Background. Breast cancer is the most common malignant tumour in women in Poland. To reduce the risk of breast cancer, appropriate prevention is necessary. Numerous studies conducted over many years in Poland and throughout the world have demonstrated a significant effect of selenium on prevention of disease, including breast cancer in women. The following paper aims to present the literature on effects of selenium (Se) on prevention of breast cancer in women. Material and methods. Based on national and international literature, the paper presents information on the role of selenium (Se) in prevention of breast cancer in women. Results. Numerous studies conducted in research centres in Poland and abroad have shown that female patients with breast cancer and individuals with gastrointestinal cancer have significantly lower selenium concentration in their blood serum and whole blood, as well as significantly lower GSH - Px activity in plasma and red blood cells, compared to healthy women. Low selenium concentration may indicate an increased risk of breast cancer. Selenium is an essential co-factor in the production of antioxidant enzymes and may affect the development of cancer. Clinical trials which assessed selenium content in food showed that its supplementation reduced cancer mortality. Conclusions. Results of numerous national and prospective international studies indicate that low selenium intake and/or concentration in serum/plasma/nails is a high-risk marker of the majority of cancers, including breast cancer in women.
Edible mushrooms (Agaricus bisporus) were enriched with an irrigation solution containing or not fortified with selenium in form of sodium selenite. Se in control mushrooms was 1.04 mcg/g D.M. and in mushrooms subjected irrigations with water solution of Se this increased to 15 mg of Se/dm) after the first flush and rose to with subsequent flushes to 30.14 mcg/g D.M. Peroxidase activity offresh mushrooms was reduced in Se-enriched Agaricus bisporus. Postharvest storage Peroxidase activity increase for 6 days and then a depression in activity .occutred. A positive correlation between peroxidase activity and fruit body mass of mushrooms was observed. Following fractionation on Sephadex G-100 column 4 peaks of peroxidases activity was observed in water extracts of mushrooms regardless of Se treatment. Molecular mass of these protein fractions were estimated by molecular mass markers as 82, 57, 45 and 22 kDa. Substantial part of peroxidase activity appears to originated from Se-dependent glutathione peroxidase.
The experiment involved Wistar rats given orally trace daily doses of selenium-75 for four weeks. The distribution of selenium-75 was evaluated in the carcass and selected organs within 28 days after the last administration. The results were compared with those reported in the literature; the kinetic data provided evidence that the absorption and distribution of selenium depend on several factors involving dosage and the route and period of administration.
The objective of this study was to investigate the effect of Selenium (Se) supply (0, 3, 6, 12, 24 mg kg−1) on the growth, photosynthetic characteristics, Se accumulation and distribution of flue-cured tobacco (Nicotiana tabacum L.). Results showed that low-dose Se treatments (≤6 mg kg−1) stimulated plant growth but high-dose Se treatments (≥12 mg kg−1) hindered plant growth. Optimal Se dose (6 mg kg−1) stimulated plant growth by reducing MDA content and improving photosynthetic capability. However, excess Se (24 mg kg−1) increased MDA content by 28%, decreased net photosynthetic rate and carboxylation efficiency by 34% and 39%, respectively. The Se concentration in the roots, stems, and leaves of the tobacco plants significantly increased with increasing Se application. A linear correlation (R = 0.95, P < 0.01) was observed between Se level and tobacco plant tissue Se concentration. This correlation indicated that the tobacco plant tissues were not saturated within the concentration range tested. The pattern of total Se concentration in the tobacco plant tissues followed the order root > leaf > stem. The Se concentration in the roots was 3.17 and 7.57 times higher than that in the leaves and stems, respectively, after treatment with 24 mg kg−1 Se. In conclusion, the present study suggested that optimal Se dose (6 mg kg−1) improved the plant growth mainly by enhancing photosynthesis, stomatal conductance, carboxylation efficiency and Rubisco content in the flue-cured tobacco leaves. However, the inhibition of excess Se on tobacco growth might be due to high accumulation of Se in roots and the damage of photosynthesis in leaves.
