Concentration of free cytoplasmic Ca2+ ([Ca2+]i) in Ehrlich ascites tumour cells loaded with fura-2 was measured in single cells applying a video imaging system. In resting cells [Ca2+li amounted to 60 - 340 nM and was increased after addition of 10 mM D-glucose or D-2-deoxyglucose by 80 - 200 nM. This increase occurred within 30 - 60 s following addition of the sugars and lasted for several minutes. Pretreatment of the cells with thapsigargin resulted in a much smaller [Ca2+]i increase after addition of glucose or deoxyglucose and, vice versa, thapsigargin added after the sugars mobilized less Ca2+ than when added before. A possible relation of the [Ca2+]i rise evoked by glucose and deoxyglucose to the Crabtree effect is discussed.
MALDI-TOF/TOF tandem mass spectrometry has been applied to determine the complete sequence of a CAPA-PVK in the Gulf Coast tick, Amblyomma maculatum. Single cell analysis allowed the identification of the amino acid sequence of Ixori-PVK (PALIPFPRV-NH2), a periviscerokinin which had previously been identified from two other ticks, Ixodes ricinus and Boophilus microplus. The identification indicates greater conservation of sequence for the CAPA-PVK/CAP2b family in ticks as compared with insects. Side-chain fragmentation experiments provided data to distinguish between Leu/Ile ambiguities. The tick CAPA peptide shows a high sequence homology with other members of the insect periviscerokinin/ CAP2b peptides, which are associated with the regulation of critical physiological processes such as diuresis. Thus, the identification of this neuropeptide will provide the experimental basis to better understand regulation of water balance in these arthropods, providing a potential opportunity to develop neuropeptide-based control strategies against these livestock pests.
Aging is a complex process involving morphologic and biochemical changes in single cells and in the whole organism. One of the most popular explanations of how aging occurs at the molecular level is the oxidative stress hypothesis. Oxidative stress leads in many cases to an age-dependent increase in the cellular level of oxidatively modified macromolecules including DNA, and it is this increase which has been linked to various pathological conditions, such as aging, carcinogenesis, neurodegenerative and cardiovascular diseases. It is, however, possible that a number of short-comings associated with gaps in our knowledge may be responsible for the failure to produce definite results when applied to understanding the role of DNA damage in aging and age-related diseases.
