Our objective was to determine the content of macro- and micronutrients, toxic heavy metals as well as total phenols in 10 broccoli (Brassica oleracea var. italica) cultivars. The experiment was designed at the Poznań University of Life Sciences, Poland, and completed during two plant growing seasons. Broccoli seedlings were planted in a field in early July and the plants were harvested between September and October. The content of general forms of nitrogen was measured by distillation with the Kjeldahl method. The content of potassium, calcium and sodium was assessed by flame photometry, and the content of magnesium, cadmium, lead, nickel, copper, zinc, manganese and iron was determined by flame atomic absorption spectroscopy (FAAS). The content of heavy metals, macronutrients and sodium in the broccoli heads depended on a cultivar. Having analyzed the levels of macronutrients in ten broccoli cultivars, it was concluded that cv. Agassi and Tradition had the highest content of nitrogen and phosphorus. A very high amount of phosphorus was also found in cv. Beaumont. The highest concentrations of calcium and potassium were found in cv. Monte Carlo. The cultivars Agassi, Beaumont, Tiburon and Tradition were characterized by the highest content of magnesium. The highest content of sodium occurred in the cultivars Beaumont and Monopoly. With respect to micronutrients, the highest content of iron, copper, zinc and manganese was revealed in cv. Agassi. The highest content of nickel was determined in the cultivars Beaumont and Monte Carlo. In addition, the cultivar Monte Carlo was characterized by the highest content of lead, and Beaumont had the highest content of cadmium. However, the maximum permissible levels of cadmium and lead were not exceeded in the heads of the ten broccoli cultivars. In both years of the study, the highest content of phenols was found in cv. Steel and the lowest one appeared in cv. Agassi.
Róże brokułów poddano obróbce cieplnej przy zróżnicowanych parametrach temp. pary wodnej (90°C- 125°C) i czasu (5-13 min.) oraz kombinacji gorącego powietrza i pary wodnej (temp. 200°C, 2 min.). Jako wariant odniesienia zastosowano gotowanie w wodzie w czasie 15 min. Z surowych i poddanych obróbce cieplnej brokułów otrzymywano ekstrakty metanolowe, w których określano efektywność zmiatania rodnika DPPH oraz oznaczano zawartość związków fenolowych ogółem. Stwierdzono korzystny wpływ obróbki parą wodną na zachowanie właściwości przeciwutleniających brokułów w porównaniu do gotowania w wodzie. Wskazano istnienie korelacji (r = -0,96) między EC50, a zawartością polifenoli.
The study was designed to determine the possible relationship between Brassica oleracea var. italica seedlings stored at 2°C in the dark for seven and fourteen days, respectively, and the level of certain antioxidant parameters in particular organs. A parallel objective of the experiment was to determine if the reaction of seedlings to low temperature might be persistent in fully developed plants until harvest time. After 14 days of chilling a significant increase in the glutathione content was observed in the seedling leaves in comparison to the non-chilled plants. During vegetation in field conditions this effect was maintained in leaves up to the stage of formation of flower buds. At harvest the highest content of glutathione was demonstrated in broccoli heads, obtained from plants, which were previously chilled in the seedling phase for two weeks. Peroxidase activity in broccoli seedlings increased each year of the three-year study due to the duration of the cooling time, whereas in the case of catalase the changes were not so distinct. At harvest time the activity of both enzymes in the leaves and flower buds fluctuated according to the particular year of study.
Broccoli (Brassica oleracea L. var. italica) contains numerous biologically active compounds. However, their levels change during processing and storage. The objective of the research study was to determine the effect of short-term storage (1 ÷ 4 days) of broccoli heads at different temperatures (3 ºC, 16 ºC, and 21 ºC) on the contents of phenolic acids, selected flavonoids (quercetin and kaempferol), and tocopherols. Broccoli heads were harvested in autumn when they reached their maximum size and their buds were 2 mm in diameter. The content of phenolics and tocopherols depended on the storage time of broccoli heads. The lowest content of phenolics was found immediately after the broccoli was harvested, whereas the highest content thereof was reported after 4 days of storage. However, the storage temperature did not impact the contents of those compounds. In the research study, the content of caffeic acid was reported to be highest in the fresh broccoli heads; the content of sinapic acid was the second-highest, and that of the ferulic acid was the third-highest. The content of kaempferol in the broccoli heads was higher than that of quercetin. No differences were reported in the content of quercetin and kaempferol at particular storage temperatures on every single day of the experiment. An increase was reported only when compared with the control sample (the 0 day of storage). In the broccoli heads, the predominant tocopherol was α-tocopherol. The lowest loss in the total tocopherols occurred while storing at a temperature of 3 ºC and the highest: at a temperature of 21 ºC.
