The content of polychlorinated biphenyls was determined in human milk collected from four regions of Poland, as well as in cow’s milk and infant formulas. Significant statistical differences in PCB levels were determined in human milk depending on the region. The highest PCB mean concentration (0.351 mg. kg⁻¹ milk fat) was determined in human milk from Gdańsk. An average of 0.218 mg PCB kg⁻¹ milk fat was determined in human milk, 6 and 17 times higher than cow’s milk and infant formulas, respectively. Higher (37%) content of PCB was found in the milk of women living in rural regions.
Preparations of anti-DNA sIgA were obtained from human milk by sequential chro­matography on protein A-sepharose, DEAE-fractogel and DNA-cellulose. The influ­ence of oligonucleotides on protein kinase activity was investigated. It was discovered that incubation of anti-DNA sIgA with oligodeoxyriboadenylate d(A)12 stimulates the phosphorylation of polypeptides of sIgA in the presence of [γ- 32P]ATP. The greatest was the incorporation of 32P into the sIgA H-chains. We also demonstrated stimula­tion of the casein kinase activity of anti-DNA sIgA by d(A)12. The stimulation of the protein kinase activity of anti-DNA sIgA by oligoriboadenylate r(A)12 was not de­tected.
Background. In case of shortage of breast milk despite proper lactation care or the poor state of the mother’s health, breast milk from human milk bank is recommended for feeding preterm infants. Objective. This study retrospectively evaluated the first year of the operation of the Regional Human Milk Bank. Material and methods. Data concerning donors was collected in the human milk bank during the cooperation. The clinical characteristics of the recipients was made on the basis of medical documentation from the Holy Family Hospital in Warsaw, Poland. Analysis of nutritional value was performed with the human milk analyzer (MIRIS AB). Results. In the first year of activity, 45 voluntary donors established cooperation, donating from 650 to 32030 ml of human milk. The content of nutrients in milk provided by donors was variable - protein 0.4-1.5 g / 100 ml, fat 1.1-7.4 g / 100 ml, carbohydrates 6.3-7.9 g / 100 ml. The average length of using donated human milk was 4 days and the average volume of milk for one infant was 282 ml. Conclusions. The donor profiles have a significant impact on the milk composition form HMB. The nutritional value can be improved by recruitment donors from mothers that gave birth prematurely and by beginning donation at earlier stages of lactation as soon as lactation is stabilized. In case of shortage of mothers own milk the immediate implementation of donors milk as a short-term support can significantly reduce the food intolerance incidence in the group of prematurely born infants
The objective of this study was to examine the relation between the content of pentadecanoic acid C15:0 (PA) and heptadecanoic acid C17:0 (HA) in human milk and the type of the mothers' diet and determine the content of these fatty acids in infant formulas and follow-up formulas available on the market. Thirty-four, healthy mothers, aged 24-33 y were take part in the study. Mothers were classified according to their diet status into one of two groups: diet low in dairy products (15) and standard diet (19). Analyzed 12 differences infant formulas, 6 infant formulas and 6 follow-on formulas. In the group of women whose diet was Iow in dairy products, the PA content of human milk fat reached on average 0.195 ±0.017% of total fatty acids, and the HA content - 0.202 ±0.012%. In the group of women eating a standard diet, a statistically significant increase was observed in the PA and HA content of milk fat which on average reached: PA - 0.406 ±0.019% and HA - 0.360 ±0.015% of total fatty acids. It was concluded that in respect of human milk, PA and HA could be regarded as biomarkers of milk fat consumption by lactating women. From among 12 investigated infant formulas (IF) and follow-up formulas (FF) which are available in retail, PA of 0.63 to 0.98% and HA of 0.37 to 0.60% of total fatty acids was found in only 4 formulas which contain bovine milk fat. In IFs which, according to the manufacturer's statement, do not contain milk fat, PA content was from 0.04 to 0.07% and HA - from 0.02 to 0.08% of total fatty acids.
