The effect of pro longed illumination (60 min) with photosynthetically active monochromatic radiation of low intensity (3 μmol-m⁻² ·s⁻¹) and high intensity (60 mmol m⁻² ·s⁻¹), corresponding to the physiological conditions and light stress conditions, respectively, was studied in the algae Nitellopsis obtusa. Illumination of Nitellopsis obtusa cells with strong light was associated with activation of the xanthophyll cycle, manifested by the deepoxidation of violaxanthin and accumulation of antheraxanthin and zeaxanthin. At the same time, the efficient singlet excitation quenching in the photosynthetic apparatus was activated, as demonstrated by the decrease in the intensity of the chlorophyll a fluorescence emission by ca 50 %. The difference of the fluorescence excitation spectra recorded before and after the light treatment match the difference absorption spectrum of the xanthophyll cycle pigments. The illumination with low light intensity resulted also in the chlorophyll a fluorescence quenching but the effect was very small (less than 10 %). The fluorescence quenching is interpreted in terms of the energy transfer between the Qy energy level of chlorophyll a and the 2¹Ag⁻ energy level of zeaxanthin. The singlet energy levels of carotenoids, corresponding to the green spectral region, are also taken into consideration in the interpretation of the excitation energy exchange between the carotenoids and chlorophylls. Possible molecular mechanisms involved in the activation of the strong and the weak excitation quenching, including violaxanthin isomerization, and possible physiological functions of such pathways of energy transfer are discussed.
Photoprotective efficiency of lutein, zeaxanthin and violaxanthin in liposomes made of digalactosyldiacylglycerol (DGDG) has been investigated. It was found that zeaxanthin is about ten times more effective than the two other xanthophylls in inhibition of chlorophyll a photo-oxidation. Kinetics of chlorophyll photobleaching suggests that exceptional photoprotective efficacy of zeaxanthin in membranes consists in effective scavenging of free radicals. High photoprotective efficiency of zeaxanthin in membranes correlates with its physiological function.
Carotenoids (neoxanthin, violaxanthin, lutein and β-carotene) present in plant cell play a role in photoprotection. Industrial pollution causes oxidative stress in plants, while carotenoids react with free radicals and dissipate the excess of excitation energy. In this way carotenoids prevent the negative influence of free radicals on metabolism and can even restore some of the damages. This is confirmed by results of our analysis of the level of xanthophylls in 16 and 17-year-old trees of Scots pine (Pinus sylvestris L.). Developed from seed collected in north-eastern, the trees grow in a relatively unpolluted control site, and in a polluted site located 2 km for away from the Phosphorus Fertilizer Works. In the polluted site the environment is contaminated with SO2, NOx and F, Al, F, Pb, Cu. The needles analysed in this study were visually undamaged. Material was collected in experimental plot from 6 trees in October'98 and April'99, between 12.00 and 13.00 hours, at full sunlight. The pigments were analysed quantitatively and qualitatively by thin-layer chromatography. The paper presents results of content and distribution of neoxanthin, violaxanthin, lutein and β-carotene of Scots pine needles from a healthy control tree and a stressed tree. Marked differences in pigment levels depended on the stage of needle development and level of pollution.
