The p21-activated kinases (PAKs) are serine/threonine protein kinases interacting with small GTPases - Rac and Cdc42. PAKs are found in most eukaryotes and play an evolutionarily conserved role in many cellular processes. Six human PAKs have been identified, and based on homology, they can be classified into two groups. This review focuses specifically on the role of Rac/Cdc42 regulated PAKs in maintaining and remodeling cytoskeletal structure in various organisms. A list of PAKs substrates and binding partners implicated directly and indirectly in cytoskeletal reorganization is presented. Also perturbations of the Rac/Cdc42/PAK pathway leading to tumorigenesis and neurodegenerative diseases are reviewed.
Ceramides, bioactive members of the sphingolipids can be generated by de novo synthesis, sphingomyelin hydrolysis and by acylation of sphingosine. Ceramides are known to regulate several cellular processes, including differentiation, growth suppression, cell senescence and apoptosis. The ceramide levels increased in several pathological conditions such as brain ischemia, hypoglycemia, inflammation and in neurodegenerative disorders. Sphingosine, a metabolite of ceramide is phosphorylated by sphingosine kinases (Sphk type 1and 2) to sphingosine-1-phosphate (S1P). Sphingosine kinases are critical regulators of the sphingolipid biostat. The aim of this study was to investigate the role of ceramide and S1P in molecular mechanisms of neuronal cells death. The human neuroblastoma cell line (SH-SY5Y) was exposed to cell-permeable C2-ceramide. Ceramide decreased the viability of SH-SY5Y cells in concentration dependent manner. The intracellular free radical generation after ceramide treatment was about 3-fold higher comparing to control. Concomitantly our study indicated that ceramide induced poly(ADP-ribose) polymerase-1 (PARP-1) activation and decreased the level of apoptosis inducing factor (AIF) in mitochondria. Ceramide diminished the expression and level of anti-apoptotic Bcl-2 protein. PARP-1 inhibitor enhanced the level of Bcl-2 protein and cells survival keeping the level of AIF in mitochondria unchanged. The recent studies indicated that ERK1/2 are involved directly in regulation of PARP-1 activity. The specific inhibitor of these kinases protected cells against death evoked by ceramide in our experimental conditions. Moreover, our study indicated, that sphingosine-1-phosphate (S1P) increased Bcl-2 gene expression and SH-SY5Y cells survival after ceramide treatment. Summarizing, our data present that PARP-1 inhibitor and sphingosine-1-phosphate (S1P) through modulation of anti-apoptotic proteins protect mitochondria and neuronal cells against death evoked by ceramide. microRNAs regulate all the cellular processes, and are strongly involved in differentiation of stem cells. Disturbances in the regulation of microRNAs expression and activity may deviate the stem cells fate, impairing their differentiation and contributing to diseases initiation or progression. In this talk the role of microRNAs in certain neurological conditions will be discussed.
The nucleophosmin 1 gene (NPM1) encodes a multifunctional nucleolar phosphoprotein that plays a crucial role in the control of various aspects of cell growth and homeostasis. In this study, the coding region of the NPM1 gene was screened in 1035 individuals of 4 Chinese cattle breeds by DNA sequencing and Polyacrylamide gel electrophoresis. A novel 12-bp deletion mutation was identified in the coding region of the NPM1 gene. The PCR products of primer NPM1-P2 exhibited 3 genotypes and 2 alleles: 178 bp (denoted as W) and 166 bp (denoted as D). Genotype DD and allele D were predominant in the studied populations. Association analysis with growth traits in the Nanyang breed (N = 265) showed that the animals with genotype DD had significantly greater birth weight, body weight, body length, and heart girth than those with genotype WD (P <0.01 or P < 0.05) at birth and after 6 months and 12 months, but not at 18 and 24 months of age. Results of this study suggest that the NPM1 gene is a candidate gene for growth traits in cattle.
