Spożycie cholesterolu z żywnością w diecie krajów zachodnich osiąga wartości nawet 400 mg dziennie, podczas gdy w rekomendacjach zaleca się, aby ilość ta nie przekraczała 300 mg dziennie, a u osób z chorobami układu krążenia 200 mg dziennie. Działanie obniżające poziom cholesterolu we krwi, przez fitosterole znane jest od 50 lat. Zdolność ta związana jest z mechanizmem hamowania jego wchłaniania w przewodzie pokarmowym. Drugi z możliwych mechanizmów związany jest ze zwrotnym transportem cholesterolu z enterocytów do przewodu pokarmowego z wykorzystaniem białek transportujących ABC. Wyżej opisane mechanizmy działania fitosteroli są odmienne od mechanizmu działania statyn, powszechnie stosowanych w terapii hipercholesterolemii. Stwarza to możliwość stosowania fitosteroli i statyn łącznie w obniżaniu zbyt wysokich poziomów cholesterolu we krwi. Dzięki temu osoby już przyjmujące statyny u których nie uzyskano wartości referencyjnych cholesterolu całkowitego i cholesterolu LDL mogą poprzez włącznie do diety fitosteroli uzyskać ich dodatkowe obniżenie.
Organotin compounds are chemicals that are widely used in industry and agriculture as plastic stabilizers, catalysts and biocides. Many of them, including tributyltin (TBT), have been detected in human food and, as a consequence, detectable levels have been found in human blood. As organotin compounds were shown to possess immunotoxic activity, we focused our attention on the effect of TBT on the basic determinants of the function of eosinophils, i.e. cell adhesiveness and motility. We used human eosinophylic leukemia EoL-1 cells, a common in vitro cellular model of human eosinophils. Here, we demonstrate that TBT causes a dose-dependent decrease in the viability of EoL-1 cells. When administered at sub-lethal concentrations, TBT significantly decreases the adhesion of EoL-1 cells to human fibroblasts (HSFs) and inhibits their migration on fibroblast surfaces. Since the basic function of eosinophils is to invade inflamed tissues, our results indicate that TBT, and possibly other organotin compounds, may affect major cellular properties involved in the determination of in vivo eosinophil function.
The present study deals with the estimation of antioxidant levels in blood of sewage and garage workers exposed to sewage water, washing water and vehicular air pollution compared with control group of population. The study areas were selected on the basis of drainage clearance and wards where workers are active as sewage workers. For garage workers, the garage selected was a large and busy one where continuous vehicles washing, repairing etc are carried out. These two exposed groups were compared to non-exposed group of population as control. The blood samples were collected from two groups of exposed population as well as control population and the antioxidant levels were estimated in blood. The present results clearly indicate that there was decreasing trend of the antioxidant level in blood for sewage and garage workers. In the control groups of population the antioxidant levels were found to be within normal range, but for sewage workers (0.14-0.36 mM) and garage workers (0.12-0.36 mM) the antioxidant levels were significantly lower. The range of antioxidant level in human blood is 0.5 to 2.0 mM. It was found to be significantly lower (P < 0.001 or 0.05) in addicted sewage and garage workers when compared with non-addicted groups. All the exposed groups were showed to be having significantly lower antioxidant level when compared to control groups. This study is a preliminary assessment to know the potent biomarkers for oxidative stress and estimation of antioxidant level in blood due to occupational exposure. This study shows a way for easy screening of biomarker assessment but further work is needed in relation to biochemical, enzymological and genetic damage study.
The different members of the secreted aspartyl proteinase (Sap) family of the human pathogenic yeast Candida albicans are proposed to play different roles during infection and are differentially expressed at various body sites. In recent reports, expression analysis has focused on the genes SAP1-6, while the expression pattern of SAP7-10 was less well studied. We analyzed the SAP7-SAP10 expression profile of C. albicans under human serum influence that may be elucidated in the course of blood infection in humans and how this in vitro expression profile is associated with hyphal formation. The phenotypes of strains were examined under scanning electron microscopy. Quantitative RT-PCR (2–ΔΔCT) was used to monitor SAP expression of C. albicans wild type cells and mutants lacking SAP9 and/or SAP10. Of the four analyzed SAP genes, only SAP7 was detectably induced in the double mutant and in the wild type strains in the model that mimics bloodstream infections. On the other hand, in the wild types (isolate 83 and CAF2-1), SAP7 was expressed 0.8- or 0.4-fold less than SAP10, respectively. Our findings suggest that Sap7 may respond to the challenge of the human blood environment. Furthermore, the results support the notion that compensatory upregulation of SAP7 and SAP8 in the Δsap9/Δsap10 mutant occurs in these conditions. SAP7-10 expression was strain-specific. Our findings point to a link between morphogenesis and expression of SAP9 in serum, where these conditions induce both hyphae and SAP9, but temporal gene expression patterns might be controlled by other factors.
Activated blood platelets shed microparticles with procoagulant activity that probably participate in normal hemostasis. We have isolated spontaneously formed microparticles from human blood and analysed them for ultrastructure, antigenic profile, and biochemical composition. In transmission electron microscopy microparticles appeared as regular vesicles with a mean diameter of 300 nm (50-600 nm). In flow cytometry almost all microparticles reacted with fluorescein isothiocyanate (FITC) labeled antibody to platelet glycoprotein complex IIb-IIIa (GpIIb-IIIa) and with FITC-annexin V but only 40-50% of microparticles reacted with FITC-antibody to platelet glycoprotein Ib (GpIb). The latter result was confirmed by double labeling of microparticles with FITC-antibody to GpIIb-IIIa and phycoerythrin (PE) labeled antibody to GpIb. Large microparticles reacted better with anti-GpIb than the small ones. A decreased level of GpIb was also demonstrated by SDS/polyacrylamide gel electrophoresis of microparticles. Compositional studies indicated, that in terms of cholesterol and protein contents, microparticles resembled platelets rather than platelet membranes as previously thought. They are, however, deficient in certain components. Thus, in comparison to platelets, microparticles had reduced contents of sialic acid (by 56.4%), galactosamine (by 48.2%), glucosamine (by 22.4%), galactose by (11.8%) and fucose (by 21.6%). Mannose content was increased by 11.8%. Total phospholipids in microplatelets were lower by 17.8%. Glycerophospholipids only were affected with phosphatidylserine being decreased as much as by 43.2%. Neutral glycosphingolipids, gangliosides and ceramides in microparticles were reduced by half. We conclude that the biochemical composition of microparticles probably reflects previous activation of progenitor platelets
Ochratoxin A (OTA) is a mycotoxin frequently found in human blood and milk samples in the colder climatic zones. In addition to dietary intake, exposure may occur by inhalation of toxin containing fungal conidia. The purpose of this work was to investigate the level of OTA in blood samples from farm workers and non-farm working controls, and to examine if serum levels of OTA were related to inhalatory exposure to conidia of Penicillium verrucosum, the main OTA producer in temperate climates. Blood samples from 210 participants were analysed for the presence of OTA and IgG antibodies against P. verrucosum conidia. The concentration of OTA was determined by HPLC (DL 10 ng/l), and the IgG level was determined by ELISA. All serum samples contained OTA (mean 397 ng/l, range 21-5534 ng/l). The OTA level in serum was unrelated to farm working, gender, age, and IgG level. The mean IgG level was significantly higher among farm workers than controls. Farm working, or increased inhalatory exposure to P. verrucosum, was not related to higher OTA serum levels. Inhalatory exposure to OTA from farm working seems to be of minor importance compared to dietary intake.
