The increasing attention paid by both the legislature and consumers concerning animal welfare has raised criticism regarding the tie stall housing system of dairy cattle, since it restricts voluntary movement and limits the social behaviour of cows. The aim of this study was to compare the welfare of dairy cows kept in a tie-stall (TS) and an open-stall (OS) system by assessing metabolic, immunological and stress-related parameters. The study involved 155 cows in 18 farms located in Tuscany. Blood samples were collected in the morning in order to measure: aspartate-aminotransferase (AST), alanine-aminotransferase (ALT), betahydroxybutyrate (BHBA), blood urea nitrogen (BUN), nonesterified fatty acids (NEFA), total proteins (TP), creatinine (Creat), calcium (Ca), potassium (K), phosphorus (P), lysozyme (SL), haptoglobin (Hp), and oxygen free radicals (OFR). At the same time, hair samples were collected to measure cortisol levels and the body condition score was recorded. The results showed that the housing system affected AST, ALT, BHBA, BUN, SL and OFR levels. Most parameters showed values within the range of reference. However, the OFR level was higher in the OS system, probably due to the higher productivity than in TS. Cortisol did not raise particular concern related to chronic stress, since the values were lower than the data reported in literature. The study revealed that the evaluation of welfare based on physiological parameters showed no severe signs of impairment in cows reared in the TS system.
Major depression is frequently associated with the hyperactivity of the hypothalamic-pituitary-adrenocortical axis, and glucocorticoid synthesis inhibitors have been shown to exert antidepressant action. The aim of the present study was to examine the effect of joint administration of metyrapone (50 mg/kg) and imipramine (5 and/or 10 mg/kg) on immobility time, plasma corticosterone concentration, the weight of spleens and thymuses and the proliferative activity of splenocytes in rats subjected to the forced swimming test - an animal model of depression. Metyrapone alone (50 mg/kg) reduced the immobility time of rats in the forced swimming test and decreased plasma corticosterone level, but did not change immunological parameters. Joint administration of metyrapone and imipramine (5 and 10 mg/kg) produced a more pronounced antidepressant-like effect than either of the drugs given alone. The forced swimming procedure significantly increased the proliferative activity of splenocytes, that parameter being reduced only by co-administration of metyrapone and imipramine. Joint administration of metyrapone and imipramine inhibited to a similar extend the corticosterone level as did treatment with metyrapone alone (about twofold); however, the plasma corticosterone level in animals treated with metyrapone and the higher dose of imipramine did not differ from the concentration of this steroid in control, not-stressed rats. The obtained results indicate that metyrapone potentiates the antidepressant-like activity of imipramine and exerts a beneficial effect on the stress-induced increase in plasma corticosterone concentration and the proliferative activity of splenocytes. These finding suggest that a combination of metyrapone and an antidepressant drug may be useful for the treatment drug-resistant depression and/or depression associated with a high cortisol level.
Ninety-seven bison (39 males and 58 females) were culled out during three successive winter seasons. No infection with BHV-1 and BHV-5 or other viruses was detected by PCR and SN methods. Advanced pathological lesions observed in males exhibiting symptoms of posthitis/balanoposthitis are probably due to secondary infection by microorganisms present in the prepuce (Corynebacterium spp., Bacillus spp., Fusobacterium necrophorum, Staphylococcus spp. (coagulase negative) and Staphylococcus hyicus). Histopathological examinations of sections from prepuce and lips of pudendum showed the presence of Onchocercinae nematode (Onchocerca sp.). A comparison of biochemical and morphological blood parameters in the bison with or without lesions in the digestive (presence of parasites), respiratory, urinary and reproductive tracts revealed that the most variable parameter during reproductive organ disorder is the leukocyte count, while disorders in the urinary and digestive tracts coincided with the age of animals. In cases of digestive disorder most changes were observed in the number of neutrophils and concentration of urea. The number of red blood cells, monocytes, the level of alkaline phosphatase (AP) and hematocrit negatively correlated with age. Negative correlation was also observed between the number of neutrophils, level of creatinine and lymphocyte proliferation index. However, positive correlation was seen between erythrocyte sedimentation rate (Spearman's correlation coefficient), level of alanine aminotransferase (ALT) and protein, and age of the bison. Positive correlation was also observed between the lymphocyte count and lymphocyte proliferation index. The attempt to determine the reference values for blood biochemical and morphological parameters showed similarities to those of cattle.
