The aim of the study was to investigate the effect of 10-day treatment (i.p.) with Padma 28 on the levels of a-fetoprotein and ß2-microglobulin and the activities of aminotransferases ALT and AST in the serum of cadmium-poisoned rats (dose of 0.36 mg/kg i.p.). It was found that acute intoxication with cadmium chloride significantly enhanced AFP and ß2-microglobulin levels and did not affect the activity of ALT and AST in the serum of rats in comparison with the control group. Padma 28 administrated for 10-days only in the dose of 1 mg/kg significantly decreased AFP and in the doses of 0.1 and 1 mg/kg increased ß2-microglobulin levels while it did not affect the activity of ALT and AST in the serum of cadmium-poisoned rats. 10-day treatment with Padma 28 has been found to increase nephrotoxic effects of cadmium in rats acutely intoxicated with this metal.
The aim of this paper was to examine the activity of lysosomal enzymes after intoxication with chlorfenvinvos and dichlorvos - phosphoorganic insecticides. Investigations were carried out on male Wistar rats weighing 180-230 g. Rats were divided into two groups: control and treated. Rats of treated group were receiving oil solution of chlorfenvinvos at doses of 0.5 LD50 or 0.1 LD5(), determinations were performed after 1 and 24 hours after treatment or dichlorvos in the dose equal to 0.5 LD50 estimations were performed after 1 h only. Rats of control group were given equivalent volume of oil. Activities of beta-glucuronidase and acid phosphatase in the blood serum and lysosomal fraction of rat liver homogenate were determined. It was found that the level of ChE inhibition and increase of BGR activity in the serum were dependant on chemical structure of used compound, dose and time after treatment. It was observed that increase of hydrolases activities in serum was accompanied by the decrease in its activity in the lysosomal fraction of liver homogenate.
The influence of acute carbon tetrachloride intoxication on hepatic and placental cytochrome P-450 content, and on NADPH-cytochrome P-450 reductase was examined in Wistar female rats. The animals were pretreated with a single dose of CC14 before pregnancy. The contents of the constituents of the MFO system were measured at term. The results revealed that the CCl4-mediated decrease at the total hepatic cytochrome P-450 level was reduced by pregnancy. Carbon tetrachloride pretreatment did not change the placental cytochrome P-450 level. A significant increase in placental microsomal cytochrome P-450 reductase activity was found after acute CC14 intoxication, which did not influence this enzyme in the liver of pregnant individuals.
