The prolactin receptor (PRLR) gene was studied as a candidate gene for the high prolificacy of Jining Grey goats. Polymorphisms in intron 1 and intron 2 of PRLR gene were detected in high prolificacy (Jining Grey) and low prolificacy (Boer, Wendeng dairy, Liaoning Cashmere and Beijing) native goats using PCR-SSCP. For intron 1, five genotypes (AA, AH, AK, HH and HK) were identified in Jining Grey goats, and two (AA and AK) in the other four breeds. The Jining Grey does of genotype HH, HK, AH and AK delivered by 0.65, 0.62, 0.59 and 0.57 more kids (P<0.01) than those of genotype AA, respectively. For intron 2, three genotypes (CC, CD and DD) were detected in Boer goats, and two (CC and CD) in the other four breeds. The Jining Grey does of genotype CD delivered by 0.55 (P<0.01) more kids than those of genotype CC.
The course and variability of cerebral base arteries in Capra hircus L was researched in 27 cerebra. It was observed that the vascular pattern of the species researched remains similar to the pattern observed in the members of Cervidae. The features characteristic for this species include the variability of descend of the rostral choroid artery as well as of the posterior cerebral artery.
The structural organisation of tunica intima in the aorta is important for its integrity, prediction, and diagnosis of atherosclerosis. The goat is a suitable model for cardiovascular studies, but the structure of its tunica intima is scarcely reported. This study, therefore, aimed to describe features of the goat aortic tunica intima by light and transmission electron microscopy. Sixteen healthy male domestic goats (capra hircus) aged between 6 and 24 months were used: 8 for light and 8 for electron microscopy. The animals were euthanised with sodium pentabarbitone 20 mg/mL and fixed with 3% phosphate buffered glutaraldehyde. For light microscopy, specimens from various regions of the aorta were routinely processed for paraffin embedding and 7 μm sections stained with Mason’s trichrome. Those for transmission electron microscopy were post fixed in osmium tetroxide, embedded in Durcupan, and ultrathin sections stained with uranyl acetate and counter stained with lead citrate. Endothelium comprises round and squamous cells, linked to the subendothelial material by a simple and sometimes lamellated basement membrane. In the subendothelial zone, a heterogenous population of cells are connected with interlinked collagen and elastic fibres. Both cells and fibres are connected to the internal elastic lamina. The composite structure and interlinkages in the tunica intima permit unitary function and increase mechanical strength, thus enabling it to withstand haemodynamic stress. The caprine arthritis-encephalitis virus (CAEV) was isolated from monocyte-derived macrophages (M/M), but not from PBMC of seropositive goats by co-cultivation with goat synovial membrane cells. Out of eight M/M co-cultures, CAEV was evidenced by the syncytia formation and presence of proviral DNA in two and four cultures, respectively. Two virus isolates from co-cultures showing cytopathic effects were further confirmed as CAEV by western blotting, PCR, and sequence analysis. The nucleotide sequence of gag gene showed 92.0% and 90.3% homology to the prototype CAEV-Co strain. Supernatants harvested from these cultures induced syncytia when cultured with uninfected cells and the resultant titer was 1035 and 1025 TCID50 per ml. New CAEV isolates are suitable candidates for further analysis of their genetic and biological properties.
The aim of the study was to assess the quality of pastures located in less-favoured areas (LFA) of Czech Republic, to evaluate the influence of that quality on the content of fatty acids of milk in grazing goats and in farm natural cheese made of their milk. Samples of forage were collected in 2008 and 2009, on 5 July, 9 August, 6 September and 4 October. Goat milk samples were taken on the same days of both years and additionally on May 31. The highest content of protein and fat of the forage dry matter and, consequently, the highest feeding value of pasture were found in July and October. It was associated with a high content of clovers and herbs in the available herbage. During grazing the content of saturated fatty acids (SFA) of goat milk fat continuously decreased while the trend for the content of monounsaturated fatty acids (MUFA) was opposite. A significant increase in herb of the total crop observed in autumn contributed to a highest content of polyunsaturated fatty acids (PUFA) in fat of goat milk. The fatty acid profile of goat cheese was related to that of the milk. Thus, a high content of PUFA, including CLA, observed in milk of grazing goats guarantees that the cheese made of that milk fulfills the requirements for functional foods.
The prevalence of coccidia infection in goats was evaluated in Western Pomerania (Poland) and West Ukraine Province. A total number of 311 goats were examined: 173 from Western Pomerania, including 139 kids (up to 6 month old), and 138 from the Lviv region including 93 juveniles. The study was based on two flotation methods, which helped to establish the number of oocysts in 1g of feces (OPG): Willis-Schlaaf for qualitative and McMaster for quantitative analysis. The species composition was established using Coudert’s (Coudert 1992) and Eckert’s (Eckert et al.,1995) keys. Nine Eimeria spp. were identified in feces samples in Western Pomerania and Lviv regions: E. arloingi, E. chrisienseni, E. jolchijevi, E. ninakohlyakimovae, E. alijevi, E. capina, E. caprovina, E. hirci, E. apsheronica. The prevalence of infection in Western Pomerania of adult goats was 74% with an intensity of 0–2500 OPG. The rate of infection in kids was 100% and the intensity ranged from 1800 to 28000 OPG. In West Ukraine Province, 100% of the adult goats and kids were infected. The coccidian intensity of infection ranged from 2600 to 120000 OPG in kids, from 50 to 4500 OPG in adults. Clinical conditions can influence the intensity of oocysts excretion in the feces of animals. In Western Pomerania the highest intensity of excretion of oocysts was during May–July and the lowest during November–January. In West Ukraine Province the peak of oocysts excretion was in May–July and the lowest in October–December. The results of the present investigation have implications for the control of coccidial infections in goats in Europe.
Epidemiological studies on caseous lymphadenitis were carried out in Poland in 1996 and 2002 among goat herds covered by a milk recording program. Between-herd seroprevalence was 13.2% in 1996 and increased to 62.5% in 2002. The average size of seropositive herds was statistically significantly higher than that of seronegative ones, however there was no statistically significant difference in the age between the herds. A statistically significant prevalence ratio (PR) was identified and relevant attributable risk for exposed animals (ARexp) was calculated for the following risk factors: presence of seropositive males in a herd (PR=8.350; ARexp=0.651), presence of superficial abscesses in animals (PR=6.142; ARexp=0.620), presence of respiratory signs (PR=2.900; ARexp=0.393), presence of animals in poor condition in a herd (PR=2.774; ARexp=0.390) and occurrence of reproductive failures in a herd (PR=1.798; ARexp=0.230). Purchase of animals from abroad, mastitis and husbandry conditions (housing system, grazing system, hygienic conditions) were not shown to be statistically significant risk factors.
The aim of this work was to trace differences in magnesium and calcium concentrations (both total and ionic form) between goats which were in late pregnancy and lactating goats. The study involved 30 goats. Blood samples were taken three times from each goat. Total concentrations of magnesium and calcium were determined with the colorimetric method and (A — 1.581 mmol dm-3; B — 2.052 mmol dm-3; C — 2.112 mmol dm-3). Ionic form of calcium based on using ion—selective analysis (A — 1.219 mmol dm-3; B — 1.126 mmol dm-3; C — 1.123 mmol dm-3). The study showed that mean value of both the total of calcium from each goat did not reach the reference level for this species. Ionic form of calcium was within the lever limit of the physiological norm. The content of magnesium in all the groups was within the range of reference concentrations (A — 1.051 mmol dm-3; B — 1.165 mmol dm-3; C — 1.117 mmol dm-3).