Background. Selenium is an element of very great importance for the proper functioning of the human body, mainly due to its antioxidant properties. Selenium exhibits a preventive effect in the case of cardiovascular disease, the immune system, male infertility and inhibits the toxic action of other agents. Selenium is important for Hashimoto’s disease. Intake of selenium in the diet slows the aging process. The biological and toxicological effects of selenium strongly depend on its chemical form. Some organisms for example: plant, yeast, are capable of metabolizing low bioavailable selenium compounds (inorganic selenium) into its high bioavailable forms (organic selenium). Objective. The aim of this study was to investigate the bio-transformation of selenium by Lactobacillus bacteria towards the characterisation of selenium metabolites. Material and Methods. The speciation of selenium was evaluated by high performance liquid chromatography with inductively coupled plasma mass spectrometry detector. The extraction of selenium species from lyophilized bacteria was executed with water, the mixture of lipase and protease, as well as lisozyme and sodium dodecyl sulphate. Results. All investigated bacteria strains cultivated in the presence of Na2SeO3 effectively uptake selenium. Surprisingly, none of the applied extraction media exhibited a strong power to release the majority of the uptaken selenium compounds. Thus a maximum of 10% of the selenium was extracted from bacteria exposed to the enzymes. However, it was found that Lactobacillus bacteria are able to metabolize inorganic ions of selenium (IV) into Se-methionine, Se-methyloselenocysteine and other unidentified forms. Conclusions. The study confirmed the ability of probiotic bacteria to biotransform inorganic selenium into its organic derivatives. Therefore, Se-enriched bacteria can be considered as an addition to the functional food.
The study aimed at application of a high performance liquid chromatography (HPLC) with UV detector for selenium speciation in samples of mineral and well waters in Poland. Basic solutions 1000 μg/l SeO3 2- and SeO4 2- were prepared by dissolving 2.190 g of reagent Na2SeO3 and 2.393 g Na2SeO4 in 1000 ml of deionised water, respectively. Recent standards SeO3 2- and SeO4 2- (single or mixed) were prepared daily. Chromatographic separations were obtained on a column with reversed phase of C18, 250*4 mm, 5 μm ODS. As mobile phase mixture of 5 mmol/l phosphate tetrabutylammonium, 50/50 water/methanol at pH 3.4 was used. Flow rate of mobile phase was1 ml/min and detector was set on λ=210 nm. Injection volumes were 5 μl. The average concentration of Se(IV) was 4.10 μg/l and Se(VI) - 4.27 μg/l and Se(II) - 0.54 μg/l although the differences between water samples originating from various sources were relatively high. The developed method can be therefore recommended for determination of Se in samples of mineral water.
For its specific physical and physicochemical properties, the water treated with low-frequency low-pressure glow plasma (GPTW) affects the growth of plants and enhances the phytoavalibility of selenium (Se) ions from the nutrient solution. The basic biometric and physiological parameters of cucumber and the uptake of Se ions applied as selenate (Na2SeO4) from the nutrient solution prepared using GPTW or distilled water (DW) were compared. In the presence of Se, the fresh weight (f.w.) of shoots of plants growing in waterdifferentiated nutrient solutions did not differ, whilst their dry weight (d.w.) and leaf area (LA) were higher in plants grown in the GPTW- than in DW-containing medium. The use of GPTW for preparation of the nutrient solution was associated with a substantial improvement of Se ions phytoavailability, compared to the regular growth medium based on DW. Despite the higher Se bioaccumulation in the GPTW- than in DW-based medium, the phytotoxicity of this element was not enhanced. GPTW-induced Se accumulation was remarkable and hence recommended for further study to understand the detailed mechanism GPTW action.