The mitogen-activated protein kinase (MAPK) cascades regulate important cellular processes, including growth, differentiation, apoptosis, embryogenesis, motility and gene expression. Although MAPKs mostly appear to be constitutively expressed, the transcript levels of some MAPK-encoding genes increase upon treatment with specific stimuli. This applies to the MAPKactivated protein kinases MK2 and MK3. By contrast, the transcriptional regulation of the related MK5 has not yet been studied. The MK5 promoters of mouse, rat and human contain a plethora of putative transcription factor sites, and the spatio-temporal expression of MK5 suggests inducible transcription of the gene. We examined the transcription pattern of MK5 in different tissues, and studied the kinetics of MK5 expression at the transcriptional and/or translation level in PC12 cells exposed to arsenite, forskolin, KCl, lipopolysaccharide, spermine NONOate, retinoic acid, serum, phorbol ester, temperature shock, and vanadate. Cells exposed to forskolin display a transient increase in MK5 mRNA, despite their unaltered MK5 protein levels. The MK5 promoters of human, mouse and rat contain a cAMP-responsive element that binds the cAMPresponsive element-binding protein (CREB) in vitro. Luciferase reporter constructs containing an 850-base pair human MK5 promoter fragment encompassing the CRE showed a basal activity that was 10-fold higher than the corresponding construct in which the CRE motif was deleted. siRNA-mediated depletion of CREB had no effect on the endogenous MK5 protein levels. Several binding motifs for heat shock factor are dispersed in the mouse and rat promoter, and temperature shock transiently enhanced the MK5 transcript levels. None of the other tested stimuli had an effect on the MK5 mRNA or protein levels. Our results indicate an inducible regulation of MK5 transcription in response to specific stimuli. However, the MK5 protein levels remained unaffected by all the stimuli tested. There is still no explanation for the discrepancy between the increased mRNA and unchanged MK5 protein levels.

Magnesium: its role in nutrition and carcinogenesis

58%
Magnesium (Mg2+) plays a key role in many essential cellular processes such as intermediary metabolism, DNA replication and repair, transporting potassium and calcium ions, cell proliferation together with signalling transduction. Dietary sources rich in magnesium are whole and unrefined grains, seeds, cocoa, nuts, almonds and green leafy vegetables. Hard water is also considered to be an important source of magnesium beneficial to human health. The daily dietary intake of magnesium is however frequently found to be below that recommended in Western countries. Indeed, it is recognised that magnesium deficiency may lead to many disorders of the human body, where for instance magnesium depletion is believed to play an important role in the aetiology of the following; cardiovascular disease (including thrombosis, atherosclerosis, ishaemic heart disease, myocardial infarction, hypertension, arrhythmias and congestive heart failure in human), as well as diabetes mellitus, gastrointestinal (GI) tract disease, liver cirrhosis and diseases of the thyroid and parathyroid glands. Insufficient dietary intake of magnesium may also significantly affect the development and exacerbation of ADHD (Attention Deficit- Hyperactivity Disorder) symptoms in children. The known links between magnesium and carcinogenesis still remain unclear and complex, with conflicting results being reported from many experimental, epidemiological and clinical studies; further knowledge is thus required. Mg2+ ions are enzyme cofactors involved in DNA repair mechanisms that maintain genomic stability and fidelity. Any magnesium deficiencies could thereby cause a dysfunction of these systems to occur leading to DNA mutations. Magnesium deficiency may also be associated with inflammation and increased levels of free radicals where both inflammatory mediators and free radicals so arising could cause oxidative DNA damage and therefore tumour formation. The presented review article now provides a summary discussion of the various research performed concerning the impact that low magnesium intake has on tumour incidence; this includes impairment of magnesium homeostasis frequently observed in tumour cells, the influence of magnesium depletion on the progression of existing tumours and the occurrence of hypo-magnesaemia when patients are treated with certain anticancer drugs.
