One of the bioindicators most often applied to assess the quality of soil is its enzymatic activity. Undesirable changes in the activity of enzymes can imply excessive presence of substances which are harmful to soil environment, such as heavy metals. Being a heavy metal, zinc is also an element essential for maintaining proper functions of live organisms. The purpose of this study has been to determine the significance of changes occurring in moderately heavy soil under the influence of zinc. The experiment was carried out in three replicates under laboratory conditions. Sandy loams of pH 5.5 and 7.0 were used for the trials. The soils were contaminated with zinc according to the following design: control (natural content), raised content (70 mg Zn2+ kg–1), weakly polluted (200 mg Zn2+ kg–1 ), moderately polluted (500 mg Zn2+ kg–1), heavily polluted (1,500 mg Zn2+ kg–1) and very heavily polluted soil (5,000 mg and 10,000 mg Zn2+ kg–1). The soil samples prepared as above were brought to the moisture content of 50% maximum water capacity and incubated at 25oC for 120 days. On day 30, 60 and 120, the activity of dehydrogenases, β-glucosidase, urease, acid phosphatase and arylsulphatase was determined. Based on these determinations, the following indices were calculated: ED50, the index for resistance (RS) and the index for resilience (RL). The tests have demonstrated that as the rate of soil contamination with zinc increased, the activity of all the analyzed enzymes was significantly depressed. The negative influence of zinc contamination on the activity of particular enzymes, irrespective of the soil pH, persisted throughout the whole experiment. In respect of their sensitivity to zinc, the enzymes can be ordered as follows: arylsulphatase > dehydrogenases > acid phosphatase > urease > β-glucosidase. Zinc contamination caused lasting changes in the soil environment, but the return to the state of equilibrium was the quickest in the case of dehydrogenases (RL = 0.276), less rapid for arylsulphatase (RL = 0.173) and the slowest for acid phosphatase (RL = 0.064). In contrast, the activity of urease, instead of regenerating, was increasingly disturbed (RL = 0.350). Soil acidification was the factor that most evidently exacerbated the negative influence of zinc on the activity of β-glucosidase and arylsulphatase. Values of ED50 for the activity of particular enzymes were varied. In the soil of pH 7.0, they ranged from 3,324 mg Zn2+ kg–1 for β-glucosidase to 412 mg Zn2+ kg–1 for dehydrogenases, and in the soil of pH 5.5, they varied from 1,008 Zn2+ kg–1 for β-glucosidase to 280 mg Zn2+ kg–1 for arylsulphatase.

Enzyme activity in forest peat soils

The aim of the study was to determine the activity of dehydrogenases and urease in forest peat soils of different fertility. There were selected 23 experimental plots localised in central and northern Poland. The research was conducted on forest fens, transition bogs and raised bogs. The biggest differences in soil physical and chemical properties were detected between fen and raised bog soils while raised bog soils and transition bog soils differed the least. Statistically significant differences between particular subtypes of peat soils were observed for soil pH-H2O, pH-KCl, C/N ratio as well as the content of organic carbon, nitrogen, calcium and potassium. The highest average dehydrogenase activity in the soil surface level was observed in fen soils, and the lowest – in raised bogs soils. The results obtained on urease activity were similar. Differences in urease activity in the studied soil types were shown. Dehydrogenase activity did not reveal statistically significant diversity. The activity of urease was negatively correlated with the content of carbon, C/N ratio, hydrolytic acidity and moisture. Also the increase in enzymatic activity accompanied by the increase in pH has been observed.
The activities of six enzymes associated with carbohydrate metabo­lism were measured both in carcinomas and in normal breast tissues. The following differences were observed. 1. The carcinoma showed higher enzyme activities than the normal nummary tissue. 2. The ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, gluta­thione reductase and catalase to lactate dehydrogenase were lower in carcinomas than in normal tissues. Similarly, the ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, glutathione reduc­tase and catalase to glucose-6-phosphate dehydrogenase were also sig­nificantly lower in carcinomas. 3. There were no significant differences in enzyme activities between I and II stage of the disease and the metastatic tissues, however, there were significant differences between I and III stage. The significance of these findings is discussed in terms of the alterations in the balance between the metabolic pathways.
Papain activity was studied in water-organic solvent mixtures using the fluorogenic substrate Dabcyl-Lys-Phe-Gly-Gly-Ala-Ala-Edans. The increase of organic solvent (MeOH, EtOH, iPrOH, TFE, MeCN, (MeO)2Et and DMF) concentration in the mixture caused a substantial decrease the initial rate of papain-catalyzed hydrolysis. More­over, the number of papain active sites decreased with the increase of DMF and MeOH concentration.
The effect of germination on the rate of formation of some endo- and exohydrolases, important in baking, in rye kernels was examined. Germination of the rye gave increase in a-amylase, endo-b-xylanase, endo-b-glucanase, b-xylosidase, a-arabinosidase, b-glucosidase as well as endo- and exopeptidases activities. The most intensive changes of activities in the group of examined enzymes were observed for a-amylase. b-amylase activity did not significantly increase on germination. The increase of endohydrolases activities in germinating rye kernels was much more markedly compared with activities of exohydrolases.
Potential activity of hydrolytic microbial extracellular enzymes were determined in a sandy beach in Sopot, (Southern Baltic Sea) in July 2001. Among of the five monitored enzymes, the highest level of activity (314.28 nM MCA g⁻¹d.w. h⁻¹) was determined in the aminopeptidase whereas chitinase had the lowest level (0.50 nM MUF g⁻¹d.w. h⁻¹) of potential activity. Usually, the ranking of the activity rates of the assayed enzymes was following: aminopeptidase > lipase > β-glucosidase > α-glucosidase > chitinase. Clear gradients in the level of enzymatic activity were determined in horizontal profile of the beach. The maximum activity of studied enzymes was recorded in sand collected under seawater and at the waterline.
Field study was performed on Haplic Luvisol soil to determine the effects of soil compaction and straw mulching on microbial parameters of soil under soybean. Treatments with different compaction were established on unmulched and mulched with straw soil. The effect of soil compaction and straw mulching on the total bacteria number and activities of dehydrogenases, protease, alkaline and acid phosphatases was studied. The results of study indicated the decrease of enzymes activities in strongly compacted soil and their increase in medium compacted soil as compared to no-compacted treatment. Mulch application caused stimulation of the bacteria total number and enzymatic activity in the soil under all compaction levels. Compaction and mulch effects were significant for all analyzed microbial parameters (P<0.001).
