This work tackles the problem of misconception when using sophisticated mathematical tools, nonlinear optimization in this particular case, to solve a navigational problem. Namely, to reach the Great Circle vertex with two rhumb line legs ensuing the optimized distance, an initial rhumb line course equal to the orthodromic course at middle latitude may be used. The initial course is thereupon optimized by the incremental value steps. The optimized distance is achieved if the rhumb line course is altered towards the vertex at the orthodrome-loxodrome intersection point. As determination of this point cannot be formulated in a closed form, an iterative solution is to be applied. The derived transcendental equation forms a basis for an iterative solution of intersection using the Newton-Raphson method. To the contrary, finding solutions to a system of nonlinear equations can mislead a researcher unable to comprehend and grasp the mathematical meanings of the algorithm. The aim of this quantitative research is to determine the perception and actual knowledge of secondary school students in the Belgrade region with respect to earthquakes as a natural disaster and security threat, and to identify the factors that influence their knowledge and perceptions. The authors used a method of surveying students to identify and describe the factors that influence student knowledge and perceptions about earthquakes. For the purpose of this research, a sample of 3,063 students was drawn from the total population of secondary school students in Belgrade (65,561 students), which equates to 4.67% of the population. The results show that the sources of information on natural disasters and their threatening consequences influence the perceptions of secondary school students. In view of the evident lack of education about natural disasters in Serbia, the results of this study can be used when creating a strategy for educational programs. This research is the first step in developing and realizing a future strategy for natural disaster management by informing and including public (school population), scientific, and administrative communities in the process.
Expression of the rat α2-macroglobulin (MG) gene undergoes dynamic changes throughout an individual's life and during the acute-phase (AP) response. Details of the participation of the STAT family of transcription factors in its control remain incompletely understood. Here we examined the involvement of STAT5b in MG gene expression during development and the AP response. Immuno-blot analysis revealed the highest nuclear level of STAT5b in the fetus and during postnatal development, whereas in the adult it decreased. Stimulation of MG expression during the AP response was accompanied by a decrease in STAT5b. Examination of STAT5b localization revealed that the relative concentrations of STAT5b were higher in the nuclear matrix than in the nuclear extract. Affinity chromatography with the extended promoter region of the MG gene (-825/+12), followed by immuno-blot analysis, revealed dynamic changes in STAT5b binding. The highest concentration of the promoter-binding form of STAT5b was observed in the fetus. As postnatal development progressed, the level of promoter-bound STAT5b decreased and in the adult liver it was the lowest. Stimulation of MG gene expression during the AP response in both the fetus and adult was accompanied by significantly decreased STAT5b binding to the MG promoter. The AP response was accompanied by lower levels of STAT5b serine and tyrosine phosphorylation in both fetus and adult. In the nuclear matrix derived from adult tissues, tyrosine phosphorylated species were completely absent. We conclude that developmental-stage differences in the mechanisms that determine STAT5b nuclear localization contribute to its activity in vivo.
