During the years 1993-2005 232 dogs suspected of autoimmune mediated diseases (18 cases in years 1993-2001; 17 in 2002; 48 in 2003; 90 in 2004 and 59 in 2005) were examined. Detection of anti-erythrocyte antibodies was performed on 213 vein blood samples using Coombs test. Antinuclear antibodies (ANA) were estimated in 124 and circulating immune complexes (CIC) in 96 serum samples. In 13 cases immunohistochemical examination of skin or inner organ sections was done. The described methods were utilized in the diagnosis and treatment monitoring of immune mediated hemolytic anemia (IMHA) and systemic lupus erythematosus (SLE) cases. The frequency of positive results was estimated in clinically defined cases. Over 71% SLE or SLE-suspected patients presented positive ANA results, and all of them had elevated concentrations of CIC. In cases of IMHA/suspected IMHA, about 78% showed positive Coombs test while IgG class titers mostly exceeded 1:640.
The aim of the study was an attempt to elucidate canine haemangiopericytoma histogenesis based on immunohistochemical analysis using a range of commercially available antibodies. Seventeen canine haemangiopericytomas were examined. Histological analysis revealed the presence of spindle cells arranged in a circular pattern around capillaries (.fingerprint. pattern) or interlacing bundles. Fingerprint patterns were the distinguishing feature in all examined haemangiopericytomas. Immunohistochemical analysis revealed positive expressions of vimentin, neuron-specific enolase (NSE), glial fibrillary acidic protein (GFAP), S-100 protein (S-100) and smooth muscle actin (a-SMA). Negative reactions in tumor cells were observed for cytokeratin (CK), desmin (DES), epithelial membrane antigen (EMA) and VIII-related antigen factor (FVIII). Positive expressions of neuronal markers may not necessarily reflect the neural origin of tumors due to their low specificity. Immunoreactivity for α-SMA and DES negativity may suggest the presence of vascular smooth muscle cells or cells of pericytic origin. Moreover, the α-SMA antibody may be useful for a differential diagnosis of canine haemangiopericytomas.
The review describes current diagnostic techniques used for TSEs diagnosis with emphasis on BSE and scrapie in sheep. The clinical signs and their reliability are presented along with the review of laboratory tests. Special emphasis has been placed on the diagnostic reliability of clinical signs including their sensitivity and specificity. Histopatological examination for BSE is discussed with guidelines for diagnostic criteria on single section examination. Other techniques for PrP detection like: SAF detection, immunohistochemistry and western-blotting have also been presented. The review describes the development of novel methods for BSE diagnosis in live animals using cerobrospinal fluid and urine.
