The clinical effects of twenty different chromosome anomalies in sheep are reviewed. It is concluded that not all anomalies give rise to reproductive problems or phenotypic variations. In particular, the centric fusion translocations appear not to have affected fertility and they have been helpful in determining the karyotype of the sheep.
Adenosine 5'-phosphoramidate (NH2-pA) is a rare natural nucleotide and its biochemistry and biological functions are poorly recognized. All organisms have proteins that may be involved in the catabolism of NH2-pA. They are members of the HIT protein family and catalyze hydrolytic splitting of NH2-pA to 5'-AMP and ammonia. At least five HIT proteins have been identified in mammals; however, the enzymatic and molecular properties of only Fhit and Hint1 have been comprehensively studied. Our study focuses on the Hint2 protein purified by a simple procedure to homogeneity from sheep liver mitochondrial fraction (OaHint2). Hint1 protein was also prepared from sheep liver (OaHint1) and the molecular and kinetic properties of the two proteins compared. Both function as homodimers and behave as nucleoside 5'-phosphoramidate hydrolases. The molecular mass of the OaHint2 monomer is 16 kDa and that of the OaHint1 monomer 14.9 kDa. Among potential substrates studied, NH2-pA appeared to be the best; the Km and kcat values estimated for this compound are 6.6 µM and 68.3 s-1, and 1.5 µM and 11.0 s-1 per natively functioning dimer of OaHint2 and OaHint1, respectively. Studies of the rates of hydrolysis of different NH2-pA derivatives show that Hint2 is more specific towards compounds with a P-N bond than Hint1. The thermostability of these two proteins is also compared.
Quantitative and qualitative studies dealing with histomorphometry of the bone tissue play a new role in modern legal medicine/forensic medicine and archaeozoology nowadays. This study deals with the differences found in case of humerus and metapodial bones of recent sheep (Ovis aries), goat (Capra hircus) and roe deer (Capreolus capreolus) specimens, both from a qualitative point of view, but mainly from a quantitative perspective. A novel perspective given by the fractal analysis performed on the digital histological images is approached. This study shows that the qualitative assessment may not be a reliable one due to the close resemblance of the structures. From the quantitative perspective (several measurements performed on osteonal units and statistical processing of data), some of the elements measured show significant differences among 3 species (the primary osteonal diameter, etc.). The fractal analysis and the lacunarity of the images show a great deal of potential, proving that this type of analysis can be of great help in the separation of the material from this perspective. The large stomach worm, Haemonchus contortus is a highly pathogenic nematode parasite of sheep (Ovis aries) and goats (Capra hircus). It sucks blood and causes acute disease, anaemia and severe economic losses due to morbidity and mortality. This paper compares characteristics of surface cuticular ridges (synlophe) and morphometrics of total body length, cervical papillae, spicules, barbs, gubernaculum and vulvar flap morphology of H. contortus recovered in the two hosts from the States of Perak and Kelantan, Peninsular Malaysia. The morphological variation in H. contortus samples recovered from the two hosts in Kelantan was found to be higher than from Perak. A discriminant function, based on three measurements of the spicule, confirmed the identity of male worms in the two hosts as belonging to the same species H. contortus.
The mRNA differential display technique was applied to investigate the differences in gene expression in the muscle tissue between black-boned and ordinary (“normal”) sheep. The gene that was differentially expressed was identified through semi-quantitative RT-PCR, and its complete cDNA sequence was then obtained using the rapid amplification of cDNA ends (RACE) method.The nucleotide sequence of the gene was not found homologous with any sheep gene described so far. Sequence prediction analysis revealed that the open reading frame of the gene encodes a protein of 763 amino acids, highly homologous with the hydroxyacyl-coenzyme A dehydrogenase/3-ketoacyl-coenzyme A (HADHA) of bovine (98%), pig (90%), sumatran orangutan (87%), human (87%), chimpanzee (87%), mouse (83%) and rat (82%) – so that it can be defined as sheep HADHA gene. This novel sheep gene was finally assigned to GeneID: 100192316. Phylogenetic tree analysis revealed that the sheep HADHA gene is genetically closely related to the HADHA gene of cattle. Tissue expression analysis indicated that the sheep HADHA is also differentially expressed In different tissues of black-boned sheep. The present study established the primary foundation for further research on the HADHA gene in sheep.
