Viral cross-protection in plants is a phenomenon, where a mild virus isolate can protect plants against damage caused by a severe challenge isolate of the same virus. It has been used on a large scale in cases where no resistant plants are available. We examined differences in cross-protection between pathotypes of Pepino mosaic virus representing Chilean 2 genotype. The potential of a mild PepMV-P22 isolate to protect tomato against more aggressive challenge isolates causing yellowing and necrotic symptoms was established. The challenge isolates were PepMV-P5-IY (yellowing), PepMV-P19 (necrotic) and PepMV-P22 K67E (artificial necrotic mutant of PepMV-P22 which differ from PepMV-P22 only by a point mutation). Efficient cross-protection was obtained using mild PepMV-P22 against PepMV-P5-IY. After a challenge inoculation with PepMV-P19 or PepMV-P22 K67E symptoms severity were significantly reduced in comparison to non-protected plants; however, necrotic symptoms appeared two months after coinfection. The real-time PCR analysis revealed that the level of accumulation of the necrotic isolate in tomato plants was even 5–7 times higher than that of PepMV-P22.
Background: The ACTN3 R577X polymorphism has been associated with an elite athlete status. Several studies have determined that the R allele is connected with power-oriented athletic performance, whereas the nonfunctional XX genotype may give some beneficial effect for endurance performance. The main aim of the study was to determine the possible interaction between the ACTN3 R577X polymorphism and an endurance athlete status in a group of Polish rowers in comparison with sedentary individuals. Material/Methods: 121 male Polish rowers, members of academic sports clubs, and 115 unrelated volunteers were recruited for the study. Genotyping for the R577X variant was performed by PCR–RFLP. Results: The genotype distribution amongst the rowers (52.06% RR, 38.85% RX, 9.09% XX) was significantly different from that amongst sedentary individuals (RR-33.5%; RX- 49.60%; XX-17,35%; P = 0.024). A significant excess of the R allele was noted in the rowers (71.48%, P = 0.008) when comparing with the controls (60.0%). Conclusions: The obtained results show that the ACTN3 X allele and XX genotype are underrepresented in Polish rowers and they are not advantageous for the endurance-type athletes in the studied population. On the contrary, the R allele seems to be useful for a top-level rower. However, additional studies are needed to clarify this problem.
The aim of this study was to establish the effect of the genotype on performance of chickens. The experimental material included 1,320 slow-growing chickens with two genotypes Hubbard JA 957 - available on the Polish market hybrid with a declared slow growth and non-commercial hybrid CxGP, for creation of which a light native breed Greenleg Partridge hens were used. Chickens were maintained up to 63 days of age. During the experiment body weight, feed intake and health of the birds were under control. Based on the observations the growth rate and the feed conversion ratio (FCR) were determined. It was found, that chickens CxGP were characterized by lower body weight, slower rate of growth and higher feed conversion ratio compared with the chickens Hubbard JA 957, but chickens CxGP had a higher health.
Natural killer (NK) cells are the most abundant lymphocyte population in the decidua. These cells express killer immunoglobulin-like receptors (KIRs), which upon recognition of HLA class I molecules on tropho- blasts may either stimulate NK cells (activating KIRs) or inhibit them (inhibitory KIRs) to produce soluble factors necessary for the maintenance of pregnancy. KIR genes exhibit extensive haplotype polymorphism; individuals differ in both the number and kind (activating vs. inhibitory) of KIR genes. This polymorphism affects NK cell reactivity and susceptibility to diseases, including gynecological disorders. Therefore we ATR-genotyped 149 spontaneously aborting women and 117 control multiparae (at least 2 healthy-born children). Several genotypes (i.e. combinations of various KIR genes) were differently distributed among the patients and control subjects. Differences were observed in the numbers and the ratios of activating to inhibitory KIRs between patients and healthy women: (i) genotypes containing 6 activating KIR genes were less frequent and those containing 6 inhibitory KIR genes were more frequent in patients than in control subjects, and (ii) an excess of inhibitory KIRs (activating-to-inhibitory KIR gene ratios of 0.33 to 0.83) was associated with miscarriage, whereas ratios close to equilibrium (0.86-1.25) seemed to be protective. In addition, the results suggest for the first time that sporadic and recurrent spontaneous abortions as well as miscarriage in the presence or absence of autoantibodies may have different KIR genotypie backgrounds.
