The effect of the compound was studied in homogenates from Musca domestica L. Manometric and spectrophotometric methods were used to examine the rate with which cytochrome oxidase disappeared at various temperatures; the influence of gamma-HCH on the activity of cytochrome oxidase at various temperatures was examined; the effect of gamma-HCH on the activity of cytochrome oxidase was tested in homogenates from insects of varying age. Gamm-HCH at the concentration of 10⁻³, 10⁻⁴ and 10⁻⁵M was found to inhibit the activity of cytochrome oxidase regardless of the age of insects; the inhibition depends but slightly on the temperature. Saturated gamma-HCH solutions stimulate the activity of cytochrome oxidase in homogenates from M. domestica. Alfa-HCH isomer failed to affect the activity of the enzyme under analogous conditions.
The oxygen consumption studies in Musca domestica L., poisoned with γ-HCH were carried out at various temperatures. Manometric method at temperatures of 20, 25, 30, 35, 40 and 45 °C, and the doses of 0,01 μg γ-HCH/insect, 0,1 μg γ-HCH/insect, 1,0 μg γ-HCH/insect were used. At the temperature near to lethal, the oxygen consumption of control insects is not linear, but shows a characteristk maximum. Similarly, the oxygen consumption of insects poisoned by γ-HCH is not linear but exhibits a characteristic peak. A considerable analogy was found in the dynamics of oxygen consumption by insects exposed to higher temperatures, as compared with those poisoned with γ-HCH.
A description of an automatic micrometric apparatus constructed by the authors. The apparatus is served by an electric motor and an interval time disconnecting swith is used. Calibration of the apparatus showed a precision of dosage, determined by the use of the variability coefficient WZ=0,6-6,5%. Calibration was made by weight using hydrargyrum and colorimetric method to determine the dispersion of the size of the single drops. Crystal violet was used as the stain. Biological testing conducted on females of the house fly (Musca domestica L.) cultivated under laboratory conditions showed in the course of the determination LD50 - gamma HCH the accuracy WZ=9,7%.
