On the basis of morphological features and RAPD markers the strains of Chalara ovoidea found in Poland on planks and on stems of beech trees were identified. As reference strains the cultures taken from CBS Utrecht were employed; they were cultures CBS 354.76 and CBS 136.88. The amplification of genomic DNA was conducted using 10 primers (OPA01-OPA10), 7 of which (OPA01-OPA05, OPA09, OPA10) gave positive results. In total 42 fragment of DNA (bands) were obtained. In case of primers OPA03, OPA04, OPA05, and OPA09 all obtained fragments for analyzed strains were fully monomorphic. This means, that no genetic variability was found using the above mentioned primers. Low genetic variability was ascertained in the analysis of frequency of occurrence of DNA fragments using other primers, namely OPA01, OPA02, and OPA10. The matrix and dendrogram of genetic affinities among different strains of Chalara, calculated using the Jaccard’s similarity coefficient suggested, that the most similar strains are the ones coming from Poland (HMIPC 16136 and HMIPC16664) as well as the strain CBS 136.88, while somewhat different from them is the strain CBS 354.76. To determine, how exactly did the dendrogram reflect genetic affinity among analyzed strains, the Mantel’s test was employed. The correlation coefficient amounted to 0.78, suggesting that the strains under study had been grouped properly. The results showed, that the fungal strains found in southern Poland represent the species Chalara ovoidea.
The distribution of chlorophyll was examined in cross-sections of 2- and 6-year-old stems as well as in the bark of the stump trunk of beech trees, utilising chlorophyll autofluorescence. The investigations were conducted using a confocal microscope. The tests carried out on 2 - 6-year old stems showed a large presence of chlorophyll in the bark, in primary and secondary rays as well as in the pith, but smaller amounts in wood parenchyma cells. There was no chlorophyll in the cork, sclerenchyma: in wood in vessels, tracheids and fibers. A reduction in the chlorophyll content in 6-year-old stems was not observed. In the bark of the trunk, chlorophyll was found in large amounts directly under the cork and in vestigial amounts in the primary phloem.
The author investigated a 0-3 cm layer of two forest brown soils: brown acid and brown leached. These forest soils were under handsome beech trees (Fagus sp.) between 100 and 120 years old. The author compared the chemical properties of the soils. The soil existing near the tree trunk (1 m2 of surface, strongly washed by rain water polluted by chemical substances) and the soil existing under the tree umbrella (40 m2 of surface in the area of the tree-tops, washed poorly by rain water). This comparison showed changes in these properties, dependent upon the degree of atmospheric contamination. It was found that, at the same level of atmospheric contamination, higher changes of properties were found in brown leached soil than in brown acid soil. Less acidified soils (with alkaline or neutral pH reaction) are more suitable in these researches. The investigations of the forest soil layer 0-3 cm (12 soil profiles) showed that changes in their chemical properties in relation to tree trunk-tree umbrella can be the basis of assessment of the degree of atmospheric contamination.
