A novel approach to DNase I-sensitivity analysis was applied to examining genes of the spermatogenic pathway, reflective of the substantial morphological and genomic changes that occur during this program of differentiation. A new real-time PCR-based strategy that considers the nuances of response to nuclease treatment was used to assess the nuclease susceptibility through differentiation. Data analysis was automated with the K-Lab PCR algorithm, facilitating the rapid analysis of multiple samples while eliminating the subjectivity usually associated with Ct analyses. The utility of this assay and analytical paradigm as applied to nuclease-sensitivity mapping is presented.
In view of new civilizational challenges of the 21st century, health promotion and education encouraging active participation in physical recreation, which should be implemented at all levels of education, within family, peer environment and local environment, become issues of particular importance. The research was carried out in the summer of 2013 and included 572 students of John Paul II State School of Higher Education in Biała Podlaska (PSW), of such degree courses as Tourism and Recreation, Public Health, Nursing, Rescue Medicine and Computer Science. The method used in the research was a diagnostic survey based on International Physical Activity Questionnaire (IPAQ). The aim of the study was to get to know the differentiation between the forms of recreational physical activity taken up and expected by the students of PWS and their gender, BMI and self-assessment of physical fitness. Basing on the results of the analysis it was concluded that the abovementioned independent variables have significant impact on the forms of physical activity chosen and expected by the surveyed. Studies of the literature on the subject and the results of our previous research entitle us to conclude that university students, who in the near future will constitute the intellectual elite of the country, should be aware of the influence of physical activity on general health, the importance of healthy lifestyle and their role in promoting healthy attitudes. However, the research showed that the students are not ready to perform such tasks, therefore the actions preparing them to lead a healthy lifestyle need to be intensified.
The investigation was conducted in a 32-year-old pine stand, growing on a fresh coniferous site in western Poland. The strip roads in the stand had been designed and cut along tree rows 5 years prior to the investigation. The analysis focused on trees growing at 12 strip roads, in 3-meter-wide stripes on both sides of the strip roads and in control zones, half-way between each two neighbouring strip roads. The trees growing on the stripes along the strip roads were divided into three zones corresponding to tree rows. The first zone (I) was formed by trees growing in the row directly at the strip roads, while the zones two (zone II) and three (zone III) were formed by trees growing in the second and third rows of trees away from the strip roads, respectively, on both sides of the strip roads. The average distances of the zones two and three from the strip roads were 1.5 and 3 m, respectively. The diameters at breast height (DBH) of all the trees growing in the respective zones were measured. Moreover, the widths of the strip roads were measured, which resulted in dividing the strip roads into two groups: the wider strip roads (average width 3.6 m) and the narrower ones (average width 2.9 m). The statistical analysis showed significant differences in DBH values between the analysed zones, for nine of the strip roads. Both in the case of the wider strip roads, as well as in the case of the narrower ones, the trees growing directly along the strip roads were usually thicker than the more distant trees. A statistically significant difference between the DBH values of trees from zone I and II was found for one strip road. In most cases the trees from zone I were statistically thicker than the trees growing in the control zone.
The article presents an example of applying multidimensional statistical methods in the ranking of provinces in terms of the amount of funds received from the selected programs implemented under the Rural Development Programme (RDP).The purpose of this paper is to build the ranking and specify the homogeneous groups of provinces in regard to the analysed attributes. On the basis of volatility analysis and a matrix of correlation coefficients, six variables characterizing this issue were chosen for the final calculation. The paper uses three methods which enable multidimensional statistical analysis, i.e. the measure of Hellwig, the Czekanowski Diagram and Prim’s dendrite. All the mentioned methods made it possible to draw the same conclusion, namely that the provinces which have been using EU funds supporting agricultural activities to the greatest extent are zachodnipomorskie, pomorskie and warminsko-mazurskie. The lowest positions are occupied by podkarpackie, małopolskie and śląskie.
In the human body, stem cells are located in niches, which are extremely complex microenvironments (with specified oxygen conditions and cellular together with extracellular matrix components arranged as a 3D structure). The influence of signals from niches seems to play an important role in maintenance of stem cells pluripotency and in their differentiation. We have been investigating the influence of the different niche components on the proliferation and differentiation of neural stem cells into specific cell types as well as the molecular mechanisms underlying this cell responses. In this study we are investigating the influence of low oxygen tension conditions on proliferation and differentiation of Human Umbilical Cord Blood of Neural Stem Cell (HUCB-NSC). Human Neural Stem Cells (NSC) in their physiological niches are exposed to 2–8% oxygen level. For that purpose, HUCB-NSC, were cultivated in two oxygen tension conditions: 21% and 5% with or without the presence of differentiation factor dBcAMP (N6,2′-O-Dibutyryladenosine 3′,5′-cyclic monophosphate sodium salt). We compared the expression of the markers characteristic for proliferation (Ki67) as well as neuronal and astroglial lineage commitment (MAP2, GFAP, β-tubulin, NF200). The presence of tested markers was revealed on the protein (immunocytochemistry) and gene expression level (Real-Time PCR). Our data show, that the low oxygen tension promote HUCB-NSC differentation into neuronal lineage.