The effects of vegetation fragmentation and mixed grazing (ie mono- or multi-species animal group) on the diet composition of sheep and red deer grazing mosaics of grassland and heather moorland was studied, using faecal cuticle analysis, in two experimental sites in Scotland during the summer of 1992 and 1993. On Site A, the influence of grassland fragmentation on diet composition was estimated for sheep and deer grazing together in plots where the grassland (20% of the area) was artificially distributed as one large, four medium or twelve small patches within a homogeneous moorland matrix (80% of the area). On Site B, differences in diet composition between animals grazing within mono-species (sheep or deer) and multi-species groups (sheep and deer together) were examined for each animal species. In this site all plots used contained a similar natural mosaic pattern of grass and heather (ie similar mixtures of patch sizes, with about 20% grass and 80% heather cover). On Site A, the proportions of grass in the diet of sheep (73%) and deer (27%) were found to be similar across all levels of grass fragmentation. A significant interaction was found between the pattern of fragmentation and the three periods in which the experiment was carried out. On Site B in 1992, sheep had more grass in their diet than did deer (52% vs 46%), and the diets of both sheep and deer responded in the same fashion when the species were grazing in mono- or multi-species groups, The consumption of grass decreased in both species throughout the period studied. Deer showed no change in the proportion of grass in their diet in the presence or absence of sheep in 1992 (deer 48% vs sheep 50%). But on Site B in 1993, the diet of sheep contained a significantly higher proportion of grasses when they were grazing with red deer (52%) than when they were grazing alone (38%). These results suggest that on grassland/heather moorland mosaics sheep may suffer intraspecies competition to a greater extent than do red deer, particularly where grass is in relatively low supply,
The abundance, density, and habitat use of roe deerCapreolus capreolus (Linnaeus, 1758) and mouflonOvis aries Linnaeus, 1758 were studied in a confined Mediterranean area in Greece with a dung survey based on the faecal accumulation rate (FAR) technique. Estimated density was modelled with generalized additive models using altitude, habitat type, and slope as potential covariates. Model selection among the set of candidate models was conducted based on their generalized cross-validation score. Roe deer had an estimated mean density of 13.9 ind./km2 and the best model included slope and habitat type as covariates. The mean density of mouflon in the study area was 22.1 ind./km2 and the best model used altitude and habitat type as covariates. For both species, the highest densities were encountered in abandoned cultivations and glades, followed by conifer forests, while the lowest densities were observed in maquis. However, use of open habitats by mouflon was much greater than it was for roe deer. The strong preference of mouflon (a grazer species) for open habitats that were abundant with grasses probably reflected food availability and contrasted with the more diverse habitat use by roe deer (a selective browser).
The feeding strategies of the Spanish ibex Capra pyrenaica Schinz, 1838 and sheep Ovis aries Linnaeus, 1758 during summer in the alpine area of Sierra Nevada in south-eastern Spain were compared. The degree of vegetation use by the two ungulates, food availability, diet composition, species selection indices and the effect of plant availability and chemical composition on diet selection were analysed. The available biomass of herbaceous and woody resources was 87 and 13%, respectively. The degree of use was 15% for herbaceous vegetation and 10.3% for woody vegetation. The most used species were those with moderate amounts in diet composition of both ruminants. The main plant groups consumed by Spanish ibex were graminoids (57.5%), shrubs (20.5%) and chamaephytes (14.3%). Sheep consumed graminoids (41.6%), forbs (32.3%) and chamaephytes (20.9%). There was a significant correlation between the two diets, with an overlap of 56.3%. The highest indices of selection were found in items with moderate-low availability and which comprised moderate levels in the diet composition of the two species. Spanish ibex mainly selected diet according to availability, while sheep selection depended on protein content. The ibex consumed cell wall-rich resources such as grasses and shrubs, while the sheep consumed higher quality feed and includes more forbs in its diet.
