Festuca arundinacea (Fa) is one of the most drought-tolerant species within the Lolium-Festuca complex. In the current work the protein level of chloroplastic Cu-Zn SOD (superoxide dismutase) in two Fa plants with extreme values of drought tolerance during exposition to water deficit was investigated. The obtain results revealed higher level of enzyme accumulation in more drought-tolerant Fa genotype. In less-drought tolerant plant the increase of dismutase level during stress treatment was only slight.
The aim of the study was to investigate the effect of different levels of protein in a diet on the incorporation of endogenous urea nitrogen (EUN) into individual amino acids (AA) of the ruminal bacteria of goats fed a low- (LP), medium- (MP), or high-protein diet (HP) in a 3 × 3 Latin square design. Three Alpine goats of about 35 kg body weight fitted with cannula into the rumen and catheter into the jugular vein were fed three isoenergetic diets containing 11% (LP), 13% (MP), or 16% (HP) crude protein in dry matter. The goats were infused for 6 days continuously with an 15N urea solution into the jugular vein. Ruminal bacteria were hydrolysed with 6M HCl. Next, butyl derivatives of free bacterial AA were obtained using HCl in butanol, then N-acylated using trifluoroacetic acid anhydride and analysed by gas chromatography using a mass-selective detector. The concentration of urea in plasma was 178, 356 and 667 mg · l–1 in goats from groups LP, MP and HP, respectively. 15N-excess during the infusion of labelled urea was significantly higher (P < 0.05) in the vast majority of AA of ruminal bacteria from goats fed the LP diet in comparison with goats fed the HP diet. Therefore, the level of protein in the diets affects the incorporation of EUN into bacterial AA. With the LP diet, EUN was incorporated mostly into glutamic acid, isoleucine and arginine, while in the case of the HP diet, into glutamic acid and arginine, as well as methionine. Regardless of the level of nitrogen in the diets, the incorporation of 15N into proline was very low. Irrespective of the dietary nitrogen level, EUN appears to be predominantly used for synthesis of glutamic acid in ruminal bacteria.
Possibility of reduction of protein level in feed mixtures of similar content of amino acids digestible in the small intestine and its influence on fattener’s performance and nitrogen retention were determined. Thirty pigs (35 kg) were divided into 3 groups. The control animals were fed with diets with crude protein level suggested by polish pig feeding standards (Normy żywienia... 1993) – 17% in grower and 15% in finisher diet. In the experimental groups (I and II) the protein level was reduced about 10 or 20% respectively. No significant effect on feed intake, weight gains and feed utilization was found. The animals from the both experimental group utilized less crude protein (P=0.001) than in Control group. The significant (P=0.001) reduction in nitrogen excreted in urine and total N-excretion in both group with reduced protein level in feed mixtures, were observed. Nitrogen retention in g/day was higher for group I compared to other groups and N-retention : N intake were significantly higher (P<0.05) in both experimental groups. The reduction of crude protein level in the experimental diets did not affect slaughter performance but in group II the tendency to increase (P=0.064) in back fat thickness and decrease meat content in carcass, were observed. The results suggest that lowering the protein level in diets for high-meat crossbred pigs (Polish Large White × Polish Landrace) × (Hampshire × Pietrain) from 170 and 150 g kg⁻¹ to 136 g and 115 g kg⁻¹ in grower and finisher diets, respectively, but supplemented with synthetic amino acids, did not impact negatively production results of fatteners, but decreased nitrogen emission to the environment and cost of feed.
Human mitochondrial polynucleotide phosphorylase (hPNPase) is an exoribonuclease localized in mitochondria. The exact physiological function of this enzyme is unknown. Recent studies have revealed the existence of a relationship between induction of hPNPase mRNA and both cellular senescence and growth arrest of melanoma cells following β-interferon treatment. The aim of this study was to verify whether the augmented hPNPase mRNA level results in increase of the protein level. In several cell lines established from five metastatic melanoma patients we did not find any such correlation. However, an elevated level of hPNPase protein was observed in interferon-induced HeLa and Jurkat cells. This increase was correlated with a slight shortening of poly(A) tails of mitochondrial ND3 transcript.
H. pylori ureA and ureB genes encoding both subunits of urease were expressed transgenically in a low-alkaloid line of tobacco (LA Burley 21). Analysis of transgene expression at both the mRNA and protein levels revealed a significant increase (up to 8-fold) in ammonia concentration correlated with an amount of UreB protein detected in the leaves, and an increase (up to 2-fold) in urease activity in transformants as compared to control plants.
