A number of factors at all stages of data processing which affect the accuracy of determination of 15N relaxation parameters in 15N-labeled proteins is discussed. Methods which allow to improve accuracy of the determined parameters are presented using data obtained for Cucurbita maxima trypsin inhibitor.
The accuracy of the territory-mapping technique for estimating the abundance of densely breeding Hawfinches was tested in an old and unfragmented lime-oak-hombeam forest in the Białowieża National Park, E Poland. Hawfinch numbers estimated from counts of the whole bird community carried out with the application of the standards of the improved mapping technique were compared with seven-year data on the bird's true numbers, which are known from parallel intensive nest searches and persistent tracking of the movements of pairs. In a forest with a dense population of Hawfinches the mapping technique underestimated their numbers by 20% in years of moderate density and by 35% during high-density years. Even though the underestimation was negatively correlated with the true density of Hawfinches, the figures obtained by both methods reflected year-to-year changes in a similar way. An improvement in mapping data is achievable either by closer attention being paid to the species during standard visits (the best ones for surveying it), or post factum by the introduction of a correction factor into the mapping-technique figures.
The aim of the study was to test whether the methods using the playback technique produce accurate data of population size when compared to the standard mapping and nest-searching methods. The three-visit method with audio-stimulation was found to produce data of the same accuracy (100%, n = 11 territories) as the standard mapping method, but the nest-searching method ahd single-visit with audio-stimulation yielded slightly lower estimates, 91% and 82% respectively, in comparison to the previously mentioned methods. However, the three-visit method with audio-stimulation was 2.2 times less time consuming than the five visits which used the standard mapping method (9h vs 20h). The three-visit method with audio stimulation could be appropriate for assessing distribution and abundance, and also for monitoring purposes.
The primarily goal of plot-less sampling methods is to reduce costs and rapid survey of plant communities. First full inventory was conducted in two 30-ha sites of Saxaul populations geo-morphologically different. In first site (site I), population had random pattern while in second site (site II) clumped pattern was observed. Crown diameters and spatial situation of all trees were recorded using distance and azimuth. Data were transferred to computer and stem map was generated with ArcGIS Software. Distance sampling methods include point-centred quarter method (PCQ), joint-point method (JP), Random pairs method (RP), T-Square method (T-Sq) and Quartered neighbour methods beside fixed area plot (FAP), n-tree and variable area transect (VAT) methods were conducted on generated stem maps. A time study was done aiding indices determined in field works. In site I, point centred quarter estimator with measurements to the second closest individual in each quadrant had the lowest relative bias (RBIAS) in estimating density followed by 3-tree and closest individual methods. In clumped pattern (site II), variable area transect method with measurements to the 4th and 5th closest individuals in each transect brought the best results. The most time consuming methods after fixed area plot, were point centred quarter estimators while methods considering measurement to the closest individual were rapid. Considering RBIAS and Time together, VAT method was the best sampling method in clumped pattern followed by point centred quarter estimator with measurements to the second closest individual in each quadrant and closest individual estimators. In random pattern, point centred quarter estimator with measurements to the second closest individual in each quadrant was the best method followed by 3-tree and closest individual estimators. But for estimating cover per unit area N-tree methods performed well. As in this site, VAT method located in 4th grade, and due to simplicity of field works related to this method, in the case that the investigator would not be able to clearly define spatial pattern of the population, this method can be recommended as well.
Scientific literature and museum exhibits are full of explicit and implicit claims about the possible postures and motion ranges of dinosaurs. For the example of the prosauropod Plateosaurus engelhardti I assessed the motion range of limbs and vertebral column in a CAD program using a 3D virtual skeletal mount. The range of motion of the forelimb is very limited, allowing the grasping of objects placed directly ventrally and ventrolaterally of the anterior torso. The manus is adapted for grasping. The powerful fore limb can barely reach in front of the shoulder, making a quadrupedal walking cycle impractical. Only a digitigrade pose of the pes with a steeply held metatarsus is feasible, and the morphology of the stylopodium and zeugopodium indicates a slightly flexed limb posture. Hind limb protraction and retraction are limited by the pelvic architecture. The neck has significant mobility both dorsoventrally and laterally, but blocks torsion. The dorsal vertebral column is flexible to a degree similar to the neck, mainly in the anterior half, but blocks torsion totally in the anterior and posterior thirds. The anterior dorsals are similar in shape to the posterior cervicals and significantly increase the motion range of the neck. The tail is highly flexible due to its large number of elements, showing more lateral than dorsoventral mobility. These results are compared to reconstruction drawings and museum skeletal mounts, highlighting a pattern of errors specific to certain widely used reconstruction methods.
