The influence of fluoride on tooth germ development, especially mineralised tissue, is well documented in numerous dental publications, but there are few reports concerning the influence of fluoride on enamel organ and dental papilla cells. The aim of the study was to assess histologically the development of tooth germs of 20-day-old rat foetuses whose mothers drank water without fluoride or with low (10 mg) and high (110 mg) contents of natrium fluoride, starting from the 12th day of the pregnancy. The fluoride contained in drinking water in low as well as high concentration accelerated the development of enamel organ and dental papilla structures in rat foetuses. The acceleration was proportional to the content of fluoride in drinking water. No disturbances caused by high concentration of natrium fluoride were observed.
The aim of this study was (a) to determine the concentration of fluoride and cadmium in urine of 1240 children ( 635 boys, 605 girls) from Gdańsk, aged 7-14; and (b) to examine whether a correlation exists between age and sex of children, the location of the schools, and the urinary levels of fluoride and cadmium. Fluoride was determined potentiometrically using a fluoride-specific electrode. Cadmium was determined by atomic absorption spectrometry. The mean fluoride concentration in urine in children attending two schools located close to a phosphate fertilizer waste disposal site was 2.14 ± 1.16 mg F¯/L, in three others 1.05 ± 0.49 mg F¯/L. The mean cadmium concentration in urine was 0.17 ± 0.19 μg Cd/L. In children aged 7 the cadmium concentration was significantly lower than in older ones.
The study was conducted on 40 women in the early postmenopausal period, aged 52.3±3.1 years with primary osteoporosis unmanageable in treatment, divided into 2 groups based on a randomized list. Group I (n-20) was administered orally fluoride 0.25 mg kg-1 24 h-1 with modified transdermal hormone therapy/HRT, and group II (n-20) was administered orally fluoride and supplement hormonal therapy(HST) in 21 therapeutic cycle. The serum concentrations of osteocalcin (OC), procollagen(PICP), insulin-like growth factor I (IGF-1), prolactin basic (PRL) and prolactin after metoclopramide (PRL/ MCP) 4 times by using radioimmunoassy methods, before treatment and after 1, 3, 12 months of treatment. Bone mineral density (BMD) L2 – L4 was determined before treatment and at 12 month with a dualenergy x-ray absorptiometry scanner (Lunar DPX-1Q). In group I women receiving fluoride and transdermal HRT IGF-1 increased significantly while the concentrations of OC and PICP significantly decreased after 3 and 12 months of treatment but no statistically significant changes in the PRL concentration occurred. In group II women receiving orally fluoride and HST, a significant decrease in the concentration of IGF-1, OC after 3 and 12 months and a significant increase in the concentration of PRL and PRL/ MCP after 1, 3 and 12 months of treatment compared with the baseline values appeared. The concentration of type I procolagen (PICP) showed no statistically significant changes. Increase in bone mineral density was statistically significant L1, L2 (p < 0.05), L3, L4 (p < 0.01) compared with the baseline in the group receiving transdermal HRT. In women receiving fluoride and orally HST increase in the bone mineral density for L1 and L2 was non-insignificant, whereas for L3 and L4 it was significantly higher compared with the baseline (p < 0.05).
An examination was made of fluoride content in the mandibular first molars of the permanent teeth of the red fox Vulpes vulpes living in north-west (NW) Poland. The teeth were first dried to a constant weight at 105°C and then ashed. Fluorides were determined potentiometrically, and their concentrations were expressed in dry weight (DW) and ash. The results were used to perform an indirect estimation of fluoride pollution in the examined region of Poland. The collected specimens (n = 35) were classified into one of the three age categories: immature (im, 6–12 months), subadult (subad, from 12 to 20 months) and adult (ad, >20 months). The mean concentrations (geometric mean) of fluoride were similar in the im and subad groups (230 and 296 mg/kg DW and 297 and 385 mg/kg ash, respectively), and significantly smaller than in the ad group (504 and 654 mg/kg, respectively, in DW and ash). Basing on other reports that the ∼400 mg/kg DW concentration of fluoride in bones in the long-lived wild mammals generally reflects the geochemical background, it was found that 57% of the foxes in NW Poland exceeded this value by 9% to 170%. This indirectly reflects a moderate fluoride contamination in the tested region.
