex sp., Chorioptes bovis, Psoroptes equi, P. ovis, Sarcoptes scabiei), 4 Ixodidae ticks (Ixodes ricinus, I. persulcatus, I. hexagonus, Dermacentor reticulatus), 1 Mallophaga species (Bisonicola sedecimdecembrii), 1 Anoplura (Haematopinus eurysternus), and 2 Hippoboscidae flies (Lipoptena cervi, Melophagus ovinus). There are few monoxenous parasites, many typical for cattle and many newly acquired from Cervidae.
Chlorazol Black E (CBE) stain has been used for the detection and identification of intestinal parasitic protozoa. In recent years, genotyping of protozoa has been performed to examine pathogenicity and for epidemiologic analysis. In this study, protozoan DNA was amplified from preserved human fecal specimens stained with CBE that were positive for Giardia intestinalis (syn. G. lamblia and G. duodenalis), Chilomastix mesnili, Pentatrichomonas hominis, and Entamoeba histolytica. DNA was amplified from 11 of the 12 (91.6%) samples examined. DNA from CBE-stained smears of G. intestinalis, E. histolytica, and P. hominis was amplified, whereas any amplification product could not be obtained from one of three smears of C. mesnili. Storage term and protozoan number had no association with results of PCR amplification. In genotyping of G. intestinalis, four out of six (66.7%) samples were of genotype AI, while the remaining two (33.3%) samples were of genotype B. The amplified DNA sequences showed high similarity (>99%) with that of G. intestinalis in the GenBank database. These results suggest that DNA remains stable in CBE-stained smears for long term. The present study demonstrates that nuclear extracts from specimens stained with CBE can be amplified by PCR and suggests that specimens stored for extended periods could be applied to genetic and prospective epidemiologic analyses.
Results of studies and literature search on parasitic and commensalic eugregarines in ground beetles (Coleoptera: Carabidae) on the territory of Poland are presented. Records of parasitisation of adult ground beetles (Carabidae) of 84 species were examined and analyzed for the presence of parasitic protozoans belonging to eugregarines (Apicomplexa: Eugregarinorida). Parasitic eugregarines were recorded in 46 species of Carabidae belonging to fifteen genera.
