Molecular markers have become a useful tool simplifying and speeding up breeding work. They are also helpful in the genetic analysis of complex agronomic traits. The investigations concern the use of already available SCAR markers for molecular analysis of breeding materials. The markers are the apple powdery mildew resistance gene Pl2, originating from Malus zumi, and the apple scab resistance gene Vf, from M. floribunda 821. The clone U 211 was found to be highly resistant to mildew under field conditions and transmitted a high level of resistance to the majority of its progeny. The presence of the Vf gene conferring resistance to scab was confirmed by molecular analysis. As the molecular markers for the Pl2 gene are not present in the DNA of U 211, it is probable that the clone U 211 is a new source of mildew resistance.
Plants respond to infection by accumulating many compounds some of which may function in disease resistance. These include: phytoalexins, antifungal proteins, chitinases, glucanases, esterases, proteaes, phospholipases, lipoxygenases, ribonucleases, peroxidases, phenoloxidases, lignin, callose, hydroxyproline and glycine-rich glycoproteins, phenolic cross-linked polysachcarides, melanin-like pigments, salicylic acid, jasmonic acid, ethylene, peptides, oligosaccharides, hydrogen peroxide and active oxygen species. Though specific avirulence genes, elicitors and elicitor receptors have been reported, the production of defense-related compounds is nonspecific and can be elicited by pathogens, pathogen products and many organics and inorganics. The molecular implications of this specificity/nonspecificity and their significance to disease resistance and practical disease control will be discussed.
These investigations aimed to evaluate the influence of acaricides used in Bulgaria (based on amitraz, cumaphos and fluvalinate) on some factors of the non-specific bee immunity in the appearance of chalkbrood. It was found that the changes of protein concentration and lysozyme amount in the haemolymph and the pH of bee intestine depend on different active substances of the drugs. Field trials canied out on 526 colonies revealed that the lower resistance of bees infected with Varroa jacobsoni was a cause of the appearance of chalkbrood. This resistance was additionally decreased by using the anti-Varroa mite product “Perizin”.
A systematic gathering of winter triticale accessions was started in Poland in 1982 by the Institute of Genetics, Breeding and Seed Science at the Agricultural University in Lublin (at present its name is: Institute of Genetics, Breeding and Plant Biotechnology at the University of Life Sciences in Lublin). First, breeding lines obtained in local breeding stations were gathered. Next, accessions were imported from the following world gene banks: Beltsville, Gatersleben, and VIR. Interesting hybrid materials obtained in research centers were also included in the collection. Now, the collection includes 2349 accessions (1329 of winter triticale and 1020 of spring triticale). The evaluation is conducted in a 4-year cycle of field experiments using the same methods. The gathered accessions represent a large range of variability of both morphological and commercial traits. The large differentiation of accessions especially concerns traits such as: plant height, number and weight of grains per spike, protein content in grain, field resistance to powdery mildew, brown rust and leaf and spike diseases.
Parental lines for breeding potato cultivars suitable for different purposes and resistant to potato cyst nematode are being developed in Plant Breeding and Acclimatization Institute, Research Division Młochów. An initial step to this programme was collecting of nineteen potato cultivars from Germany, Netherlands and Great Britain, highly resistant to different pathotypes of Globodera rostochiensis (Ro1-Ro5) and/or to G. pallida (Pa1-Pa3). Cooking quality of these cultivars was tested and compared to Polish potato cultivars. The following parameters of cooked tuber flesh were evaluated: taste, darkening and aroma. Cooking quality of particular cultivars, which expressed the complex resistance to potato cyst nematode is comparable to that of commonly grown Polish cultivars, susceptible or resistant to pathotype Ro1. It was concluded that there exist a possibility of breeding the cultivars joining good cooking quality with complex resistance to potato cyst nematodes.
The usefulness of fungus culture filtrates and fusaric acid as selecting agents for Fusarium resistance breeding in tulip was examined on in vitro cultures of shoots and embryonic calli of seven tulili genotypep differing in resistance to Fusarium oxysporum Schlecht, f. sp. tulipae Apt. (F.o.t.) and four virulent F.o.t. isolates. Fusaric acid influenced the shoot growth of all cultivars tested in a similar way, irrespectively of their greenhouse resistance to basal rot. Also, the sensitivity of calli of the cultivars studied to fusaric acid did not correspond with their resistance to F.o.t. evaluated in the greenhouse screening. The phytotoxity of F.o.t. culture filtrates did not depend on their fusaric acid contents. There was a negative correlation between cultivar's resistance to F.o.t in greenhouse tests and the sensitivity of their shoots to fungus culture filtrates in in vitro tests. This indicates that defence mechanism against F.o.t. in tulip tissue may have a nature of hypersensitive response. Considering the results of our study, it may be concluded that the use of fusaric acid or fungus culture filtrates for the in vitro selection of somaclones resistant to F.o.t. in tulip is not feasible.
