The data concerning endosperm type in Chondrilla juncea L. from Europe, Asia and Australia have been the subject of controversy. This study investigated the problem embryologically in an individual plant growing on an experimental field. Two endosperm types, nuclear and cellular, were observed to be present in inflorescences of the same plant. Both types of endosperm were formed after open pollination as well as after emasculation; however, cellular endosperm predominated slightly after open pollination. The development of endosperm preceded embryo development independently of endosperm type. Intraspecific variability of endosperm type in the investigated plant seems to be genetically conditioned, and it may be modified by external conditions.
Callus induction, organogenesis, and autonomous endosperm were induced by in vitro culture of unfertilized ovules of six genotypes of Viola odorata L. (2n = 20). On media supplemented with auxins and cytokinins the ovules enlarged; the frequency of enlarged ovules depended on plant genotype, the stage of ovule development and the hormones used. The highest frequency of enlarged ovules was on MS medium with 2,4-D and kinetin. Autonomous endosperm was observed in ovules after 6 days of culture on MS medium with 0.1 mg l-1 2,4-D. No egg cell divisions were found in examined ovules. The highest rate of callus induction was on MS medium with 2,4-D and BAP. Roots regenerated from callus on the same medium and on media with different concentrations of auxin and cytokinin. Karyological variability of callus cells and regenerated roots was observed, with euploid and aneuploid chromosome numbers in roots (2n = 13, 15, 17, 18, 20) and diploid and polyploid in the callus (2n = 20, 30, 40). The origin of callus is discussed.
Autonomous endosperm (AE) was found in 39 (17.3%) of in vitro cultured unpollinated ovaries of Brassica napus L. cv. Topas, with average frequency 1.5 ovule with AE per ovary. AE was induced on MS medium and on MS + 2 mg/l BAP + 0.1 mg/l NAA , MS + 2 mg/l BAP + 2 mg/l 2,4-D, and in ovules exposed to 1 h auxin treatment (MS + 20 or 40 mg/l 2,4-D) and then transferred to hormone-free MS medium. The highest frequency of AE induction occurred after 1 h auxin treatment (34.5%). The structure and size of AE nuclei suggest their origin from secondary nucleus or polar nucleus/nuclei. The control experiment with unpollinated, bagged flower buds left on plants, revealed no AE induction in situ. Gynogenic embryos were not observed in the material studied.
