In this study we have developed a modified CLARK and DIAMOND (1992, 1993) method using the polymerase chain reaction to amplify amoebic ribosomal RNA genes that allow either specific detection of Entamoeba histolytica s. str. or amoeba species identification. DNA was isolated from cultured protozoa or from stool samples (cysts andlor trophozoites). Cultures of xenie or axenic material (also stool samples) were treated with Easy Genomic DNA Prep or Genomic DNA Prep Plus (A&A Biotechnology, Poland) for DNA isolation. With these new procedures, DNA can be obtained effectively and with high degree of purity. 13 amoeba strains were investigated. Three strains produced an 876 bp fragment with Psp5 and Psp3 primers (specific product for E. histolytica s. str). Three strains gave a specific 876 bp product for E. dispar with NPsp5 and NPsp3 primers. Two strains were E. moshkovskii as identified by KpnI digestion of PCR products obtained with RD5 and RD3 primers. Four strains (one of them was cultured in two laboratories) were E. invadens as identified by HinfI or HhaI digestion of PCR products obtained with RD5 and RD3 primers. Characteristic RFLP patterns with these enzymes was also obtained for E. terrapinae and Blastocystis hominis. This RFLP analysis show significant promise as a medical diagnostic tool particularly useful for species identification.
The present investigation covered 400 specimens of Platichthys flesus and 22 of Scophthalmus maximus caught in the period of October 1993-December 1994 in the Gulf of Gdańsk (the South-east Baltic). The following parasite species were found: Bothriocephalus scorpii, Hysterothylacium auctum, Cucullanus heterochrous, Cuculanellus minutus, Anisakis simplex, Raphidascaris sp. (probably R. acus), Echinorhynchus gadi, and Pomphorhynchus laevis. The occurrence of parasites was studied in relation to the season of fishing and the length of the fish body.
The authors deal with the problem of malaria induced by Plasmodium falciparum and imported to Poland by people returning from tropics. They stress significance of the variable clinical pattern involved and of chloroquine resistance. Basing on their own observations of a definitely diagnosed malaria (22 out 35 cases observed and suspected of malaria) the authors discuss diagnostic and therapeutic difficulties encountered in 7 patients with Plasmodium falciparum-induced malaria. Two representative cases of malaria (Plasmodium falciparum) have been discussed in detail. One of the cases with malaria imported from tropics, presented severe course and an atypical clinical pattern (with involvement of central nervous system) which made appropriate diagnosis difficult and delayed application of specific causal treatment. The other unusual case involved a nurse who contracted Plasmodium through a small skin wound with which patient's blood came into contact when the nurse was drawing blood for testing. Course of the disease was grave, with deep anaemia and central nervous system and kidney involvement. In both cases the disease had favourable outcome due to complex anti-malarial therapy and multispecific medical intervention.
The nematodes were collected from livers of Baltic cod caught off the Gulf of Gdańsk. This paper consists of a description of the third stage larvae of Contracaecum osculatum (Rud.). This nematode is also pathogenic for man and its occurrence on the Polish coast has epidemiological significance.