Fluoride alters the expression and post-translational modifications of extracellular matrix proteins in dentin. The aim of our study was to determine the effects of fluoride on type I collagen expression during the early stages of tooth germ development in rats. Pregnant dams were divided into three groups and fed a standard diet. From the fifth day of pregnancy the three groups received tap water with, respectively, trace amounts of fluoride (C), a low fluoride concentration (FL) or and a high fluoride concentration (FH). Changes in type I collagen expression and distribution were evaluated. The expression of type I collagen was restricted to the extracellular spaces of cells of mesenchymal origin. In the youngest animals the most intense immunoreactivity for type I collagen was detected in predentin of the FL group. Although the intensity of immunostaining increased in proportion to the age of the animals, the largest increase in the groups investigated was detected in the FL group. We concluded that a low concentration of fluoride can act as a stimulator of type I collagen deposition in the extracellular matrix of dentin, while high concentrations of fluoride have an opposite effect, acting as an inhibitor of type I collagen formation in dentin.
The influence of diet and fluoride on odontogenesis in rats was investigated. 20 foetuses, 20 days old, were divided into four groups. The control group was fed with the standard diet and drank water with 0.16 mg F/l. The second, third and fourth groups were fed with the deficit, experimental diet and drank distilled water with 10 and 110 mg of natrium fluoride per litre or without fluoride. In each group, the observed tooth-bud development assumed different stages. The less advanced cap stage assumed the tooth-bud in the group fed with the deficit diet and given distilled water without fluoride. In the remaining groups, the development of observed first molars in mandible assumed the different level of its advancement in the same stage of odontogenesis — bell stage.
The effect of soluble fluorine species in different concentrations on the catabolic activity of Desulfotomaculum ruminis bacteria has been studied. The fluoride concentrations tolerated by and toxic to the bacteria have been determined. Celem pracy było ustalenie fizykochemicznych warunków wiązania fluorków z fosfogipsu przez nienasycone substancje organiczne zawarte w torfie z utworzeniem biologicznie biernych wiązań C-F. Badania prowadzono metodą statyczną w szerszym zakresie pH środowiska od 2 do 11. Największy ubytek F- wynoszący 93-94% zaobserwowano dla roztworów równowagowych o pH zawartym w zakresie 4.23-4.45. Przebieg widm IR wykonanych dla osadów powstałych po odparowaniu tych roztworów świadczy o powstawaniu wiązań C-F.
Tereny bytowania łosia europejskiego znajdują się w części Polski północno-wschodniej i wschodniej. Jest to obszar mało uprzemysłowiony o stosunkowo czystym środowisku naturalnym bez skażeń związkami fluoru. W badaniach naszych interesowała nas jaka jest zawartość fluoru w zębach tych zwierząt oraz kształtowanie się zależności pomiędzy wiekiem a zawartością fluoru w zębach. Stwierdziliśmy wzrost zawartości fluoru w zębach wraz z wiekiem.
The study characterizes specific therapeutic waters in Polish health resorts, which contain iodide, fluoride, silicon, sulphides, iron(II), carbon dioxide and radon in concentrations that ensure the therapeutic status of water and specify how it can be used for medical treatment. Based on the physicochemical analysis, it has been verified that among 160 waters from 39 health resorts, 37 contain Fe(II) in concentrations > 10 mg dm–3, which means that they can be classified as ferruginous. Twelve waters containing fluoride in concentrations > 2 mg dm–3 were recognized as therapeutic fluoride waters. Five waters were classified as siliceous ones owing to the content of silicon compounds above 70 mg dm–3 (H2SiO3). Twenty-eight of the analysed waters contain sulphides (H2S+HS–) in concentrations > 1 mg dm–3 and 64 waters contain iodide in concentrations > 1 mg dm–3. Carbon dioxide, which occurs in 72 waters in concentrations above 1 g dm–3 (acidulous waters), proved to be the most widespread component. Waters which contain radon in an amount corresponding to the radiation of 74 Bq (2 nCi) in 1 dm3 are classified as radon waters. Ten of the analysed waters were found to contain this element. The waters were highly varied in chemical composition and type of specific components. Sulphurous waters are mainly used for mineral baths; ferruginous waters are drunk; fluoride and silicon-rich waters are taken for mineral baths and iodide waters are used for mineral baths and inhalations. Using these waters in balneotherapy is an important part of spa treatment and defines different healing profiles of Polish health resorts.
Oznaczono stężenia fluorków w łożyskach gromadzonych w ciągu kilku lat, pochodzących od kobiet zamieszkujących tereny bezpośrednio sąsiadujące z zakładami chemicznymi emitującymi związki fluoru. Stwierdzono tendencję do obniżania się stężenia fluorków w badanym materiale wraz z poprawą sytuacji ekologicznej wokół źródła emisji. Zjawiska tego nie potwierdzono statystycznie, co wyklucza przydatność łożysk ludzkich jako bioindykatora w ocenie stopnia skażenia środowiska związkami fluoru.
