The study was aimed at investigating whether keeping colonies on small-cell combs (cell width of 4.93 mm), in combination with natural selection resulting from the lack of V. destructor treatment, leads to an intensification of hygienic behaviour. We also sought to determine whether the very transfer of colonies treated against V. destructor (without natural selection) onto small-cell combs causes an intensification of hygienic behaviour. Hygienic behaviour was monitored in both varroatosis-treated and untreated colonies kept on small-cell combs, as well as in two control groups consisting respectively of varroatosis-treated and untreated colonies kept on combs with standard size cells (cell width of 5.56 mm). Regardless of whether they had been treated or not, the colonies kept on the small-cell combs removed dead pupae (needle test) faster than the colonies kept on the standard-cell combs, both treated and untreated. In this way, we demonstrated that hygienic behaviour can be intensified by transferring colonies onto small-cell combs, even without natural survival selection resulting from the lack of treatment against V. destructor. The practical use of this method, however, requires further studies of various bee populations, as they may differ significantly in the intensity of hygienic behaviour and, possibly, in their aptitude for being kept on small-cell combs.
The aim of research was the assessent of the hygienic behaviour of worker bees. The hygienic behaviour was assessed by means of two test types: the traditional needle test and a test proposed by the authors that consisted in measuring the removal rate of cardboard pieces. The experiments were conducted over three seasons and comprised a total number of 60 colonies. It was proved that the cardboard removal test may replace the needle test in the selection process of bees resistant to brood diseases, but primarily in those bees that have a good expression of hygienic behaviour. The best test results are to be expected in tests which last for 24 hours.
Background. Alcohols are the most commonly used active substances in preparations for quick hand disinfection. They should be bactericidal in very short contact time. PN-EN 13727 + A2: 2015-12 standard, for testing hygienic and surgical handrub disinfection preparations, provides mandatory test conditions of disinfectants in contact times with the range of 30 s to 60 s (hygienic handrub disinfection) and 60 s to 5 min (surgical handrub disinfection). A short contact times for hand hygiene products require a short time of neutralization process. For contact times less than or equal to 10 minutes, the estimated neutralization time is 10 s ± 1 s. Neutralization is a process that abolishes the action of disinfectants. Correct application of this process allows for proper use of disinfectants in practice and its biocidal effect. Objectives. Verification of the effectiveness of 10-second neutralization time of alcohol based preparations for hygienic handrub disinfection. Materials and Method. Neutralization of two products with different ethanol content (89% and 70%) for hygienic handrub disinfection according to PN-EN 13727 + A2: 2015-12 was investigated. The effectiveness of the neutralizer was assessed by determining toxicity of neutralizer, activity of residual effects of the tested products and their derivatives produced during neutralization (10 s) for test organisms (Staphylococcus aureus ATCC 6538; Pseudomonas aeruginosa ATCC 15442; Enterococcus hirae ATCC 10541; Escherichia coli K12 NCTC 10538). Results. The 10-second neutralization time was sufficient to eliminate the residual activity of products for hygienic handrub disinfection with differentiated ethanol concentration. The neutralizer used did not show toxicity to bacteria and did not produce toxic products with tested preparations after neutralization. Conclusions. The use of 10-second neutralization time allows in a precise way designate the contact times for hygienic handrub disinfection products.
The aim of this study was to determine which method of hygienic behaviour assessment is more reliable: the evaluation of the pierced brood removal rate or the evaluation of the freeze-killed brood removal rate. Additionally, we aimed to determine whether freeze-killed brood should be placed in colonies when defrosted or still frozen. Defrosted freeze-killed brood was removed faster within a 24 h period. The removal rates for pierced brood and frozen freeze-killed brood were similar in hygienic colonies. In non-hygienic colonies, pierced brood was removed at a significantly slower rate than frozen or defrosted freeze-killed brood. The mechanisms of removing frozen and defrosted freeze-killed brood were similar to each other and different from those observed in the case of pierced brood. The defrosting of brood prior to its introduction into colonies seems inadvisable, as it accelerates brood removal. Our results confirm the hypotheses of those researchers who believe that the frozen freeze-killed brood removal test is not always appropriate. A good solution is, therefore, to perform the frozen freeze-killed brood and pin-killed brood removal tests simultaneously. The time from the beginning of the tests to the moment 50% and 75% of dead-brood cells have been cleaned up should be assumed as the appropriate duration of the hygienic behavior evaluation tests.
