The paper presents an attempt to assess the reaction of seeds to mechanical loads taking into account their geometry expressed as seed thickness and 1000 seed weight. The initial material comprised 33 genotypes of grain legume plants and included cultivars registered in the country and breeding lines that are subject to pre-registration trials. The analysis of variance revealed significant diversity of the cultivars and lines of the species studied in terms of each of the analysed trait. The highest weight of 1000 seeds were obtained for white lupine seeds and peas, the lowest for andean lupine seeds. The maximum deformation and energy were obtained for white lupine seeds, the lowest for pea seeds, the maximum force and module the lowest values were determined for narrow-leafed lupine and pea. The highest values of protein were obtained for andean and yellow lupine, a fat content for andean and white lupine. The fatty acid profile as much as 70% or more were linoleic and oleic acids. Against the background of all the species are distinguished by white lupine seeds with a high content of oleic acid and the lowest of linoleic acid, for yellow lupine were obtained the inverse ratio of the two acids.
The nitrogen fixation response to copper nutrition in faba bean, yellow lupin and soybean was studied. Copper nutrition significantly increased the pod yields of all tested grain legumes but faba bean gave the greatest Cu-use efficiency for pod and grain production. The accumulation of dry matter in vegetative parts, nodules, N and leghemoglobin concentration in nodules and nitrogen accumulation in the whole plants were increased by copper supply in faba bean and yellow lupin in contrast with soybean. Cu nutrition significantly increased the Cu concentrations in nodules of all cultivated plants. The differential sensitivity of N₂ fixation in tested grain legume species to copper nutrition could be connected with the level of phenols in nodules and depended on both the host plants and strains of rhizobia, which differ in their ability to produce catechol-like siderophores. Copper requirements by symbiotic N₂ fixation could also depend on the nature of phenols in nodules (presence of o-dihydroxyphenols or number of hydroxyls in molecule).
Two plasmid vectors were introduced into soybean (Glycine max (L.) Merr.) and azuki bean (Vigna angularis Willd. Ohwi & Ohashi) using different transformation systems. Azuki bean epicotyl explants were prepared from etiolated seedlings and co-cultivated with Agrobacterium tumefaciens for 2 days. Adventitious shoots were developed from the callus of the explants on a regeneration medium containing hygromycin, and the shoots were excised and transferred to a rooting medium containing hygromycin at the same concentration. Rooting shoots were transferred to soil and grown in a glass-house to produce viable seeds. PCR analysis confirmed clearly the presence of the hpt gene in most of the azuki beans regenerated under hygromycin selection. A soybean embryogenic suspension culture was generated from immature cotyledons, and used for the introduction of plasmids by particle bombardment. Hygromycin-resistant embryogenic clones were isolated after 8 weeks of hygromycin selection, and then the green clones were matured on the differentiation medium. After desiccation, the embryos were germinated on the rooting medium, and the plants were transferred to soil in a glass-house. More than 50% of the regenerated soybean plants tolerant to hygromycin yielded the hpt fragment on PCR analysis. The azuki bean transformants were obtained more rapidly and with higher efficiency than the soybean transformants.
The effect of processing conditions (soaking in water for 0-3 h, cooking in water or 1-3% salt solution for 15 or 30 min) was tested. The water absorption was affected both by soaking and cooking, but the salt content had little effect. The descriptors connected with visual testing and those connected with texture were influenced more by cooking, than by soaking; relationships between different characteristics were either linear or logarithmic or hyperbolic. They were related to the water content in cooked lentils, which had, however, no significant effect on the flavour acceptance. It rose with the intensity of salty flavour till a flat optimum at medium saltiness of 2% salt in the cooking liquid.
The results of plant breeding trials with populations of fodder pea strains and broad bean hybrids were the basis of consideration on the interrelationship between some traits - the yield structure elements. Developed by Eaton, a relatively new method of yield component analysis called the two-dimensional partitioning method (TDP) was applied to analyse the data. The method, which combines multiple regression and ANOVA, allows for concise tabular presentation and simple interpretation of the distribution of traits in one direction and the sources of variance according to ANOVA model in the other direction. Additionally, the interpretation of the results was supported by such standard statistical techniques as ANOVA, simple and multiple regression and path analysis. The main components of pea yielding were plant height and the number of pods per plant. Among the analysed characters of broad bean the number of nodes with pods on the main stem, which turned out to be the determinant of broad bean yielding, might be strongly affected by environmental conditions. The number of nodes with pods might be considered a selecting character of high potential yielding of broad bean genotypes.
