Rapid, reliable diagnosis is a necessary condition for the successful treatment of infections. Such diagnostic assays are continually being developed. The paper presents a method for selecting the molecular target for PCR-based diagnostics based on the comparison of RAPD patterns. A sequence encoding Candida glabrata CBS138 hypothetical protein was selected. The limit of detection for PCR and real-time PCR reactions with DNA extracted from blood samples spiked with Candida glabrata was estimated at 1 CFU/ml. The application of the assays developed in this study would thus seem to be promising as a complementary method in the diagnostics of C. glabrata infections.
The objective of this research was to determine the suitability of boiling water test in the prediction of seed viability in leek seed and to compare this test with standard germination and vigour test. Basic principle of boiling water test is the evaluation of radicle protrusion from the seeds immersed to boiling water for certain time. The viability of aged and unaged leek seeds in boiling water test was correlated with standard germination and vigour test. In this research, the mean germination of unaged leek seeds varied from 29,63% to 70,18% depending on the cultivars. While the range of boiling water test in unaged leek seeds was between 35,57% to 51,06%, the range for emergence was between 27,63% to 72,05%. The viability of aged and unaged leek seeds in boiling water test was correlated with standard germination test. According to the results of this research, it might be assessed that boiling water test is a rapid and simple method to measure seed viability by evaluating radicle protrusion of leek seeds in boiling water. In conclusion, boiling water test as a new viability test for leek seed could be used as a rapid method for determining the viability in leek seeds.
Recently developed the agility and skill tests (AS) were reevaluated to assess the children’s agility together with the soccer shoots to ball for goal. Children (male) soccer players (N = 68) (age = 11.6 ±0.5 yrs; height = 147 ±6.6 cm; weight = 35.5 ±5.6 kg) participated in this study. Test-retest and comparisons, including 20 m Sprint, Long Jump, T-Drill Test, AS (with ball), AS (goal success) and Zigzag Tests were used to assess the children’s sprinting, jump power, and agility with shooting ball. All handled test results showed that there were significant differences between the test-retest sessions (p < 0.01) except for the 20 m Sprint. There were a near perfect correlations between test and retest values (r = 0.90–0.99). A higher level of correlation between the AS tests (goal success, with ball) (r = 0.99) was found. There were very high levels of correlations between the AS (goal success and with ball) tests and Zigzag tests (r = 0.71, r = 0.70 respectively). These results suggest that the AS tests are reliable and valid agility and skill tests for children players. Because the AS tests have unique values and are composed of soccer specific agility and active shooting skills, they are very important in identifying the talent and ability of children.
