Excessive building dampness causes a number of adverse phenomena. That is why excessive dampness should be prevented, and if it occurs, drying is necessary. The removal of moisture from the building is an expensive, long-term and difficult process. Permanent drying requires not only drying the walls, but also the elimination of the causes. Due to this, complex activities are required. The article discusses the problem, with special emphasis on the drying methods.
The article presents basic principles of flow cytometry (FCM), which is a highly advanced technology based on measuring physical and chemical parameters such as: light scatter at different angles and fluorescence. The essential principle of flow cytometry is identifying cell surfaces and intracellular antigens using specific monoclonal antibodies conjugated with fluorochromes. Cells labeled with fluorochromes pass through the flow chamber and, when exposed to the laser beam, they emit and scatter light, causing alterations in the intensity of the light. Light scatter and fluorescence is measured at the same moment. Light signals are converted into digital signals by a photomultiplier system and analyzed by computer. Flow cytometer and computer software provide multiparameter qualitative and quantitative analysis of single cells in a quick, precise and repeatable manner.
Opracowano metody oznaczania kwasu 2,6-diaminopimelinowego (DAPA) przy użyciu zestawu HPLC (Waters) i kolumny z odwróconą fazą (Nova-Pak C18) poprzez elucję gradientową oraz detekcję UV przy 340 nm lub fluorescencyjną (wzbudzenie 229 nm, detekcja 470 nm). Po hydrolizie próbek DAPA przeprowadzano w pochodne przy użyciu o-ftaldialdehydu (OPA) w obecności 2-merkaptoetanolu (metoda 1) lub OPA w obecności etanotiolu (metoda 2) z ewentualnym usunięciem nadmiaru innych aminokwasów (metoda 3). Metodą 1 można rozdzielić poszczególe izomery DAPA. Na podstawie otrzymanych wyników można wnioskować, że metody 1 i 2 mogą być wystarczająco czułe i selektywne do oznaczania DAPA w bakteriach przy użyciu detekcji UV. Natomiast zastosowanie metody 3 z detekcją fluorescencyjną pozwala uzyskać zadowalające wyniki nawet przy bardzo niskich stężeniach DAPA.