Arabidopsis thaliana AtNUDT7 Nudix pyrophosphatase hydrolyzes NADH and ADP-ribose in vitro and is an important factor in the cellular response to diverse biotic and abiotic stresses. Several studies have shown that loss-of-function Atnudt7 mutant plants display many profound phenotypes. However the molecular mechanism of AtNUDT7 function remains elusive. To gain a better understanding of this hydrolase cellular role, proteins interacting with AtNUDT7 were identified. Using AtNUDT7 as a bait in an in vitro binding assay of proteins derived from cultured Arabidopsis cell extracts we identified the regulatory protein RACK1A as an AtNUDT7-interactor. RACK1A-AtNUDT7 interaction was confirmed in a yeast two-hybrid assay and in a pull-down assay and in Bimolecular Fluorescence Complementation (BiFC) analysis of the proteins transiently expressed in Arabidopsis protoplasts. However, no influence of RACK1A on AtNUDT7 hydrolase catalytic activity was observed. In vitro interaction between RACK1A and the AGG1 and AGG2 gamma subunits of the signal transducing heterotrimeric G protein was also detected and confirmed in BiFC assays. Moreover, association between AtNUDT7 and both AGG1 and AGG2 subunits was observed in Arabidopsis protoplasts, although binding of these proteins could not be detected in vitro. Based on the observed interactions we conclude that the AtNUDT7 Nudix hydrolase forms complexes in vitro and in vivo with regulatory proteins involved in signal transduction. Moreover, we provide the initial evidence that both signal transducing gamma subunits bind the regulatory RACK1A protein.
The purpose of our study was to assess whether the chemical composition of drinking water in the city of Szczecin, Poland, creates the correct environment in the oral cavity for promoting regeneration processes in the mineralized tissue of teeth. The drinking water samples were collected from a water treatment station, as raw and treatment water, and from consumer’s water pipes. The concentration of calcium and magnesium was determined by F-AAS method, fluoride by ion-specific electrode, and pH by pH-Meter. The average concentrations of calcium (46.9 mg/dm³), fluoride (0.73 mg/dm³), and pH level (7.46) determined in tap drinking water create a sufficient environment in the oral cavity for promoting regeneration processes in tooth mineralized tissues. Because of the low concentrations of magnesium in drinking water (14.1 mg/dm³), this element should be supplemented in a person’s diet.
Celem pracy było wykorzystanie kwasowej biopsji szkliwa do oceny zawartości fluorków i magnezu w powierzchniowych warstwach szkliwa zębów pochodzących z wykopalisk archeologicznych, przy uwzględnieniu ich wieku chronologicznego, wieku osobniczego oraz głębokości trawienia tkanki. Materiał badawczy stanowiły 44 zęby przedtrzonowe i trzonowe pobrane z czaszek pochodzących z dwóch serii szkieletowych z Wrocławia datowane na X-XII w. oraz na XV-XVI w. Grupę kontrolną stanowiło 16 zębów usuniętych ze wskazań periodontologicznych i protetycznych od osób dorosłych w wieku 32-68 lat zamieszkałych na terenie Szczecina. W grupach badanych wyselekcjonowano podgrupy różniące się wiekiem osobniczym: adultus (25-45 lat) i maturus (45-65 lat). Stosując metodę kwasowej biopsji szkliwa uzyskano mikropróbki tkanki trzech powierzchniowych warstw szkliwa z powierzchni policzkowej zębów. Pomiary stężenia fluorków przeprowadzono metodą chromatografii gazowej, natomiast stężenie magnezu mierzono przy użyciu spektrometru absorpcji atomowej. Stężenia fluorków we wszystkich warstwach szkliwa wzrastały wraz z obniżaniem wieku chronologicznego zębów. Podobną tendencję zmian zaobserwowano w przypadku magnezu, ale tylko w dwóch najbardziej powierzchniowych warstwach szkliwa. We wszystkich grupach, zarówno w przypadku fluorków jak i magnezu, stwierdzano spadek stężenia badanych pierwiastków wraz z głębokością trawienia szkliwa. Nie wykazano różnic statystycznie istotnych w stężeniach fluorków i magnezu pomiędzy podgrupami wyselekcjonowanymi na podstawie wieku osobniczego.
Badania prowadzono w celu ustalenia optymalnych warunków roztwarzalności fosfogipsu pochodzącego z Gdańskich Zakładów Nawozów Fosforowych w wodnych roztworach: KOH, Ca(OH)2, HNO3, kwasu cytrynowego, K2CO3, KC1 i NH4Cl. Stwierdzono, że pod wpływem wodnych roztworów KOH, Ca(OH)2, K2CO3 następuje neutralizacja fosfogipsu z jednoczesnym unieruchomieniem zawartych w nim fosforanów. W przypadku KOH i K2CO3 następuje roztwarzanie głównego składnika fosfogipsu, tj. CaSO4. K2CO3 powoduje znaczny wzrost roztwarzalności fluorków zawartych w fosfogipsie.