Introduction. The Total Antioxidant Capacity (TAC) of human milk reflects the concentration and the activity of many components which prevent oxidative degradation of fats and proteins. This study compares the effectiveness of ABTS and DPPH tests with regard to the recovery, precision and sensitivity (detection and quantification limit) of (TAC) values in human milk. Material and methods. TAC values were determined in twenty five samples of human milk obtained from healthy mothers, residents of Gdańsk, on the 14,h day postpartum. Results. The average TAC of human milk determined by ABTS assay was 19.61 ±3.311 mg TE (Trolox Equivalents)/100 cm3, the average values obtained by the DPPH assay reached 9.95 ±4.36 mg TE/100 cm3. For each milk sample the TAC determined by the ABTS test was significantly higher than the values produced by the DPPH test. The above findings can be attributed to the presence of substances whose spectra overlap with DPPH’ spectra. ABTS test was characterised by a higher sensitivity and repeatability of the determination of TAC in human milk compared to the DPPH test. Conclusions. Comparing the calculated values for the validation parameters of both methods and taking into account the solubility of DPPH only in polar matrices, slower reaction of selected antioxidants with DPPH radical, and the presence in human milk constituents absorbing electromagnetic radiation in the absorption of DPPH be assumed that the ABTS test is more appropriate method of determining of TAC in breast milk.
The concentrations of lead, cadmium, zinc, cuprum and chrominum in human milk were determined using GF AAS. The samples originated from women living in the Upper Silesian Industrial District (Poland). The mean concentration of lead was about 50 ng/g, cadmium 16 ng/g, zinc 50 μg/g, chromium 45.5 ng/g, and copper 1.1 μg/g. The mineralization of the samples was carried out both by means of concentrated mineral acids (HN03; HN03+HCI04 and HN03 + H202) under normal pressure for 12 hours, and mineralization with the microwave digestion system (which is most recommended).
Wprowadzenie. Karmienie piersią uznawane jest za najkorzystniejszy, naturalny sposób żywienia niemowląt. Skład ilościowy i jakościowy mleka kobiecego jest idealny do realizacji wszystkich potrzeb żywieniowych niemowlęcia do 6 miesiąca życia, przy założeniu, że sposób żywienia matki karmiącej jest prawidłowy. Cel badań. Celem pracy było oszacowanie spożycia energii oraz białka, tłuszczów, węglowodanów i cholesterolu w grupie 100 kobiet karmiących piersią, w wieku od 19 do 38 lat, z województwa mazowieckiego. Materiał i metoda. Badanie przeprowadzono w okresie od września 2010 do marca 2011. Wyniki uzyskano przy użyciu kwestionariusza ankietowego oraz metody 3-dniowego bieżącego notowania. Wyniki. Wartość energetyczna racji pokarmowych była zgodna z normą u 9% badanych kobiet. Aż 91% respondentek charakteryzowało się zbyt niską wartością energetyczną diety. Spożycie białka zgodne z normą odnotowano u 64% badanych kobiet, tłuszczu u 45%. Zalecenia spożycia kwasów tłuszczowych wielonienasyconych realizowało 61% badanych kobiet, nasyconych 15%, węglowodanów 36%. Spożycie cholesterolu było za wysokie u 45% badanych kobiet. Odnotowano istotny statystycznie wpływ wykształcenia na wielkość spożycia tłuszczu ogółem oraz kwasów tłuszczowych nasyconych i wielonienasyconych, sacharozy oraz wartość energetyczną całodziennych racji pokarmowych. Kobiety z wykształceniem wyższym spożywały od 12% do 20% więcej wymienionych składników stosunku do kobiet z wykształceniem średnim/zawodowym. Wnioski. Istnieje potrzeba dalszej edukacji kobiet w okresie laktacji na temat prawidłowo zbilansowanej diety, gdyż w ocenianych racjach pokarmowych kobiet karmiących wykazano wiele nieprawidłowości.
