The effect of crude extracts of neem (Azadirachta indica) leaf, neem seed and garlic (Allium sativum) at concentrations ranging from 5% to 30% of the material in 100 ml of Potato Dextrose Agar on mycelial growth of Fusarium oxysporum f. sp. lycopersici was assessed. All the extracts inhibited mycellial growth at various levels. Dry neem seed extract gavel 100% inhibition of mycelial growth. Fresh neem leaf extract reduced mycelial growth with increasing concentration while in garlic there were no differences in growth inhibition among the various concentrations used. However garlic extracts decreased sporulation with increasing concentration and cultures grown on extract amended agar plates remained viable.
We report the first case of chafer beetle [Anisoplia austriaca (Herbst 1783)] mortality caused by Actinomucor elegans var. elegans in wheat fields of the Kurdistan province, Iran. For three years, dead larvae of Anisoplia austriaca were collected from wheat fields of the Kurdistan province. Similar isolates of a fast-growing fungus were recovered from all samples. The fungal isolates were identified as A. elegans var. elegans based on morphological and cultural characteristics. The identity of the species was further confirmed using sequence data of the ITS (Internal Transcribed Spacer) region of ribosomal DNA. Koch’s postulates were fulfilled by the inoculation of the larvae of A. austriaca and Galleria mellonella (Linnaeus, 1758) (as the model insect) using the spore suspension of A. elegans var. elegans. The viability of sporangiospores was evaluated using a spore dilution technique on germination medium. The results on the pathogenicity (100% mortality in A. austriaca larvae) and viability tests (germination: 95.45%) demonstrated that A. elegans var. elegans can be considered as a potential biocontrol agent against the chafer beetle. Field experiments are still required to evaluate the capacity of A. elegans as a biological control agent.
Fusarium species are causal agents of fungal diseases occurring frequently in numerous agriculturally important plants, including potato, garlic and are one of the common pathogens of tomato, causing root rot in the west part of Iran. Therefore, the objectives of this study were to isolate and identify disease-causing Fusarium species from infected tomatoes based on the morphological and molecular characteristics. Twenty-five isolates of Fusarium were obtained from infected root of tomato plants collected from the fields in different regions of western Iran. Based on morphological features, the strains were classified into four following Fusarium species: F. oxysporum, F. redolens, F. proliferatum and F. verticillioides. The phylogenetic trees based on tef1 and tub2 dataset clearly distinguished closely related species. All of the isolates were evaluated for their pathogenicity on healthy tomato seedlings in the greenhouse. This is the first report on molecular identification of Fusarium species isolated from tomato plants cultivated in Iran.
Bakanae disease on rice has been recorded almost in all countries where paddy is grown commercially, especially in Asian countries, including Malaysia and Indonesia. Bakanae disease was widespread in Peninsular Malaysia and three provinces of Indonesia with the range of disease severity from scale 1 to 5 and disease incidence from 0.5 to 12.5% during 2004-2005 main growing seasons. A total of five Fusarium species belonging to section Liseola and their allied i.e. Fusarium fujikuroi, F. proliferatum, F. sacchari, F. subglutinans and F. verticillioides were isolated and identified on the basis of their morphological characteristics. Literature data showed that the bakanae disease of rice all over the world is caused by F. fujikuroi and probably some other Fusarium species from section Liseola or allied. However, from pathogenicity tests that have been carried out by using variety MR 211 of rice it was evident that F. fujikuroi was highly virulent and the only species involved in causing bakanae disease. Therefore, this species was the only one detected to be able to produce gibberellic acid - (GA3) with Rf value 0.40 and 0.62, developed in solvent systems isopropanol:ammonia:water (10: 1: 1), v/v/ v and chloroform:ethyl acetate:formic acid (5: 4: 1), v/v/v, respectively. This knowledge would be invaluable in developing our understanding on the interaction between F. fujikuroi and the host plants.
