The influence of pre-sowing seed treatments on germination, seedling growth, ion accumulation and plant antioxidant system in spring wheat (Triticum aestivum L.) cultivar MH-97 was examined under control and saline conditions. Albeit, all pre-sowing seed treatments were effective in improving seedling fresh and dry weight under saline conditions, the effect of hormonal priming with kinetin was very pronounced particularly in improving seedling vigor. The plants raised from seeds primed with kinetin have maximum root and shoot lengths under saline conditions while pre-sowing chilling treatment increased shoot length maximally whereas halopriming increased root length maximally under non-saline conditions. The Na⁺ uptake was decreased while K⁺ uptake was increased in plants due to hormonal priming with kinetin and hydropriming under stress conditions. Except halopriming, all seed treatments significantly increased leaf protein contents under non-saline conditions, however decreased under saline conditions. Halopriming resulted in a significant increase in protein concentration. Catalase (CAT) activity was significantly increased under salinity stress, however, hormonal priming and halopriming treatments maximally increased CAT activity. All the seed treatments were effective in increasing superoxide dismutase (SOD) level but hormonal priming with kinetin maximally increased its activity under stress conditions. Although all pre-sowing seed treatments were effective in alleviating the adverse effect of the salt stress on wheat plants, their effects on altering the concentration of different ions, seedling growth and plant antioxidant system were different in the wheat plants.
Effect of D-mannose treatment on different antioxidants, phenolics, protease activity, lipid peroxidation, DNA damage and cell death was investigated in coleoptiles of etiolated wheat seedlings. Modulations in these biochemical parameters were monitored up to 96 h after treatment at 24 h intervals. With accelerating effect on initial signs of cell death, i.e., appearance of long DNA fragmentation and no effect on initiation of terminal stage, i.e., internucleosomal nDNA fragmentation, mannose treatment (1 % = 56 mM) diminished the antioxidant activities in wheat coleoptiles. Mannose treatment decreased the catalase activity at all intervals, while APX and POD activities decreased at 72 h. Peroxidation of lipids increased at 72 h after mannose treatment. Levels of most of antioxidants, i.e., SOD, peroxidases and phenolics were raised during initial time period (24–48 h) of mannose treatment probably as an attempt to counter the stress effect. Protease activity gradually increased and protein content decreased with time in both treated and non-treated coleoptiles. Sharp decrease in CAT, APX and peroxidase activities and increase in lipid peroxidation at 72 h overlaps with apoptotic internucleosomal nDNA fragmentation in this organ. This coincidence points towards the importance of compromised antioxidant defense and involvement of reactive oxygen species in initiation of terminal stage of programmed cell death in wheat coleoptile. In conclusion, accelerating effect on DNA fragmentation and lipid peroxidation along with diminished antioxidant activities at the time of internucleosomal nDNA fragmentation, provide evidence for pro-apoptotic effect of D-mannose in wheat coleoptile.
Suaeda fruticosa Forssk is a leaf succulent obligate halophyte that produces numerous seeds under saline conditions. Seeds are a good source of high quality edible oil and leaves are capable of removing substantial amount of salt from the saline soil besides many other economic usages. Little is known about the biochemical basis of salt tolerance in this species. We studied some biochemical responses of S. fruticosa to different exogenous treatments under non-saline (0 mM), moderate (300 mM) or high (600 mM) NaCl levels. Eight-weekold seedlings were sprayed twice a week with distilled water, hydrogen peroxide (H₂O₂, 100 µM), glycine betaine (GB, 10 mM), or ascorbic acid (AsA, 20 mM) for 30 days. At moderate (300 mM) NaCl, leaf Na⁺, Ca2⁺ and osmolality increased, along with unchanged ROS and antioxidant enzyme activities, possibly causing a better plant growth. Plants grew slowly at 600 mM NaCl to avoid leaf Na⁺ buildup relative to those at 300 mM NaCl. Exogenous application of distilled water and H₂O₂ improved ROS scavenging mechanisms, although growth was unaffected. ASA and GB alleviated salt-induced growth inhibition at 600 mM NaCl through enhancing the antioxidant defense system and osmotic and ion homeostasis, respectively.
Tomatoes reportedly have a positive response to seed priming. The present study evaluates the effects of ethanol priming on germination, seedling vigour and antioxidative responses of tomato seeds. Priming was achieved by exposing seeds of ‘Roma’ and ‘Nagina’ to 2, 4 and 6% aerated ethanol solutions for 24 h. Priming with low levels (2 and 4%) of ethanol improved seed germination, seedling vigour and enhanced antioxidative activity that results in better performance of tomato seeds. However, priming with 6% ethanol failed to improve seed germination and seedling development which relates to the decreased anti-oxidative activity in tomato seeds due to high ethanol level.
In this study, the biocontrol abilities of water-soluble and volatile metabolites of three different isolates of Trichoderma (T. asperellum,T. harzianum and Trichoderma spp.) against soil borne plant pathogen Rhizoctonia solani were investigated both in vitro and in vivo. The results showed for the first time that mycelial growth inhibition of the pathogen was 74.4–67.8% with water-soluble metabolites as compared to 15.3–10.6% with volatile metabolites in vitro. In vivo antagonistic activity of Trichoderma isolates against R. solani was evaluated on bean plants under laboratory and greenhouse conditions. We observed that T. asperellum was more effective and consistent, lowering disease incidence up to 19.3% in laboratory and 30.5% in green house conditions. These results showed that three isolates of Trichoderma could be used as effective biocontrol agents against R. solani.
Azo dyes are the main group of dyes used in different industrial applications. These dyes are highly toxic for aquatic life, so their removal is of utmost importance before they can be disposed of in a main water body. The present study focused on degrading/mineralizing the synthetic reactive dye wastewater. Initial experiments were done with four indigenous white rot fungi. P. chrysosporium (PC) showed more potential toward degradation of synthetic dye wastewater than other three fungal strains, so it was selected for further optimization of different fermentation parameters. Maximum decolorization (84.8%) of reactive dye wastewater was obtained at pH 5, inoculum size 4 mL, and 30ºC. After optimizing experimental parameters, the effects of different nutritional factors like carbon and nitrogen sources were also studied. Decolorization of synthetic dye wastewater was increased from 84.8 to 89.2%, when rice bran was used as an additional carbon source. However, no increase in decolorization of synthetic dye wastewater was observed in the presence of nitrogen supplements. The screened fungal strain decolorized the wastewater up to 90%. The effect of different nutritional factors enhanced the degradation capability of the fungal strain under study. UV-visible and FTIR analyses confirmed the degradation of synthetic dye wastewater into simpler, non-toxic products.