The morphological and histological changes which occur in the cold-induced apical meristem of broccoli (B. oleracea var. italica cv. ‘Fiesta’) during transition from the vegetative to the generative phase have been demonstrated in our previous investigations. Now, a light microscopic study on the micromorphological parameters of the tunica and corpus of the meristem was undertaken to ascertain their association with devernalisation. Changes in the apex of broccoli seedlings grown for 26 days at 24oC, succeeded by 35 days of cold treatment (16oC - vernalization) followed by 14 days of warm treatment (24oC - devernalization), were studied and described. Microscopic analysis according to the new method of slide preparation showed cytohistological zonation and the spatial distribution of cells in the central and peripheral zones of the broccoli shoot meristem more precisely than possible hitherto. Comparison between subsequent stages of meristem reorganization after devernalization revealed variation in the structure of the apex during its transition to the evocation phase of flowering. The results of measurements show that the tunica width changes irregularly, increasing from its minimum value (112.39±5.78 μm) through the medium one to the maximum values (260.32±2.50), to decrease again, while its height increased and decreased sinusoidally from its minimum value (21.52±4.30μm) to the maximum one (76.98±7.44μm) and then decreasing again to the minimum value (21.52±4.30). The stratification of the tunica as well as its width and height revealed relatively low variation within the analyzed time. These structural traits correlated with cold/warm treatments may be further utilized in the broccoli breeding programme against developmental disturbances.
The factors regulating callus proliferation and bud regeneration from stigma tissues are not sufficiently understood. To study the regenerative capacity of pistil elements, pistil of broccoli was cultured under simple culture conditions. Stigmas with style from broccoli pistils undergo somatic embryogenesis on Murashige and Skoog basal medium. Callus initiation occurred on basal medium supplemented with 4 mg·l-1 BAP, 1.6 mg·l-1 2,4-D, 250 mg·l-1 casein hydrolysate and 30 g·l-1 sucrose. Proembryo induction was observed after two callus subcultures. Calluses with globular embryos were cultured on basal medium with 2 mg·l-1 BAP, 1 mg·l-1 IAA and 40 g·l-1 sucrose for development, maturation and germination of somatic embryos. A population of somatic embryos was maintained on medium containing 1 mg·l-1 BAP and 2 mg·l-1 NAA only. Adding NAA to MS medium containing BAP considerably enhanced root formation. After acclimatization, all plantlets developed well and produced phenotypically normal flowers.
Temperature is a factor which strongly affects the yield and broccoli head quality. Temperature determines the date of head initiation and thus affect the length of vegetation period. An experiment the aim of which was to assess the impact of temperature on the yield and the length of the period from the broccoli head initiation until the beginning and end of harvest as well as the length of the harvest period was carried out in years 1997–1999. The seedlings of cv. Fiesta were planted in April, May, June and July. The phase of head initiation was determined on the basis of analyses of microscopic preparations. The highest total yield was obtained from May and June planting dates, the highest marketable yield was recorded in the case of the May date planting. A higher correlation was found between the sum of temperature and mean daily air temperature in the period from the initiation to the beginning and end of harvest and mean weight of marketable heads than between temperature and the total yield. The highest correlation was determined between the mean air temperature and the number of days from the head initiation to the last harvest.
Brokuł jest warzywem, którego znaczenie gospodarcze w Polsce systematycznie rośnie. Jest ceniony przez konsumentów za walory smakowe i zdrowotne. Wielkość plonu i jakość róży brokułu zależą bezpośrednio od zaopatrzenia roślin w pierwiastki w ciągu sezonu wegetacyjnego. Przedstawiono wyniki stanu odżywienia makroelementami roślin brokułu, na podstawie ich zawartości analizowanej w częściach wskaźnikowych (nerw główny liścia), w dwóch fazach rozwoju. Analizę uzależniono od dwóch czynników agrotechnicznych: metody uprawy (siew wprost do gruntu i rozsada) oraz rozstawy roślin (20, 30, 40 i 50 cm x 67,5 cm). W prezentowanej pracy zakres zawartości badanych składników wyniósł kolejno w fazie wzrostu wegetatywnego i fazie wiązania róż (g·(100 g)-1 s.m.): NH4 + 0,11-0,80 i 0,09-0,43; NO3 - 0,49-5,73 i 0,34-3,30; P 0,17-0,31 i 0,24-0,41; K 2,37-5,74 i 2,22-3,09; Ca 0,92-2,27 i 0,93-2,80; Mg 0,16-0,41 oraz 0,20- 0,44. Poziom analizowanych makroelementów był zróżnicowany nie tylko pomiędzy obiektami doświadczenia, ale również pomiędzy latami badań. Prezentowane dane pozwalają na diagnozę stanu odżywienia brokułu w fazach determinujących wielkość i jakość plonu.
Italian broccoli (Brassica oleracea L.var. botrytis italica), Monaco variety from field culture and Lorix variety from greenhouse cultivation as well as savoy (Brassica oleracea L.var. sabauda) Langendijker variety from greenhouse cultivation were investigated. The plants were stored in the controlled atmosphere (CA) consisting of 4% CO2, 3% O2, 93% N2, at temperature 0-1°С and in the air (K), for comparison. Vitamin C content, its degradation kinetics, antioxidating properties and sensoric features (general view, colour) were analysed. It was shown that CA slowed down the rate of vitamin C degradation about twice as compared to air storage. The degree of vitamin C degradation is different depending on plant variety. The slowest degradation is observed in the Italian broccoli of Monaco variety stored in CA, and the quickest one in savoy stored in the air. Vitamin C decomposition applies to the 1st order kinetics (control samples) or 0th order kinetics (CA).