Objective: To determine transforming growth factor (TGF) β1, interleukin (IL) 4, and IL-10 concentrations in human milk and to assess the relationship between allergic disorders in mothers and the content of the interleukins in their milk. Material and methods: Thirty allergic and 46 healthy mothers were included in the study. Colostrum was collected 2-3 days after delivery. Cytokine concentrations were determined with commercial enzyme-linked immunosorbent systems. Results: TGF-β1was found in milk from 23 women in the control group (53.49%) and 11 in the allergy group (37.93%). When TGF-β1 was present, the median concentration was higher in the allergy group than in the control (61.5 and 30.4 pg/mL, respectively; P < 0.004). IL-10 was present in the colostrum of all the women and the median IL-10 concentration did not differ between the allergy (50.5 pg/mL) and control (51.5 pg/mL) groups. The probability of occurrence of a positive IL-4 value in the allergy group was greater than in the control group (chi-squared [df = 1] = 2.60, P < 0.053). Median IL-4 level did not differ significantly between the two groups (0.5 and 0.5 pg/mL respectively). Conclusions: TGF-β1 was detected less often in the colostrum of allergic mothers than in that of mothers without allergy (but the difference was not statistically significant). IL-4 was found more often in the colostrum of allergic mothers than nonallergic ones. The allergy status did not correlate with IL-10 concentration.
Oznaczono pozostałości chlorowanych węglowodorów: y-HCH, DDT i jego metabolitów DDE i DDD, w mleku kobiecym pozyskanym w roku 1996 w Olsztynie. Uzyskane wyniki badań porównano z wcześniejszymi badaniami przeprowadzonymi w latach 1986 i 1976 w tym samym regionie.
Badano skład kwasów tłuszczowych cis oraz zawartość izomerów trans w mleku 31 matek karmiących w trzech okresach laktacji i w zależności od sposobu żywienia. Nie stwierdzono istotnych różnic w udziale kwasów nasyconych, jedno- i wielonienasyconych w poszczególnych okresach laktacji i w zależności od składu diety szpitalnej i domowej. We wszystkich badanych mlekach obecne były izomery trans kwasów C16:1, C18:1, C18:2, C18:3, z przeważającym udziałem izomerów trans kwasu oleinowego. Średni udział izomerów trans w badanych mlekach obniżył się o ok. 14% po zmianie sposobu żywienia ze szpitalnego na domowy.
Human milk is rich in polyunsaturated fatty acids, as well as lysozyme, vitamin C and other bioactive compounds. The effect of lyophilization on the content of antioxidants (vitamin C and catalase CAT), bactericidal compounds (lysozyme), total antioxidant capacity (TAC) and lipid peroxidation in human milk was investigated in this study. Samples of mature human milk were collected from five healthy women who gave birth on the scheduled date and without complications. Freeze drying resulted in the removal of 88.2 % of the initial water content from milk. The human milk lyophilizate was readily soluble in water. Lyophilization had no effect on the content of primary (lipid peroxides LP) and secondary (thiobarbituric acid reactive substances TBARS) products of lipid oxidation. Freeze-drying led to a significant decrease in the vitamin C content and TAC values of milk (by 31 % and 16.5 %, respectively). Catalase and lysozyme were resistant to freeze-drying. Lyophilization induced a decrease in lysozyme content (9 %) and catalase activity (11 %) but these changes were not statistically significant. Low-temperature dehydration and rehydration of human milk lyophilizates promote satisfactory retention of biologically active ingredients and prevent the oxidation of human milk lipids. The results of this study indicate that lyophilization can be considered as an effective method for prolonging the shelf life of human milk.
The dietary components used by 102 breast-feeding women included in the study were reflected in the content of fatty acids in their breast-milk. Fatty acid profile in the milk was changing during lactation. Higher correlation ratios were found between the proportion of fatty acids in the analysed samples of human colostrum and the supply of fatty acids in the diet of women during pregnancy, when compared to mature milk. An inhibitory effect of trans fatty acids upon the synthesis of long-chain unsaturated fatty acids from their precursors, the linoleic acid and alpha-linoleic acid was observed.
