The aim of the study was to assess the seroprevalence of Coxiella burnetii in cattle herds in different regions of Poland. A total of 1150 serum samples collected from 443 cattle herds from 14 provinces were tested using complement fixation test. The seroprevalence was different in individual regions of Poland. The average percentage of seropositive herds was 40.41% and these herds were identified in each province tested.
This work presents results of the research on the occurrence of Coxiella burnetii and Francisella tularensis in the tissues of wild-living animals and ticks collected from Drawsko County, West Pomeranian Voivodeship. The real-time PCR testing for the pathogens comprised 928 samples of animal internal organs and 1551 ticks. The presence of C. burnetii was detected in 3% of wild-living animals and in 0.45–3.45% (dependent on collection areas) of ticks. The genetic sequences of F. tularensis were present in 0.49 % of ticks (only in one location – Drawa) and were not detected in animal tissues. The results indicate respectively low proportion of animals and ticks infected with C. burnetii and F. tularensis.
An epidemiological study was carried out to determine the herd prevalence of Chlamydophila abortus and Coxiella burnetii antibodies in goats covered by a milk recording program in Poland. The survey took place in 2007 and 48 herds located in different parts of the country were involved. A representative sample from each herd was taken by a simple random sampling allowing to detect seropositivity of a herd on a 95% level of confidence. In total 918 goats were tested for specific antibodies against both germs with the use of enzyme-linked immunosorbent assays. In addition, history of reproductive failures was recorded in these herds. The survey revealed that the herd prevalence of C. abortus was 4.2% (2 herds) while no C. burnetii antibodies were found. Abortions were reported to be a problem in 80% of herds while repeating estrus was encountered in 46% of herds. Reproductive failure concerned two seropositive herds as well. Since the germ is present in the population, it has to be taken into consideration in diagnostic process. Nevertheless, the results of the present study indicate that C. abortus infection occurs infrequently in Polish goats. As no antibodies against C. burnetii were detected in the screened sample the risk of goat-to-human transmission of both bacteria in Poland seems to be very low.
Q fever is a worldwide zoonosis that is manifested as a reproductive failure in animals and by polymorphic, nonspecific symptoms in humans. The infection may be acquired through the respiratory or alimentary route or an arthropod bite. The diagnosis of Q fever relies mainly upon serology: indirect immunofluorescence assays (IFA), complement fixation (CF) tests or enzyme-linked immunosorbent assays (ELISA) are used to detect antibodies against Coxiella burnetii in sera of infected animals. The aim of our study was to evaluate the agreement of the commercially available ELISA kit and CF test using statistical methods (Kappa value). We used serum samples that were collected from 122 dairy cows from one herd in central Poland. The general health status of the herd was good, and the animals were clinically normal. Our results showed a low agreement (Kappa value = 0.376) between the commercially available ELISA and CF test.
Apoptosis is a genetically controlled mechanism of cell death involved in the regulation of tissue homeostasis. The aim of this study was to investigate the influence of Borrelia afzelii, Coxiella burnetii, and Bartonella henselae bacteria on apoptosis measured as the level of caspase 3 activity in human fibroblast cells HEL-299. Our findings show that C. burnetii bacteria may inhibit the process of apoptosis in the host cells for a long time. This can permit intracellular survival in the host and mediatingthe development of chronic disease.
The importance of Q fever from the veterinary and medical point of view was characterized, and the fact that the World Organisation for Animal Health (OIE) had classified it as a listed disease was mentioned. The reason for this classification was the zoonotic potential of Coxiella (C.) burnetii and its ubiquitous, global occurrence. The main properties of this microorganism were described. The epizootic of the Q fever outbreak in late 2009 in the Netherlands and control methods applied in goat and sheep farms were shortly described. The main reservoirs of the infection and transmission of C. burnetii to human beings were mentioned. A summary report on trends and sources of zoonoses and zoonotic agents in the European Union member countries in 2008 was cited in relation to Q fever monitoring in animals and in humans. The role of wildlife and domestic animals, particularly ruminants, as the reservoir of Q fever and the role of ticks in transmitting the infection to human beings and domestic animals were evaluated. Cows, sheep and goats, in particular their feces, urine, amniotic fluid and aborted fetuses, were indicated as the most important sources of infection. The aerogenic transmission of C. burnetii by aerosols from these sources was evaluated as the most frequent one. Other sources of infection of animals and humans were also mentioned. The symptoms of Q fever in humans and animals were described. The importance of Q fever to public health was underlined, noting that C. burnetii is highly pathogenic for human beings. In connection with this statement the role of veterinary prophylaxis of this zoonosis was emphasized. Diagnostic laboratory tests were mentioned as well as the general and specific prophylaxis of Q fever and the use of antibiotics in domestic ruminants. The authors made efforts to present important recent achievements in the research on Q fever and its etiological agent.
