Chromosome morphology was studied in lily genotypes L. candidum, L. x formolongi, L. henryi and L. pumilum, and cultivars ‘Alma Ata,’ ‘Expression,’ ‘Marco Polo,’ ‘Muscadet’ and ‘Star Gazer’ belonging to the horticultural group Oriental hybrids. All genotypes tested represented 2n = 2x = 24 chromosomes. Chromosomal markers were established after Feulgen and silver staining, from analysis of the chromosome length and position of the primary and secondary constrictions. For each chromosome the arm index was calculated. Based on these data, idiograms were drawn. For the genotypes analyzed the markers were the secondary constrictions, as confirmed by silver staining. Chromosome length can be used as a marker in only a few cases. From 4 to 10 chromosomes could be identified using secondary constrictions as markers, depending on the genotype. Markers are proposed for each possible species x cultivar and cultivar x species combination.
The effects of TDZ and paclobutrazol on the primary regeneration on tulip flower stalk explants of six cultivars and subsequent shoot multiplication were examined. Explants, flower stalk slices, were excised from cooled and subsequently forced bulbs. The explants were incubated for two months in darkness on medium containing NAA and cytokinins, 2iP and BAP, as control, or TDZ (0.5-4 mg l-1) and paclobutrazol (0.05-0.4 mg l-1). Then, the regenerating explants were subcultured on medium with TDZ and NAA applied at low concentrations. Different regeneration capabilities were found depending on cultivar and growth regulators. The percentage of explants forming leaf-like structures ranged, on the control medium, from 80% in 'Blue Parrot' and 'Prominence' to below 30% in 'Apeldoorn' and 'Mirjoran'. TDZ, applied at optimum for each cultivar concentration, greatly increased the regeneration potential up to 70-100%. Paclobutrazol, added to the TDZ-containing medium, significantly enhanced the response of explants, resulting in high numbers of leaf-like structures formed per explant (13.7-22.8). The structures developed gradually into characteristic forms: the growing up cotyledonary leaf, the probable root primordium formed at its base, the growing downwards stolon and the shoot meristem developed finely on its tip. It is suggested that such primary regeneration may have a nature of somatic embryogenesis. Then, the adventitious shoots developed and formed clusters, which were divided into 2-3 smaller ones every two months. The growth regulators, used at initial stage, markedly influenced subsequent shoot multiplication. Thus, the most intensive shoot formation was noted with TDZ at concentrations of 0.5-2 mg l-1 and paclobutrazol of 0.05-0.1 mg l-1.
W pracy przedstawiono wyniki badań nad wpływem biopreparatu Biosept 33 SL w ograniczaniu rozwoju Puccinia malvacearum na malwie ogrodowej w warunkach uprawy polowej. Substancjami biologicznie aktywnymi tego środka ochrony są przede wszystkim endogenne flawonoidy grejpfruta. Biopreparat Biosept 33 SL zastosowany profilaktycznie (0,05% i 0,1%) w odstępach tygodniowych, wczesną wiosną, na wybijające liście, silnie ograniczał rozwój rdzy na malwie w porównaniu do roślin kontrolnych nieopryskiwanych. Po 30 dniach od ostatniego zabiegu ochrony, liczba zainfekowanych liści na roślinie i liczba skupień teliospor na zainfekowanych blaszkach liściowych i ogonkach liściowych była około pięciokrotnie mniejsza w porównaniu do roślin kontrolnych niechronionych. Stosowanie biopreparatu Biosept 33 SL w odstępach dwutygodniowych w mniejszym stopniu ograniczało rozwój rdzy na malwie w porównaniu do roślin opryskiwanych w odstępach tygodniowych. Efekt ochronny biopreparatu Biosept 33 SL był słabszy w porównaniu do środka porównawczego Amistar 250 SC 0,05% i 0,1% (substancja aktywna azoksystrobina).
Krzyżowanie międzygatunkowe i międzyrodzajowe jest ważną metodą we współczesnej hodowli. Celem tego krzyżowania jest najczęściej włączenie genów z form nieuprawnych do materiału hodowlanego, warunkujących lepszą zdolność przystosowawczą do niesprzyjających warunków uprawy i odporność na czynniki patogeniczne. W wyniku krzyżowania oddalonego mogą zaistnieć warunki do powstawania zarodków apomiktycznych oraz do eliminacji całego lub części genomu jednego, albo obojga partnerów. Dlatego jest ważne, aby jak najwcześniej zweryfikować status genetyczny powstających siewek lub zregenerowanych roślin. Weryfikację można prowadzić wykorzystując uprzednio opracowane markery. W tej pracy omówiono metody testowania mieszańców oddalonych na podstawie markerów morfologicznych, cytologicznych i molekularnych.
