In the present investigation the seeds of Pigeon pea (Cajanus cajan (L.) Millsp) were treated with different doses of gamma radiation (05KR, 10KR, 15KR, 20KR, 25KR, 30KR, 35KR, 40KR, 45KR, and 50KR) and concentration of Ethyl Methane Sulphonate (05mM, 10mM, 15mM, 20mM, 25mM, 30mM, 35mM, 40mM, 45mM, and 50mM) for studying seed germination, seedling height, (shoot and root), seedling injury, seedling vigour index, and seedling survival of plants at 30th day. The seed germination percentage was decreased with increased in the concentration/doses when compared to control. The LD50 (Lethal dose) value was determined based upon the seed germination percentage. The 50 percentage of seed germination and reduction was observed in 20KR of gamma rays and 25mM of EMS and it is considered as LD50 value for both the treatments. The decrease in seed germination was more prominent with gamma rays than that of EMS treatments. The seedling parameters of gamma rays and EMS treated seedlings were progressively decreased with increase dose/concentration in all mutagenic treatments when compared to control. The maximum seedling parameters were observed in 05KR of gamma rays and 05mM of EMS. Minimum seedling parameters were observed in 50mM of EMS and 50KR of gamma rays respectively.
The L5178Y (LY) murine lym phoma sublines LY-R and LY-S are dif fer en tially sen si tive to ion iz ing ra di a tion. The high ra di a tion sen si tiv ity of LY-S cells is re lated to im­paired rejoining of DNA double strand breaks. We found previously that the g-ray-induced base dam age is higher in the more radiosensitive LY-S subline. Here, we ex am ine the role of the re pair of ion iz ing ra di a tion in duced base dam age in re la tion to the radiosensitivity dif fer ence of these sublines. We used the GS/MS tech nique to es ti mate the re pair rates of six types of base dam­age in g-irradiated LY cells. All mod i fied DNA bases iden ti fied in the course ofthis study were typ i cal for ir ra di ated chromatin. The to tal amount of ini tial base dam age was higher in the ra di a tion sen si tive LY-S subline than in the ra di a tion re sis tant LY-R subline. The re pair rates of 5-OHMeUra, 5-OHCyt, 8-OHAde were sim i lar in both cell lines, the re pair rates of FapyAde and 8-OHGua were higher in the radiosensitive LY-S cell line, whereas the re pair of 5-OHUra was faster in its radio resistant coun ter part, the LY-R. Al to gether, the re pair rates of the g-ray-induced DNA base dam age in LY sublines are re lated nei ther to the ini tial amounts of the dam aged bases nor to the dif fer en tial le thal or mutagenic ef fects of ion iz ing ra di a tion in these sublines.
The effect of gamma irradiation and EMS treatment on seed germination and seedling height of Chick pea (Cicer arietinum. L.). In this regard Co-4 variety of chick pea was subjected to different doses/concentrations of gamma rays (20, 30, 40, 50, and 60 kR) and EMS (10, 20, 30, 40 and 50 mM) for inducing mutation. The effect of mutagen was observed on the basis of percentage of seed germination, seedling height reduction at 15th day and survivability. From the result it was observed that, the percentage of seed germination, seedling height reduction at 15th day and survivability were significantly decreased with increasing doses/concentrations of mutagen. The effective doses/ concentrations which caused 50 % growth reduction were 40kR in gamma rays and 30 mM in EMS.
The mutagenic effects of different dose/concentrations of gamma rays (30, 40 and 50 KR) and ethyl methane sulphonate (1.0, 1.5 and 2.0 mM) on sesame (Sesamum indicum L.) varieties VRI-1 were investigated. The characters studied include; days to first flower, plant height, number of branches per plant, number of capsule per plant, number of seeds per capsule and seed yield per plant in M2 and M3 generations. Both negative and positive shifts in mean values were recorded as a result of the physical and chemical treatments. The results indicate the possibilities of evolving higher yield variants through proper selection. Thus, economic traits like number of capsule per plant, number of seeds per capsule and hundred seed weight in M3 generation offer scope for selection and improvement.
