The effect of sulforaphane on human lymphoblastoid cells originating from a patient of a high cancer risk was studied. Sulforaphane (SFN) is a naturally occurring substance of chemopreventive activity. In our study, changes in cell growth, induc­tion of apoptosis and phase 2 enzymes as well as glutathione level were examined. Apoptosis was tested by confocal microscopy at three stages: change in mitochon­drial membrane potential, caspase activation and phosphatidylserine externalization. We show that SFN increases the activity of the detoxification system: it in­creases quinone reductase activity at low concentration (0.5-1 uM) and raises glutathione level in a dose-dependent manner. At higher doses (2.5-10 uM) sulforaphane is a cell growth modulator, as it caused cell growth cessation (IC50 = 3.875 uM), and apoptosis inducer. The results obtained suggest that sulforaphane acts as a chemopreventive agent in human lymphoblastoid cells.
Since decades, it has been suggested that regular, low to moderate consumption of red wine, a major component of Mediterranean diet, at main meals, may contribute to explain the healthy properties attributed to this traditional dietary style. Despite preclinical in vitro/in vivo data have shown many signifi cant pharmacological activities of grape phytochemicals, mostly polyphenols, evidence in humans is still debated. This lack of consensus may be due to the equilibrium between the two main components of wine relevant for health: alcohol and phytochemicals. Because ethanol is a major risk factor in oral carcinogenesis, in this commentary, we briefl y discuss the relationship between the type of alcoholic beverage and oral cancer in European countries.

The products of plant origin are a rich source of biologically active substances, both nutritive and referred as anti-nutritive. A large group of these compounds are substances with antioxidant activity that fights against free radicals. In the family of Brassicaceae vegetables, Brassica, is the largest and most widely consumed a group of plants in Europe and all over the world. They are characterized by different levels of nutrients. However because of their large and frequent consumption, they may become a significant source of nutrients and bioactive compounds in the daily diet. The beneficial effects of Brassica vegetables on human health have been somewhat linked to phytochemicals. They prevent oxidative stress, induce detoxification enzymes, stimulate immune system, decrease the risk of cancers, inhibit malignant transformation and carcinogenic mutations, as well as, reduce proliferation of cancer cells. Brassica vegetables contain a lot of valuable metabolites, which are effective in chemoprevention of cancer, what has been already documented by numerous studies. Due to the presence of vitamins C and E, carotenoids and antioxidant enzymes such as catalase, superoxide dismutase (SOD) and peroxidase, these vegetables are considerable source of antioxidants, and due to the presence of polyphenols and the sulfur-organic compounds exert also antimutagenic action. Moreover, these vegetables are also rich in glucosinolates, which are unstable compounds and undergo degradation into biologically active indoles and isothiocyanates under the influence of enzyme presented in plant tissues- myrosynase. These substances through the induction of enzymatic systems I and II phase of xenobiotics metabolism may affect the elimination or neutralization of carcinogenic and mutagenic factors, and consequently inhibit DNA methylation and cancer development. Despite many healthy benefits upon eating of cruciferous vegetables, it has been also seen a negative impact of their certain ingredients on the human body.
Solar ultraviolet radiation (UV) is a major cause of non-melanoma skin cancer in humans. Photochemoprevention with natural products represents a simple but very effective strategy in the management of cutaneous neoplasia. The study investigated the protective activity of Calluna vulgaris (Cv) and red grape seeds (Vitis vinifera L, Burgund Mare variety) (BM) extracts in vivo on UVB-induced deleterious effects in SKH-1 mice skin. Forty SKH-1 mice were randomly divided into 4 groups (n=10): control, UVB irradiated, Cv + UVB irradiated, BM+UVB irradiated. Both extracts were applied topically on the skin in a dose of 4 mg/40 µl/cm2 before UVB exposure - single dose. The effects were evaluated in skin 24 hours after irradiation through the presence of cyclobutane pyrimidine dimers (CPDs) and sunburn cells, tumor necrosis factor-alpha (TNF-), interleukin (IL)-6 levels. The antioxidant activity of BM extract was higher than those of Cv extract as determined using stable free radical DPPH assay and ABTS test. One single dose of UVB generated formation of CPDs (p<0.0001) and sunburn cells (p<0.0002) and increased the cytokine levels in skin (p<0.0001). Twenty hours following irradiation BM extract inhibited UVB-induced sunburn cells (p<0.02) and CPDs formation (p<0.0001). Pretreatment with Cv and BM extracts resulted in significantly reduced levels of IL-6 and TNF- compared with UVB alone (p<0.0001). Our results suggest that BM extracts might be a potential candidate in preventing the damages induced by UV in skin.