ZBTB7A is a known proto-oncogene that is implicated in carcinogenesis and cell differentiation and development. Fully understanding the function of ZBTB7A in cellular processes could provide useful strategies for cancer treatment and development-associated disease therapy. Here, global mapping of ZBTB7A transcription factor binding sites was developed by utilizing microarray technology in HepG2 cells. The data obtained from the microarrays was further validated via chromatin immunoprecipitation-PCR (ChIP-PCR) and real time-PCR, and it was revealed that ZBTB7A may be one of the regulators of neural development. ZBTB7A target signal pathways were identified in signal pathway and GO (Gene Ontology) analyses. This is the first report on the global mapping of ZBTB7A downstream direct targets, and these findings will be useful in understanding the roles of ZBTB7A in cellular processes.
Proteasome is a multi-activity enzyme involved in a ubiquitin-dependent turnover of cytoplasmic and nuclear proteins. It recognizes and digests short-lived regulatory proteins, influencing cellular processes as crucial as progression of the cell cycle, transcription, oncogenesis and flux of substrates through metabolic pathways. The enzyme is responsible also for the housekeeping chores, degrading misfolded or oxidatively damaged proteins. Defects in the proteasome action play a causal role in development of a number of diseases, among which are cerebral ischemia and neurodegenerative disorders such as Huntington’s, Alzheimer’s, and Parkinson’s diseases. Being a multifunctional proteolytic machinery, the proteasome must act under a strict control to prevent massive degradation of all intracellular proteins, which would result in a cell death. One of the levels of such a control is the proteasome structure itself. The core particle called 20S proteasome is a barrel-like structure made up of four rings of seven subunits each. The outer (α) rings play predominantly a structural role forming a kind of a gated channel leading to the proteolytic chamber. The inner-β-rings harbor six active sites, concealed inside the cavity formed by the β subunits. So far, the only proteasome-targeting agents used in clinics are competitive inhibitors, directly blocking the enzyme’s active sites. However, the multi-subunit barrel-like structure of the 20S proteasome encourages to test compounds which can target allosteric interactions between subunits and influence the gating mechanism, involved in the control of the substrates’ uptake. Such modulators may provide a precise and substrate-specific regulation of the proteasome catalytic performance. Additionally, targeting the allosteric interactions may enable not only inhibition but also stimulation of the proteasome, which is crucial in managing disorders connected with the proteasome not sufficient activity, such as neurodegenerative diseases. A variety of protein ligands, interacting with the outer ring of the 20S proteasome and modulating its activity, is already known. They can serve as templates for design of putative small-molecule allosteric drugs. In an effort to find synthetic compounds able to enhance or suppress the performance of the proteasome active centers we utilize one of such protein ligands – HIV-1 Tat protein. The protein is known to inhibit the core proteasome and to interfere with the physiological PA28 activator in its binding to the 20S. G48RKKRRQRRRPS59 fragment of HIV-1 Tat (Tat1) occurred to be very efficient in the 20S proteasome inhibition. By single and multiple alanine substitutions we have recognized “hot spots” in the sequence of Tat1. NMR and molecular dynamics calculations allowed us to correlate these putative pharmacophores with the structural turns. By introduction of a non-peptide turn-inducing modification to the Tat1 sequence we have obtained the derivatives highly toxic for human cultured cancer cells HeLa.S3. Rsp5 is a conserved ubiquitin ligase involved in regulation of numerous cellular processes. A growing number of publications describing new functions of the ligase have appeared in recent years. Rsp5 was shown to be involved in the control of intracellular trafficking of proteins via endocytosis and multivesicular body sorting. Moreover, nuclear functions of Rsp5 in response to various stresses have been discovered. Rsp5 is also involved in the regulation of unsaturated fatty acid and sterol synthesis and phospholipid composition. Here, an overview of Rsp5 functions with emphasis on its involvement in the regulation of lipid biosynthesis will be presented.
