The purpose of the work was to adapt the methods of thrombocytes isolation from the peripheral blood, their absolute number determination and an attempt to evaluate their phagocytic activity. The elaborated technique of thrombocytes isolation in the ACD liquid enabled to separate in homologous plasma approximately 80 per cent of those cella, i.e. 23 850 in 1 mm³ of the plasma (± 7160). The above method allowed to preserve, like in the full blood, proper conditions for thrombocytes. It was found that the number of thrombocytes from healthy fish was 29 720 ± 6830 in one mm³ and the value of phagocytic index and per cent of phagocytic cells were 1.48 ± 0.38 and 18.83 ± 4.13, respectively. There was found 5 various forms of thrombocytes differing with the shape of the cell and nucleus, and the distribution and colour of the cytoplasm.
The aim of this study was to determine the influence of anticoagulants on dog platelet activation. The investigation was carried out by evaluating the expression CD61 and fosfatydyloserin as a marker of activation. In vitro data platelet activation was obtained by PMA and for the stabilization of the cell membrane procaine was used. Thrombopoesis was evaluated by reticulated platelets. It was shown that the platelets number was dependant on anticoagulants, and was greater in EDTA-3K blood, and the expression of CD61 was the largest on normal platelets in EDTA-3K platelet-rich plasma (PRP). The number of activated platelets was the highest among aggregates and the highest percentage of CD61+AnV+ did not depend on the use of anticoagulants. The percentage of platelets and microparticles with the expression of those antigens influenced by procaine, microparticles and aggregates influenced by PMA was higher in citrate PRP than EDTA-3K PRP. It was observed that anticoagulants did not have any influence on the percentage of reticulated platelets and the highest number of them can be found in among aggregates.

Blood platelet are of crucial importance in hemostasis. Little attention has been paid to their bacteriocidal activity and their potential role in parasitic infections. Activation of blood platelets may result from direct contact with a parasite or IgE, IgG, complement factors, lymphokines, CRP. Blood platelets reveale cytotoxic activity through release variuos inflamatory mediators, phagocitic activity and cooperation with other cells of immunologic system. The precise mechanism of all platelet cytotoxic effects remains ucleare.