A simplified amplified-fragment length polymorphism (AFLP) method was used to genotype Pichia pastoris strains obtained by transformation of P. pastoris strain GS115 with a single integration vector. A total of 14 transformants and 3 control strains were analyzed, which generated 16 different band patterns. A clonal variation was obtained after the transformation process due to genetic differences generated during the transformation event of the host strain. Furthermore, the cluster analysis showed that the transformants with lesser genetic differences with respect to the P. pastoris host strain are the recombinant strains with the highest level of recombinant protein production.
The examined population of Jersey cows was characterised by the occurrence of three genotypes of the casein protein fraction and four genotypes of whey protein.
Forty safflower genotypes were grown under normal irrigation and drought stress. In the first experiment, the allelopathic potential of shoot residues was evaluated using the sandwich method. Each genotype residue (0.4 g) was placed in a sterile Petri dish and two layers of agar were poured on that. Radish seeds were placed on agar medium. The radish seeds were cultivated without safflower residues as the controls. The length of the radicle, hypocotyl, and fresh biomass weight and seed germination percentages were measured. A pot experiment was also done on two genotypes with the highest and two with the lowest allelopathic activity selected after screening genotypes in the first experiment. Before entering the reproductive phase, irrigation treatments (normal irrigation and drought stress) were applied. Shoots were harvested, dried, milled and mixed with the topsoil of new pots and then radish seeds were sown. The pots with safflower genotypes were used to evaluate the effect of root residue allelopathy. The shoot length, fresh biomass weight, and germination percentage were measured. Different safflower genotypes showed varied allelopathic potential. The results of the first experiment showed that Egypt and Iran-Khorasan genotypes caused maximum inhibitory responses and Australia and Iran-Kerman genotypes resulted in minimum inhibitory responses on radish seedling growth. Fresh biomass weight had the most sensitivity to safflower residues. The results of the pot experiment were consistent with the results of in vitro experiments. Residues produced under drought stress had more inhibitory effects on the measured traits. Safflower root residue may have a higher level of allelochemicals or different allelochemicals than shoot residue.
In this paper we introduce a new way of analysing habitat preferences of plant communities using the genotype and genotype-by-environment interaction effects (GGE) biplot method. We took into consideration real data on six plant associations belonging to the Molinio-Arrhenatheretea class. A total of 241 phytosociological relevès collected with the Braun-Blanquet method were described by the Ellenberg indicators. The two-way classification relevès-by-indicators was analysed with GGE. As a result it is proposed the modified biplot that contains ellipses for making the plot clearer. Next, we present the equations that described both axes and allow adding into the biplot any other relevès or a mean of set of relevès. Based on the generated biplot it is possible to present an ecological characteristics of any type of plant community including its acceptable and typical habitat requirements. The ellipse ranges provide important information about the homogeneity and ecological spectrum of the analysed syntaxon. The constructed equations allow comparing new data with data analysed previously. Summing up, the GGE biplot method is useful for analysing and graphically presenting ecological preferences of plant communities in a simple and comprehensive way.
The study was conducted to examine slaughter parameters of three genotypes of meat type chicken: F1 crossbred derived from crossing of light hen of indigenous breed Greenleg Partridge with heavy meat type cocks, F2 crossbred being an effect of re-crossing obtained crossbred C x GP with meat type males and medium growing Hubbard JA 957, designed for a longer, 9-week production. Chicken were reared till 63rd day of age. Examined parameters were: dressing percentage, breast and leg meat yield and fatness. F1 crossbred (C x GP) were characterized by rather low body weight, typical for slow growing chicken and good musculature, especially breast. Re-crossing with meat type cocks affected significant (P < 0.01) improvement of the slaughter parameters. F2 crossbred (C x (C x GP) reached high body weight, typical for medium growing chicken. In comparison with Hubbard JA 957 F2 crossbred had lower body weight, the same dressing percentage, better breast and worse leg musculature, and less abdominal fat. High breast meat percentage in carcass and less fatness suggested, that these chickens can be used in meat production, as a medium growing material designed for a longer fattening period (9 weeks).