Background. Persistently high incidence of ‘diseases of affluence’ (including cancer) motivates numerous research teams to look for causes of morbidity, as well as to search for preventive methods and effective therapeutic measures. The paper aims to present the literature on effects of selenium (Se) on prevention of gastrointestinal and lung cancer. Material and methods. Based on national and international literature, the paper presents information on the role of selenium (Se) in prevention of cancer – with special consideration given to gastrointestinal and lung cancer. Results. The results of national and international research show the importance of selenium in prevention and treatment of cancerous diseases, including digestive tract cancer and lung cancer. Numerous studies have shown that the risk of cancer for people with low selenium levels is twice as high as for people with high levels of selenium in blood serum. The most prominent relationship between low selenium level in serum and cancer is observed in gastrointestinal cancer, lung cancer, breast cancer in women, and prostate cancer. Conclusions. Many clinical and experimental studies carried out nationally and internationally gathered evidence which indicate the vital role played by selenium in prevention and treatment of diseases – especially of cancer. It should be emphasised that the intake of selenium in amounts necessary for proper functioning - given numerous deficiencies (first in the soil, then in food products coming from selenium-deficient soil) – is merely the first step, as it has already been proven in numerous publications that carbohydrates (especially simple sugars, sweets, cakes, etc.) ‘destroy’ selenium in the human body.
The aim of this study was: (a) to estimate the concentration of selenium in plasma of 136 residents (59 men and 77 women) of the Gdansk region, northern Poland, aged 19-70; and (b) to compare the obtained results with data corresponding to healtly populations living in different European countries. Selenium in plasma was determined by atomic absorption spectrometry using the hydride generation method. The mean selenium concentration in plasma of the investigated persons was: 72.3 +/- 14.1 mug/l, in men 75.9 +/- 12.8 mug/l, in women 69.5 +/- 14.7 mug/l. No age-dependent differences in plasma selenium were found in the investigated population. In 22% of the investigated persons the selenium level in plasma was lower than 60 mug/l. The obtained results indicate that a significant part of this population is deficient in selenium and should be supplemented with this element.
Experiments were performed on 12 sheep divided into 2 groups and fed a standard diet. The trial group received the same nutritive fodder supplemented with 0.3 ppm of sodium selenite (Na₂SeO3). The concentrations of selenium, triiodothyronine and thyroxine in serum were determined every 4 weeks during 20 weeks, respectively. Oral supplements of sodium selenite increased the serum concentration of selenium in sheep from 0.34 ± 0.19 to 4.25 ± 0.69 μmol/l. Contrary to those in the control group, serum Se content ranged from 0.34 ± 0.08 to 0.36 ± 0.10 μmol/I. Alterations in the metabolism of thyroid hormones showed that supplements of selenium produced an insignificant decrease of thyroxine with a slight increase in triiodothyronine concentration.
Owing to its biological properties the microelement selenium has attracted enormous interest. It has been established that selenium stimulates the human immune system and has anti-carcinogenic effect. The main sources of selenium are high-protein foodstuffs of plant and animal origin, as well as high-protein dairy products. The aim of this study was to detect selenium content in confectionery products using speciation analysis in order to determine inorganic forms of selenium such as Se2- SeO32- and SeO42- anions. The hydride generation method combined with the atomic absorption spectroscopy was used for the final determination of selenium forms. The determination of selenium was conducted using aqueous extraction and digestion of samples with concentrated acids. The speciation determination of selenium was conducted in ten confectionery products. The correlation between the total content of selenium and its individual forms (-II), (IV),(VI) of different oxidation degree was also examined. It was shown that there was no correlation between the total selenium and inorganic forms of selenium. That means that speciation analysis is the only correct analysis of selenium content in foods.
This paper presents statistical analysis of results of determination of the total content of arsenic, antimony, selenium and microcomponents in 49 samples of groundwater from an area of western Poland (Wielkopolska). The total content of arsenic reached up to 3.00 ng/ml, antimony 1.25 ng/ml, selenium < 0.15 ng/ml, at detection limits of 0.16 ng/ml for As, 0.18 ng/ml for Sb and 0.15 ng/ml for Se. The samples showed low values of the content of the elements considered relative to the values obtained for water samples from other regions. As the groundwater samples were collected from isolated water bearing horizons, the values determined for them could be treated as geogenic.
